Book excerpt: Chile (capsicum annuum L.) is a major cash crop grown in New Mexico for its edible and ornamental pods. Its farm gate value in 2014 was approximately $38 million. Rhizoctonia solani (RS) is a soilborne fungal pathogen that causes seed rot and damping off in chile, leaving bare patches in affected fields. Under no management, plant infection in a field increases over time. Extreme cases necessitate replanting of fields, increasing production costs. This study characterized the virulence of RS isolates recovered from chile and rosemary and used the most virulent isolate to evaluate the efficacy of selected microbial seed treatments. RS-susceptible chile seeds (var. New Mexico 6-4) were surface-disinfested in 10% sodium hypochlorite solution for 15 min and either pre-germinated (PG) or not pre-germinated (NPG). Microbial experiments were two-way factorial treatment structures with complete randomized block design. Data were subjected to ANOVA. Microbial seed treatments consisted of combinations of 0.5% solutions of two species of Bacillus, 1% suspensions of four species of arbuscular mycorrhizal fungi in the genus Glomus, two species of Trichoderma and water only. After microbial treatment, PG and NPG seeds were placed into petri dishes or soil that was either infested with RS or not. Shoot length and fresh weight were collected 30 days after sowing treated NPG seeds and a disease rating scale was developed to quantify the level of infection in treated PG seeds every 4 days for a period of 12 days. In all tests, there was no seed rot or damping off in control treatment due to RS (non-treated and non-inoculated NPG or PG seeds). Conversely, all treated NPG and PG seeds in RS-infested soil and petri plates containing RS displayed severe symptoms of seed rot and damping off. None of the microbial treatments reduced seed rot or damping off caused by the extremely virulent isolate of RS used (RS AG-4, HGI). Lesions on seedlings grown in RS-infested soil were re-cultured and tested on fresh PG seeds, where symptoms and signs of seed rot and damping off reoccurred and pathogen was re-isolated.
