Book excerpt: The feasibility of sperm cells to act literally as gene vehicles was discussed using one of the most modern techniques in this aspect which is known as restriction enzyme mediated integration sperm mediated gene transfer (REMI-SMGT)technique. Several objectives were pursued in this book; such as to test the validity of REMI-SMGT, to evaluate the efficiency of this technique in generating mammalian species rather than the sheep that made only by one group and to observe the possibility of doing so by using rabbits as a model for this approach. Add to that, it was aimed to reduce the cost of REMI-SMGT by substituting liposomes and highly cost effective media with a high efficient, non-cost effective substitute. Moreover, it was directed to identify if any band substitution was occurred by simple seminal proteins - exogenous DNA incubation by one gel double stained non-radioactive method to identify the role of seminal proteins charge on the nature of seminal fluid inhibitory effect that exerted against the invading DNA. This might help us to approach closely to the nature of inhibitory activities in seminal proteins that prevent the entry of foreign DNA into the head of the sperm.

Although mice including genetically modified mice offer many possibilities, other non-murine species are absolutely required in some circumstances. Rabbit is one of these species, which has been widely used in biomedical studies. This animal is genetically and physiologically closer to humans including cardiov- cular system and metabolism characteristics. Rabbit is thus more appropriate than mice to study some diseases such as atherosclerosis and lipid metabolism. Because of its larger size, surgery manipulation, bleeding, and turn-over studies are much easier performed in rabbits than in mice. Furthermore, transgenic rabbits can be produced using microinjection and other methods such as lentiviral v- tors. Cloning in rabbits has been proved possible, even though still laborious and time-consuming. Hopefully, functional rabbit ES cell lines will be available in the coming years. Gene deletion or knock-out in rabbits will then become possible.
