Book excerpt: Modification of Ca2+ cycling function of sarcoplasmic reticulum (SR) is a major component of mechanisms underlying thyroid status-dependent alterations in contractility and energy metabolism of heart and skeletal muscle. We hypothesized that thyroid hormone status-dependent alterations occur in the expression and function of SR Ca2+-cycling proteins, and their regulation by Ca2+/calmodulin-dependent protein kinase II (CaM kinase II) and cAMP-dependent protein kinase (PKA), leading to characteristic changes in contraction-relaxation cycle of cardiac and skeletal muscle. We utilized hyperthyroid and hyperthyroid rabbit models to investigate this hypothesis. Cardiac contractile function parameters were assessed in isolated perfused heart preparations; contractile function of skeletal muscle (soleus) was studied in situ in anesthetized rabbits. SR function was studied in vitro using isolated junctional SR (JSR) and longitudinal SR (LSR) vesicles. We found the following. (i) Thyroid hormone induced overexpression of ryanodine receptors (RyR2) in the JSR of rabbit hearts, which is closely associated with enhanced Ca 2+-release activity of SR and enhanced cardiac systolic function in the hyperthyroid state. (ii) There was significantly increased expression of SR Ca2+-ATPase isoform II (SERCA2) in hyperthyroid cardiac JSR and decrease of phospholamban (PLN) in both hyperthyroid LSR and JSR compared to euthyroid. These changes are in consistent with elevated SR Ca2+ sequestration function and cardiac diastolic function in the hyperthyroid state. (iii) CaM kinase II-mediated phosphorylation of Ca2+-cycling proteins (RyR2, SERCA2 and PLN) was significantly lower (30--50%) in JSR and LSR of hyperthyroid heart; exogenous PKA-mediated phosphorylation per unit of immunoreactive PLN was significantly higher in the hyperthyroid heart. These findings demonstrate selective down regulation of the expression and function of CaM kinase II in the hyperthyroid rabbit heart in the face of upregulated expression of RyR2 and SERCA2. (iv) In rabbit soleus muscle, thyroid hormone-induced enhancement of contractile properties is associated with co-ordinate down regulation of the expression and function of phospholamban and CaM kinase II, and selective upregulation of the expression and function of the SERCA1 but not SERCA2 isoform of the SR Ca2+ PUMP. These findings imply unique, interactive roles for phospholamban and CaM kinase II in SERCA2 isoform-specific physiological regulation of SR Ca2+ pump function.

Book excerpt: Life Science studies in space were initially driven by the need to explore how man could survive spaceflight conditions; the effects of being launched un der high accelerations, exposed to weightlessness and radiation for different periods of time, and returned to Earth in safety. In order to substantiate the detailed knowledge of potentially adverse effects, many model experiments were launched using organisms which ranged from bacteria, plants, inverte brates, rodents and primates through to man. Although no immediate life threatening effects were found, these experiments can be considered today as the precursors to life science research in space. Many unexplained effects on these life forms were attributed to the condition of weightlessness. Most of them were poorly recorded, poorly published, or left simply with anecdotal information. Only with the advent of Skylab, and later Spacelab, did the idea emerge, and indeed the infrastructure permit, weightlessness to be considered as an ex tended tool for research into some fundamental mechanisms or processes as sociated with the effect of gravity on organisms at all levels. The initial hy pothesis to extrapolate from hypergravity through 1 x g to near 0 x g effects could no longer be retained, since many of the experiment results were seen to contradict the models or theories in the current textbooks of biology and physiology. The past decade has been dedicated primarily to exploratory research.
