It is natural that as the field of biochemical dynamics has achieved new levels of refinement its students have increasingly adopted the concepts and methods of transition-state theory. Indeed, every dynamical problem of biochemistry finds its most elegant and economical statement in the terms of this theory. Enzyme catalytic power, for example, derives from the interaction of enzyme and substrate structures in the transition state, so that an understanding of this power must grow from a knowledge of these structures and interactions. Similarly, transition-state interactions, and the way in which they change as protein structure is altered, constitute the pivotal feature upon which molecular evolution must turn. The complete, coupled dynamical system of the organism, incorporating the transport of matter and energy as well as local chemical processes, will eventually have to yield to a description of its component transition-state structures and their energetic response characteristics, even if the form of the description goes beyond present-day transition-state theory. Book excerpt: The writing of this book was prompted by the need for a comprehensive of current data on organic acids suitable for both newcomers and collection established researchers in this field. The only previous text of the kind was the excellent review by Nordmann and Nordmann (1961), and at that time the main method of analysis was paper chromatography with liquid chromatography being used in a limited way. Only three diseases in which organic acids accumulate were known (primary hyperoxaluria, phenylketonuria and alcaptonuria). Since then, with the development of gas chromatography and mass spectrometry, and the further development ofliquid chromatography, knowledge concerning the nature of the organic acids in physiological fluids has been greatly extended. At the same time, the number of organic acidurias has increased dramatically, there being now some 40-50 known diseases of this type. During the past 15 years or so, there have been several reviews, dealing with either specific diseases or groups of diseases (Gompertz, 1972, 1974; Tanaka, 1975), or presenting the proceedings of symposia (Stern and Toothill, 1972) or workshops (Marner et al. , 1974). This present text deals comprehensively and in detail with the organic acids in human physiological fluids in health and in disease states, and is particularly concerned with the methods necessary for their separation, determination and indentification.

Book excerpt: In May of 1991, Victor Van Buren, who was then with Springer Verlag in New York City, asked us for timely topics in the earth sciences that would be appropriate for publication as a book. We all quickly agreed that recent interest and research activity on the role of organic acids in geological processes would make a timely book on this diverse and controversial topic. As coeditors, we outlined chapter topics for such a book that maintained a good balance between geological and geochemical interests. Specific authors were then sought for each of the chapter topics. We had exceptional success in getting leading researchers as authors, and their response was universally enthusiastic. This approach has been most gratifying in that it provides a cohesion and conciseness that is not always present in books representing compilations of papers from symposia. This book does not resolve the controver sies that exist regarding the significance of organic acids in geolog ical processes. However, it does present both sides of the controver sies in terms of available data and current interpretations. Readers may judge for themselves and envisage research necessary to resolve these controversies in the future. We thank the authors of this book for their participation, dedication, and cooperation.
