Book excerpt: This Special Issue gathers original studies (eight articles and one review) that aim to improve our knowledge concerning mycotoxin toxicity. The most recent research concerning the impact of zearalenone (ZEA) and the immune response is presented in a review that aims to explore the immunotoxicity produced by ZEA on different types of immune cells (phagocytes related to innate immunity and lymphocytes related to acquired immunity) as well as on immune organs, and also to identify the mechanism of action used by ZEA to modulate the immune response. The role of zearalenone in macrophage-mediated innate immunity after bacterial lipopolysaccharide (LPS) stimulation was the subject of another study, suggesting that the intake of ZEA-contaminated food might result in decreasing innate immunity. Another important study related to the co-exposure to mycotoxins and other food/feed contaminants showed that combining patulin with cadmium induces enhanced hepatotoxicity and nephrotoxicity both in vitro and in vivo, while the exposure to both deoxynivalenol and porcine circovirus has an additive effect on inflammatory cytokines by inducing the mRNA expression of interleukine-1 beta (IL-1β) and interleukine-6 (IL-6). The use of feed compounds rich in bioactive constituents for mitigating the mycotoxins effects in swine was the subject of other three studies herein, which have shown the efficacy of antioxidants from dietary grapeseed and sea buckthorn meal waste to decrease the toxicity of a diet contaminated with aflatoxin B1 or both aflatoxin B1 and ochratoxin A at the level of organs involved in the metabolism and excretion (liver and kidney), as well as at the level of secondary lymphoid organs (mesenteric lymph nodes). The use of different models (cell co-culture model, rat model) are recommended by another two studies in order to evaluate the ochratoxin A toxicity or for better prediction of the mycotoxins risk for human cancer.

Mycotoxins occur more frequently in areas with a hot and humid climate, favourable for the growth of molds, they can also be found in temperate zones.In comparison to monogastric species, ruminant animals are generally less susceptible to the adverse effects caused by contamination of feeds with mycotoxins because presence of rumen microflora in rumen. However, a number of mycotoxins resist rumen degradation, causing distinct clinical signs of intoxication. Dairy cattle can be exposed to different number of mycotoxins because complex feeding diet originating from different feed materials such as roughage and concentrates. Exposure of diary catte to these mycotoxins may result in unexpected health problem. Due to apre-existing negative energy balance, cows in the transition period are considered to be particularly sensitive to the exposure to feeds. The variation of mycotoxinoccurence due to different factors, including agricultural practices and climatic conditions. Aflatoxins (AFs) are found in maize and peanuts, aswell as in tree nuts and dried fruits. OTA is found mainly in cereals, but significant levels of contamination may also occur in wine, coffee, spices and dried fruits. Other products of concern are beans, roasted coffee and cocoa, malt and beer, bread and bakery products, wines and grape juices, spices, poultry meat and kidneys, pig kidneys and pork sausages. The production of secondary metabolism is the effect of Fungal metabolism Secondary metabolites, are formed from a relatively small number of branch points of primary metabolic pathways. Among them some of these secondary metabolites are used as antibiotics, while others are very toxic and carcinogenic to humans and animals.Economic impact associated with mycotoxicosisincluding:Slower growth rates, Poor conception rates, inconsistentmanure, Increased disease susceptibility, Reduced production performance, loss of animal and human life, veterinary and public cost of treatment.

Book excerpt: According to the presented studies, the health condition of animals in rearing and breeding should be regularly monitored. This would allow early detection of delicate deviations in the body of clinically healthy individuals. Unfortunately, regular monitoring of the health of animals in commercial production is not performed. It follows that this type of research should be an introduction to further, more inquisitive steps. This can form the basis for further courses of action, indicating which organs or tissues field doctors or researchers should be interested in and what to pay attention to in order to find the correct answer, concerning the situation in the animal body. In the future, we should determine biomedical markers for use in precision veterinary medicine. In human medicine, this has been practiced with great success. The problem, however, is that we are getting to know more and more substances produced by mold fungi. This causes a build-up of new interpretative problems, causing health conditions (diagnosis), as well as analytical problems. To fully understand the results we need new techniques to assess toxicological and chemical hazards, including those related to undesirable substances. We need a solid knowledge of the biological pathways underlying the toxicity and tolerance to interference factors toxicological processes. We hope that the presented study will allow for a better understanding of mycotoxicoses that bother us and our animals, which will allow for more effective preventive actions.

Considering their potential toxic and carcinogenic effects, mycotoxin exposure assessment has particular importance in the context of health risk assessment. The magnitude of a given exposure allows the derivation of the associated risk and the potential for the establishment of a disease. Although food ingestion is considered a major route of human exposure to mycotoxins, other contexts may also result in exposure, such as specific occupational environments where exposure to organic dust also occurs due to the handling of organic materials. Animals could be exposed to mycotoxins through consumption of contaminated feed, subsequently entering in the food chain and thus constituting a source of exposure to humans. Human biomonitoring is considered a new frontier for the establishment of the human internal exposure to mycotoxins. Although several studies have summarized the potential outcomes associated with mycotoxin exposure, major gaps in data remain in recognizing the mycotoxins that are the cause of diseases. This book contributes provides research that supports the anticipation of potential consequences of the exposure of humans and animals to mycotoxins, future risk assessments, and the establishment of preventive measures.

As part of a comprehensive effort to curtail the adverse health effects posed by mycotoxins, substantial research has been conducted to determine the mechanism of action of mycotoxins. Although much information has been obtained regarding the action of several mycotoxins, future research topics should continue to address several areas of critical concern. In vitro studies in different cell lines could detail and explain many of these mechanisms, while in vivo can give a real scenario in the development of a toxic effect. This Special Issue of Toxins collected the most recent reports on the mechanism of action of mycotoxins on single or combined mycotoxins studied in vivo or in vitro, the identification of known and unknown mycotoxins metabolites and other metabolites in different cell lines and animals or matrices (including organs, urine, or blood), and the development of analytical skills to study these mechanisms. A total of eleven papers, eight research papers and three review articles, are included in this reprint.
