This gene was previously determined to be down regulated in a rat model system of ovarian cancer and in a high percentage of human ovarian cancer cell lines. Unlike traditional tumor suppressor genes, which follow Knudson's two-hit hypothesis, the LOT1 open reading frame (ORF) was determined to be devoid of any inactivating mutations. Since the 5' regulatory region of a gene may also regulate a gene's expression, we sought to clone and functionally characterize the LOT1 cis-acting region. A region upstream of the LOT1 5' end was cloned and was demonstrated to have transcriptional activity in luciferase reporter assays using human ovarian cancer cell line A2780.

We have set up the infrastructure at Mayo for an Ovarian Cancer Research Program utilizing the rich resources of clinical material and linking work from molecular geneticists with that of dedicated clinicians. We are proposing to combine several powerful strategies to clone many of fie genes involved in ovarian cancer development. The first project focuses on identifying genes that are over-expressed or under-expressed during the development of ovarian cancer using subtraction suppression hybridization cDNA libraries and Gene Expression Array Analysis in collaboration with Millennium Predictive Medicine. The second project focuses on the role of gene amplification in familial versus sporadic ovarian cancer. The third project is to characterize two common fragile sites, FRA6F (6q21) and FRA6E (6q26) which are derived from chromosomal regions frequently deleted in ovarian cancer and that also contain genes involved in replication senescence. Thus we have three interactive projects whose overall focus is to identify key genetic targets in the development of ovarian cancer. This Program Project is centered within a larger institutional effort to better understand the biology of the development of ovarian cancer and to devise better strategies for prevention, early detection and treatment of this lethal disease.

Book excerpt: The objectives of this study were to (1) analyze gene expression in a model of human ovarian cancer from a benign to a malignant phenotype in cells grown in organotypic culture and (20 confirm gene expression patterns by RNA analysis. Two human cell lines were used, 96.9.18 and 1.24.96, and RNA extracted from cells at various stages of malignant change were queried on Affymetrix HU133A and B chips in triplicate (96.9.18) or HU133A plus chip in triplicate (1.24.96) . Data analysis has revealed a number of genes that are dysregulated in both 96.9.18 and 1.24.96. An analysis of the interaction of these genes showed that 5 genes are downregulated between the two cell lines and 183 genes are upregulated during malignant progression. Downregulated genes are too few to analyze but the upregulated genes are involved in apotosis, cytokine-cytokine receptor interactions, toll-like receptor signaling pathway, and the MAPK signaling pathway.
