Book excerpt: Excerpt from The Journal of Biological Chemistry, 1918, Vol. 33 The finding of occult blood in stools is of great importance in the diagnosis of ulcer and cancer of the stomach. However, the usual chemical methods are so unreliable in their results that they are at present discredited by many surgeons and internists. A review of the literature of this subject shows not only that the number of reagents proposed is very large, but that there also exists a considerable number of procedures for the use of these reagents. The two most widely used are gum guaiac and benzidine. The phenolphthalein reagent is difficult to prepare and moreover is entirely too sensitive for ordinary use. In a previous paper we have shown how the benzidine reagent may give erroneous results unless great care is exercised in cleaning all the apparatus as well as in the use of reagents of definite concentrations. In the routine examination of stools we have found it difficult to get good controls with benzidine; moreover, it is too sensitive. An alcoholic solution of gum guaiac possesses the following disadvantages: (1) Not being of fixed composition or purity, a solution of definite concentration cannot be readily prepared. (2) Gums from different sources vary greatly in their sensitiveness as well as in the stability of the blue compound formed. (3) For the best results, solutions must be freshly prepared. In order to make use of the excretion of hippuric acid during the 6 hour period as a measure of the rate of synthesis of glycine, it must be shown that the excretion of hippuric acid depends upon the amount of glycine available and not upon the rate of absorption of benzoate from the intestine, nor upon the rate of formation of hippuric acid, nor upon the rate of elimination of hippuric acid by the kidneys. Book excerpt: Excerpt from The Journal of Biological Chemistry, 1907, Vol. 2 I have thought it worth while to bring together here a number of observations relating to bacterial processes in the intestinal tracts of persons suffering from severe forms of anaemia, in the belief that they afford a clue to the etiology of many cases of advanced blood disease that are ordinarily described as "primary." Most of the instances which form the basis of this study have given the blood-picture of so-called idiopathic, pernicious anaemia. The observations relate especially to seventeen cases of anaemia. Of these nine showed those changes in the morphology of the red cells (including the presence of megaloblastic forms) which are regarded as most characteristic of primary pernicious anaemia (Cases I, III, V, VII, VIII, X, XII, XIII, XVII). In four other cases (Cases II, IV, VI, and IX) the diagnosis of pernicious anaemia was made but appears less certain as megaloblasts were not seen. Of the remaining four cases one is an adult from whose history and blood-picture a distinguished consultant leaned to the diagnosis of pernicious anaemia, while another practitioner, upon the same evidence, suspected that he was dealing with an anaemia secondary to gastric carcinoma (XV). Another case is one of severe anaemia which was regarded as secondary, but for which no explanation could be found at autopsy (Case XI). Another case is that of a young child in the care of Dr. Holt, whose history is especially instructive as showing the rapidity of the process of blood regeneration upon the subsidence of a subacute intestinal process to which the anaemia was apparently secondary (Case XVI). Book excerpt: In the twenty years since their inception, modern dynamic light-scattering techniques have become increasingly sophisticated, and their applications have grown exceedingly diverse. Applications of the techniques to problems in physics, chemistry, biology, medicine, and fluid mechanics have prolifer ated. It is probably no longer possible for one or two authors to write a monograph to cover in depth the advances in scattering techniques and the main areas in which they have made a major impact. This volume, which we expect to be the first of aseries, presents reviews of selected specialized areas by renowned experts. It makes no attempt to be comprehensive; it emphasizes a body of related applications to polymeric, biological, and colloidal systems, and to critical phenomena. The well-known monographs on dynamic light scattering by Berne and Pecora and by Chu were published almost ten years ago. They provided comprehensive treatments of the general principles of dynamic light scat tering and gave introductions to a wide variety of applications, but natu rally they could not treat the new applications and advances in older ones that have arisen in the last decade. The new applications include studies of interacting particles in solution (Chapter 4); scaling approaches to the dynamics of polymers, including polymers in semidilute solution (Chapter 5); the use of both Fabry-Perot interferometry and photon correlation spectroscopy to study bulk polymers (Chapter 6); studies of micelIes and microemulsions (Chapter 8); studies of polymer gels (Chapter 9).
