Book excerpt: Endurance exercise can affect immune function and eccentric exercise can result in increased oxidative stress. Fish oil has anti-inflammatory properties, and can be beneficial in conditions such as cardiovascular disease and may also modulate immune and oxidative responses to exercise. However, the effects of fish oil supplementation on immune function and oxidative stress after an exercise bout are currently unknown. This thesis investigated the effect of supplementation with fish oil capsules on the immune response to a bout of endurance exercise and on markers of oxidative stress following a bout of eccentric exercise. In the first study, sixteen males underwent 6 week's supplementation with either fish oil or placebo followed by an endurance exercise bout. Blood samples were taken pre-exercise, immediately, 1h and 3h post-exercise. Samples were analysed for a range of immune and inflammatory markers. Fish oil increased PBMC IL-2 and NK cell function compared to control in the recovery period. In the second study, twenty males underwent 6 week's supplementation with either fish oil or placebo followed by a bout of eccentric exercise. Blood samples were taken pre-exercise and immediately, 24h, 48h and 72h post-exercise. Samples were analysed for a range of markers of oxidative stress and muscle damage. Fish oil decreased plasma TBARS and DNA damage in the days following the exercise bout. This thesis demonstrates that fish oil supplementation increases PBMC IL-2 production and NK cell activity in the recovery period after a bout of endurance exercise and also reduces plasma TBARS and induced DNA damage after a bout of eccentric exercise. These results show that fish oil can protect against infection with URTI in the recovery period following exercise and may also protect against exercise-induced oxidative stress. However, this may also inhibit any oxidative stress-mediated adaptation to exercise.

Book excerpt: The purpose of this study was to investigate the effect of four weeks of fish oil supplementation and four weeks of fish oil supplementation and exercise training on measures of oxidative stress, including superoxide dismutase (SOD), glutathione peroxidase (GPx), and myeloperoxidase (MPO). Twenty-two normal to overweight (BMI=23.0-29.9) individuals (10 males, 12 females) age 18-45y participated in this 4 week intervention study. Subjects were randomly assigned to a fish oil supplementation group (n=10) or a fish oil supplementation and exercise group (n=12). Fish oil supplementation for both groups involved the consumption of 8 gel caplets/day of fish oil supplement (4g/day). Exercise training consisted of 45 min/d, 5d/week of brisk walking or mild jogging at 60% of VO2 max. Variables were measured 1) prior to intervention, 2) after an acute exercise bout prior to intervention, 3) after the 4 wk intervention, and 4) after an acute exercise bout following intervention. No significant differences (p [greater than] 0.05) were found between groups after intervention in fasted samples or acute exercise samples for SOD, although there was a trend for a group*time interaction in acute exercise samples (p=0.08). In fasted samples there were no differences between groups for GPx(p [greater than] 0.05) or MPO (p [greater than] 0.05). Book excerpt: Asthma, a chronic inflammatory disease of the airways, affects nearly 25 million Americans. The vast majority of these patients suffer from exercise-induced bronchoconstriction (EIB), a complication of asthma. Although traditionally treated pharmacologically, nutritional strategies provide a promising alternative for managing EIB as the prevalence of asthma may be due in part to changes in diet. Our objective was to determine the effects of novel nutritional strategies on hyperpnea-induced bronchoconstriction (HIB) in asthmatic individuals. HIB uses rapid breathing to identify EIB in a research or clinical setting. Fish oil, a combination of the omega-3 fatty acids eicosapentaenoic acid (EPA) and docsahexaenoic acid (DHA), has been shown to be effective in suppressing EIB. However, its use in combination with other nutritional supplements, the optimal fish oil formula, and its effect on smooth muscle contractility have not been fully explored. An in vivo study (study 1) was conducted in individuals with both asthma and HIB to determine whether a combination of fish oil and vitamin C was more effective than either one alone in alleviating HIB. Pulmonary function was significantly improved with both fish oil and the combination treatment but not with vitamin C alone. In study 2, individuals with both asthma and HIB were supplemented with DHA alone since the optimal formula for fish oil has yet to be ascertained; previous in vitro studies have suggested DHA may be the more potent omega-3 fatty acid in fish oil. However, no significant changes in pulmonary function or airway inflammation were seen with DHA supplementation. For study 3, canine airway smooth muscle tissue was treated with fish oil to determine the in vitro effect of fish oil on smooth muscle contractility. Acute treatment with fish oil relaxed smooth muscle strips that had been contracted with 5-hydroxytryptamine. These minor relaxations in smooth muscle tension with fish oil may represent significant changes at the level of the smaller airways. These studies have confirmed that fish oil represents a viable treatment modality for asthmatic individuals with EIB and suggest that fish oil may influence airway smooth muscle contractility.
