Book The Dynamic Nature of Mitochondria

Download or read book The Dynamic Nature of Mitochondria written by Andreas S. Reichert and published by CRC Press. This book was released on 2024-04-15 with total page 211 pages. Available in PDF, EPUB and Kindle. Book excerpt: Mitochondrial research has exploded over the last ~150 years. This book gives an amazing view on a conceptual change in our understanding of mitochondrial biology. It becomes clear that mitochondria are extremely dynamic in nature, controlling life at multiple levels. Mitochondria rule energy conversion, adapt cells well to changing stress and nutrient conditions, and regulate many cellular processes including immunity. The dynamic nature of mitochondria occurs at an intramitochondrial level but also includes its ability to interact with other organelles and to modulate multiple signalling pathways. It is thus not surprising that alterations or inabilities to ensure this dynamic behaviour is linked to ageing and human diseases. The following sections give an updated view on mitochondria: Mitochondrial ultrastructure: molecular mechanisms shaping the inner membrane Mitochondrial cristae and lipid dynamics: from super-resolution microscopy to lipid-OXPHOS interplay Mitochondrial control of cellular homeostasis: from redox signalling to interorganellar contact sites Mitochondria in health and disease: from mtDNA release to Complex I assembly Advanced methods in mitochondrial biology and metabolism research Integrative view on mitochondrial research and outlook The field of mitochondrial research has always been full of surprises and has helped science to advance tremendously. It developed hand in hand with landmark developments in technology, such as super-resolution microscopy (nanoscopy), and is currently influencing an increasing number of scientific disciplines. There is still much ‘new’ to find out about this ‘old’ organelle and I think that you can find interesting and also unexpected aspects of mitochondrial biology in this book. I hope the book will enhance your scientific curiosity and inspire your own research.

Book Investigating the Dynamic Nature of Mitochondrial Networks in C Elegans

Download or read book Investigating the Dynamic Nature of Mitochondrial Networks in C Elegans written by Saroj G. Regmi and published by . This book was released on 2015 with total page 270 pages. Available in PDF, EPUB and Kindle. Book excerpt: Mitochondria are dynamic organelles that undergo constant fusion and fission events and this dynamic homeostasis defines the overall morphology of a mitochondrial network. Although these fusion and fission events are important for the maintenance of mitochondrial function, the physiological role of these processes remains poorly characterized. Using the nematode C. elegans as a model organism, this thesis explores various aspects of mitochondrial dynamics by: (i) examining its role in the context of aging; (ii) investigating the localization of CED-9, a Bcl-2-family member and regulator of mitochondrial dynamics; and finally, (iii) developing tools to study mitochondrial dynamics. Using transgenic animals expressing a matrix-targeted GFP under the control of a muscle-specific promoter (P[subscript myo-3] mitoGFP), I observed that body-wall muscles display a tubular inter-connected mitochondrial network that fragments with increasing age. Therefore, I aimed to determine if mitochondrial morphology is a suitable bio-marker of aging. To that end, I analyzed various aspects of mitochondrial morphology (e.g. mitochondrial length, circularity, area) in animals raised at different temperatures. I found that in animals grown at 15°C, the onset of mitochondrial fragmentation occurred later in life than those animals grown at 25°C. Furthermore, I analyzed mitochondrial morphology in both short- and long-lived animals. To complement this analysis, I characterized mitochondrial morphology in five- and seven-day old animals to determine if those displaying a fragmented mitochondrial morphology exhibit shorter lifespans when compared to animals displaying tubular mitochondrial morphology. The results suggest that while mitochondria in body-wall muscles undergo age-dependent fragmentation and a loss in volume, these changes are not a cause of aging but rather a consequence of the aging process. Using super-resolution microscopy, I investigated whether CED-9 localizes not only to mitochondria but also to the ER. Data collected from this study indicate that a subpopulation of CED-9 is dually localized to mitochondria and the ER. This may be important for the maintenance of ER-mitochondria contact. Finally, I describe the generation of fluorescent markers, including a photo-convertible DENDRA-based construct, as a means to study aspects of mitochondrial dynamics. Collectively, these represent valuable tools for the study of mitochondrial fusion and fission, as well as mitophagy.

