Available in PDF, EPUB and Kindle. Book excerpt: Interest and research in urinary enzymology were incited about three decades ago by reports that urinary enzymes are elevated in diseases of the kidney and urinary tract. Of the more than 40 hydrolases, oxidoreductases, transferases, and lyases identified in human and animal urine, only ten or so are being used as diagnostic indicators. Recognition of the quantitative distribution of enzymes in the various anatomical and functional parts of the nephron and advances in our understanding of the handling of proteins by the kidney have made it possible to associate urinary enzyme activity patterns with physiological and pathophysio logical functions of the nephron. Confidence in the diagnostic value of urinary enzymes is not unanimous among clinicians and among scientists. The main reason for the difference in opinion may well be that the variability in data exceeds the variability one is accustomed to in the diagnostic enzymology of blood plasma enzymes. In contrast to plasma enzymes, which are protected by an "enzyme friendly" milieu, enzymes released into the urine encounter an "enzyme hostile" environ ment: no or little protective protein, variable pH, variable volume, variable metabolite and salt concentrations, variable concentrations of enzyme in hibitors. Through advances in methodology some of these factors can now be controlled; standardization of urine collection periods and preanalytical treat ment are as important as optimization of assay methods.

Book Standard Methods of Clinical Chemistry

Download or read book Standard Methods of Clinical Chemistry written by Samuel Meites and published by Elsevier. This book was released on 2013-10-22 with total page 307 pages. Available in PDF, EPUB and Kindle. Book excerpt: Standard Methods of Clinical Chemistry, Volume 5 presents a wide variety of approaches to analytical procedures in clinical chemistry. This 24-chapter volume discusses the principles, reagents, procedure, and calibration of various clinical chemistry methods. The first three chapters cover the basic protocols in clinical chemistry laboratories, including collection and preservation of specimens, error sources determination, and the automatic chemical analysis. These topics are followed by surveys on determination of blood ammonia, bilirubin, total and free cholesterol, sweat chloride, glucose, and blood and urine lead. Other chapters examine the analysis of magnesium, methemoglobin, osmolality, pH, phenylalanine, and alkaline and acid phosphatase enzymes. The final chapters focus on the methods of colorimetry and turbidimetry for total protein determination. This book is directed primarily toward clinical chemists.

Book Nutritional and Toxicological Significance of Enzyme Inhibitors in Foods

Download or read book Nutritional and Toxicological Significance of Enzyme Inhibitors in Foods written by Mendel Friedman and published by Springer Science & Business Media. This book was released on 2013-03-09 with total page 563 pages. Available in PDF, EPUB and Kindle. Book excerpt: Soybean protei ns are wi de 1 y used inhuman foods ina vari ety of forms, including baby formulas, flour, soy protein concentrates, soy protein isolates, soy sauces, textured soy fibers, and tofu. The presence of inhibitors of digestive enzymes in soy proteins impairs nutritional quality and possible safety of this impportant legume. Normal processing conditions based on the use of heat do not completely inactivate these inhibitors, so that residual amounts of plant protease inhibitors are consumed by animals and man. Inhibitors of digestive enzymes are present not only in legumes, such as soybeans, lima beans, and kidney beans, but also in nearly all plant foods, including cereals and potatoes, albeit in much smaller amounts. The antinutritional effects of inhibitors of proteolytic enzymes have been widely studied and can be ameliorated by processing and/or sulfur amino acid fortification. A more urgent concern is reports that rats fed diets containing even low levels of soybean-derived inhibitors, which are found in foods such as soy-based baby formulas, may develop over their lifespan pancreatic lesions leading eventually to neoplasia or tumor formation. On the other hand, recent stUdies suggest that certain enzyme inhibitors from plant foods may prevent cancer formation in other tissues. A key question, therefore, is whether inhibitors from plant foods constitute a human health hazard.
