Book excerpt: The current volumes of The Hippocampus reflect the prodigious amount of work aimed at discovering the functions of this structure over the past decade. The hippocampus ideally lends itself to two types of study: (1) because of its regular and relatively simple cytoarchitecture, it can serve as a model for cortical processing in general, and (2) because of its size and central location, the role of the hippocampus in the total ecology of brain function poses an important challenge. We attempted to divide the contributions to Volumes 3 and 4 according to these two types of experimental aims. As always, however, when one makes dichotomies, one finds them inadequate in treating certain data and our attempt is no exception. There are contributions that do not fit the classification and there are others which fit both. In addition, there are manuscripts which we wanted to include but which the authors were not ready to submit at this time. Larry Squire, Mortimer Mishkin, and others are making important contributions which donot appear in these volumes except among references throughout. But this was also the case for Volumes 1 and 2 where we sorely miss the irreplaceable contributions of James Olds, Ross Adey, and Brenda Milner. Volume 4 is concerned primarily with the role of the hippocampus in the ecology of the brain in regulating behavior and experience. The contri butions have the potential to raise the level of our understanding considerably.

Book excerpt: Circadian rhythms play a prominent role in psychiatric health with disruption in rhythms associated with poor mental health. Corticosterone (CORT) is an important hormone in entraining the biological rhythms of many cells throughout the body and coordinating peripheral rhythms with the central master clock. In aim 1, we tested the hypothesis that excess CORT during the circadian trough would lead to a flattening of period genes (Per1 and Per2) rhythms in limbic brain areas, and thus impact emotional behaviors. This was not expected to be mirrored when excess CORT occurred during the circadian peak. Male rats were injected daily with 2.5 mg/kg CORT or vehicle for 21 days at either ZT0 or ZT12 and sucrose preference, open field, and forced swim behaviors measured during the dark phase of the light cycle. After three weeks of injections, a reduction in sucrose preference was observed in animals injected with CORT at ZT0 and the reduction significantly correlated with reductions in Per2 mRNA expression in the central amygdala (CeA) and bed nucleus of the stria terminalis (BNST). No changes in behavior were observed in animals injected with CORT at ZT12, and period gene expression was mostly unaffected except for an increase in Per2 within the prefrontal cortex. In aim 2, we hypothesized that disruption of limbic period genes in the CeA or BNST would be sufficient to induce behavioral changes. DsiRNA was used to directly reduce Per2 levels in either the CeA or BNST and behavior was assessed. Despite reductions in Per2 expression in the CeA, no behavioral changes were observed. In contrast, a reduction in Per2 expression in the BNST was sufficient to reduce sucrose preference. The results demonstrate that CORT significantly contributes to the circadian expression of Period genes in certain limbic brain areas and disruption in diurnal CORT or Per2 expression can lead to impaired emotional behavioral responses.

Both structurally and functionally, there are many peculiarities that differentiate the brain from all other organs. The brain is our connection to the world around us and by governing nervous system and higher function, any disturbance induces severe neurological and psychiatric disorders that can have a devastating effect on quality of life. Our understanding of the physiology and biochemistry of the brain has improved dramatically in the last two decades. In particular, the critical role of cations, including magnesium, has become evident, even if incompletely understood at a mechanistic level. The exact role and regulation of magnesium, in particular, remains elusive, largely because intracellular levels are so difficult to routinely quantify. Nonetheless, the importance of magnesium to normal central nervous system activity is self-evident given the complicated homeostatic mechanisms that maintain the concentration of this cation within strict limits essential for normal physiology and metabolism. There is also considerable accumulating evidence to suggest alterations to some brain functions in both normal and pathological conditions may be linked to alterations in local magnesium concentration. Stressful stimuli also appear to exert their effects by acting upon individuals with susceptible genotypes. Over the past 50 years, animal models have been developed to study these dynamic interactions between stressful stimuli and genetically susceptible individuals during prenatal and postnatal development and into adulthood. Stress and Mental Disorders: Insights from Animal Models begins with a discussion of the history of psychiatric diagnosis and the recent goal of moving toward precision psychiatry, followed by a review of clinical research on connections between stressful stimuli and the development of psychiatric disorders. Chapters are also included on neuroendocrine, immune, and brain systems involved in responses to stress. Additional chapters focus on the development of animal models in psychiatry and the susceptibility of the developing organism to stressful stimuli. Subsequent chapters are devoted to animal models of specific stress-sensitive psychiatric disorders, including schizophrenia, autism spectrum disorders, bipolar disorder, anxiety disorders, depression, and post-traumatic stress disorder. These chapters also focus on identification of promising molecular targets for development of new drug therapies. The section concludes with a chapter on animal models of resilience to stress-induced behavioral alterations as a newer approach to understanding why some animals are susceptible to stress and others are resilient, even though they are essentially genetically identical.