Book Organelle Contact Sites

Download or read book Organelle Contact Sites written by Mitsuo Tagaya and published by Springer. This book was released on 2017-08-16 with total page 254 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book provides the first comprehensive coverage of the quickly evolving research field of membrane contact sites (MCS). A total of 16 chapters explain their organization and role and unveil the significance of MCS for various diseases. MCS, the intracellular structures where organellar membranes come in close contact with one another, mediate the exchange of proteins, lipids, and ions. Via these functions, MCS are critical for the survival and the growth of the cell. Owing to that central role in the functioning of cells, MCS dysfunctions lead to important defects of human physiology, influence viral and bacterial infection, and cause disease such as inflammation, type II diabetes, neurodegenerative disorders, and cancer. To approach such a multifaceted topic, this volume assembles a series of chapters dealing with the full array of research about MCS and their respective roles for diseases. Most chapters also introduce the history and the state of the art of MCS research, which will initiate discussion points for the respective types of MCS for years to come. This work will appeal to all cell biologists as well as researchers on diseases that are impacted by MCS dysfunction. Additionally, it will stimulate graduate students and postdocs who will energize, drive, and develop the research field in the near future.

Book Cyclin Dependent Kinase 5 Cdk5

Download or read book Cyclin Dependent Kinase 5 Cdk5 written by Nancy Y. Ip and published by Springer Science & Business Media. This book was released on 2009-02-28 with total page 326 pages. Available in PDF, EPUB and Kindle. Book excerpt: Cyclin Dependent Kinase 5 provides a comprehensive and up-to-date collection of reviews on the discovery, signaling mechanisms and functions of Cdk5, as well as the potential implication of Cdk5 in the treatment of neurodegenerative diseases. Since the identification of this unique member of the Cdk family, Cdk5 has emerged as one of the most important signal transduction mediators in the development, maintenance and fine-tuning of neuronal functions and networking. Further studies have revealed that Cdk5 is also associated with the regulation of neuronal survival during both developmental stages and in neurodegenerative diseases. These observations indicate that precise control of Cdk5 is essential for the regulation of neuronal survival. The pivotal role Cdk5 appears to play in both the regulation of neuronal survival and synaptic functions thus raises the interesting possibility that Cdk5 inhibitors may serve as therapeutic treatment for a number of neurodegenerative diseases.

Book Mitochondria in Health and Disease

Download or read book Mitochondria in Health and Disease written by Paolo Bernardi and published by Frontiers Media SA. This book was released on 2021-09-01 with total page 299 pages. Available in PDF, EPUB and Kindle. Book excerpt: Mitochondrial biology reinvented itself and became a new world that has attracted new scientists influencing every field of biomedical research. Mitochondrial research is growing and changing, as reflected by the exponential rise in the number of conferences covering mitochondrial biology and the role of mitochondria in diseases ranging from neurodegenerative diseases, metabolic diseases and genetic muscular dystrophies toimmunopathologies and cancer. As the awareness of the essential role of mitochondria in pathology rose, a demand for new approaches to measure mitochondrial function resulted in the robust development of new forms of microscopy and spectroscopy that opened windows into previously unknown aspects of mitochondrial biology. Two Conferences provided an outstanding representation of this state of affairs, the Gordon Research Conference Mitochondrial Dynamics and Signaling (Ventura, California March 17-22, 2019) and the FASEB Conference Mitochondrial Biogenesis and Dynamics in Health and Disease (Palm Springs, California May 19-24, 2019). These conferences well reflected the explosion of the field of mitochondrial communication within the cell, between cells and across organs, as well as the budding of a new field on the definition of individual mitochondria and the identification of subtypes with diverse structural features that may serve different specific functions. Through our participation in these meetings, we conceived the idea to cover some of these topics in the Research Topic “Mitochondria in Health and Disease” of Frontiers in Physiology - Mitochondrial Research Specialty Section. Fitting the tradition of Frontiers, our contributors have generated a platform incuding both solid data and new concepts, as radical and courageous as they can be. We are pleased with the outcome and we hope that our readers will share our enthusiasm.
