A detection limit of 18 attograms was obtained within a one-second signal integration time. The concentration detection limit was 8.9 x 10−14 mole per liter. An average of one-half rhodamine 6G molecule was present within the 11 pL excitation volume. However, during the signal integration time a total of 22,000 analyte molecules passed through the excitation in a 0.42 microliter volume.

Indirect Laser-induced fluorescence detection and capillary electrophoresis were used to quantify lactate and pyruvate in single red blood cells. The assay of specific enzyme activities was achieved by monitoring the highly fluorescent enzymatic reaction product, NADH. LDH activity was found not to be a unique marker for diagnosis of leukemia. Reactions of single LDH-1 molecules were investigated by monitoring the reaction product with LIF detection.

Book excerpt: The feasibility of using laser induced fluorescence to measure total S2 number densities in a laboratory environment is studied. A rate equation analysis modeling transitions within the B to the 3rd power negative summation u-to the 3rd power negative summation g system of S2 is developed and applied to model the frequency prime = 9, frequency double prime = 6 transition at 3214.6 angstroms. Excitation spectra were obtained over the range of 2890 to 2810 angstroms clearly showing the frequency double prime = 0, frequency prime = 8 and frequency double prime = 0, frequency prime = 9 absorption bands. Fluorescence scans of the frequency prime = 8, frequency double prime = 1 through 24 and frequency prime = 9, frequency double prime = 1 through 25 were taken. After these broad spectra were identified, further investigations of the frequency prime = 9, frequency double prime = 6 transition included fluorescence intensity versus temperature, laser power, and pressure. Additional measurements of quenching, radiative lifetime and self absorption are made.

Chapter 2. Initial studies demonstrated the ability to synthesize a BODIPY- tagged zirconocene complex in which the fluorescence of the tag was not quenched by the metal center and thus was suitable for single-molecule studies. Though treatment of microscope coverslips and subsequent single-molecule analysis revealed individual molecules, XPS data indicated that the complex was not binding intact to the glass surface. The binding mode of a tagged palladium complex that had been observed at the single-molecule was established by PM-FTIRRAS, suggesting the formation of a network incorporating the complex. Chapter 3. A diethylenetriamine ligand capable of being used to tether a platinum center to a glass surface for single-molecule studies of a ligand exchange reaction was synthesized. Subsequent work using amine and thiol groups tethered to a glass surface demonstrated the viability of utilizing sulfur-based ligands tethered to a glass surface for the single-molecule observation of a ligand exchange reaction at a platinum center. A thiourea ligand capable of being tethered to the surface of a glass microscope coverslip was synthesized or purified from commercial sources for use in the single-molecule ligand exchange studies. A form of BODIPY containing a triethoxysilyl group for tethering the fluorophore to the surface of a coverslip was synthesized and an optimum concentration range was found for dispersing the molecule on a coverslip surface for the determination of a delay between a molecule binding to the surface and emitting fluorescence to lead to its detection. Chapter 4. A pair of test molecules containing isolated donor and acceptor BODIPY molecules, as well as a molecule containing both donor and acceptor molecules, was synthesized for the evaluation of their feasibility as FRET partners for the single-molecule analysis of the Trost-Tsuji reaction. Single-molecule and ensemble-level studies of these molecule revealed reduced fluorescence of the FRET pair, and demonstrated the feasibility of differentiating the three types of molecules at the single-molecule level based on their fluorescence intensities by use of appropriate filter sets.

For example, various workers have achieved direct measurement of quantum jumps, Doppler sidebands, and other fundamental phenomena such as ion crystallization of ions in vacuum trapped by electromagnetic fields. By using laser induced fluorescence and a novel hydrodynamically-focused flow to confine the molecules and reduce the scattering volume, single molecules of the protein B-phycoerythrin with the equivalent of 25 rhodamine 6G chromophores have also been detected. In contrast to the far- field optical approach, recent advances with various near-field spectroscopies such as scanning tunnelling microscopy (STM) have provided images of single molecules of benzene and CO on Rh surfaces and images of liquid crystal molecules on graphite to name a few examples.

Book excerpt: The purpose of this research was to develop laser-induced fluorescence detection in high-throughput screening of heterogeneous catalysts and single cells analysis. First, a high-throughput in situ screening approach for solid heterogeneous catalysts by laser-induced fluorescence imaging is introduced. We demonstrated LIFI has good detection performance and the spatial and temporal resolution needed for high-throughput screening of heterogeneous catalysts. We further applied the LIFI screening system for combinatorial discovery of heterogeneous catalysts and reaction condition optimization for naphthalene oxidation, as well as combinatorial study of catalytic performance of zeolites in acylation of aromatics. In the second part of this dissertation, we used laser-induced native fluorescence coupled with capillary electrophoresis (LINF-CE) and microscope imaging to study the single cell degranulation. On the basis of good temporal correlation with events observed through an optical microscope, we have identified individual peaks in the fluorescence electropherograms as serotonin released from the granular core on contact with the surrounding fluid.
