Firstly, there is considerable confusion in the literature about the appearance of the different kinds of neuroglia by light and by electron microscopy, and this is complemented by widespread disagreements among distinguished neuropathologists about the international classification of tumours of the central nervous system. Secondly, there is an increasing volume of experiments on the physiology and biochemistry of tissue cultures of neurons and different kinds of neuroglia, whose validity depends upon the accurate identification of both the parent tissue and also of the cells subsequently growing in culture. The biochemical classifica tion in recent years has often tended to become independent of the cellular identification, which makes the use of the neuroglial cell names doubtful and the significance of the biochemical properties of the cells difficult to relate to the physiological properties in vitro or in vivo (Table 1).

Book excerpt: Several model systems have been used to understand the cellular and molecular mechanisms of differentiation of mammalian nerve cells. Each model system has unique advantages and disadvantages and is suited for the study of only certain aspects of differentiation. In this book, the techniques of these models and the usefulness and limita tion of each model system are discussed. An awareness of the use and misuse of each model system is important for a rational interpretation of data and for a reasonable comparison of data obtained from different model systems. With the use of clonal lines of neuronal cells and hybrid neural cells (neural cells x nonneural cells), many new concepts have emerged con cerning the regulation of differentiated functions, the relationship between the expressions of individual differentiated functions, and the relationship between differentiation and malignancy. It is becoming increasingly clear that a misregulation of synaptogenesis and synaptic remodeling and dysfunctional neuronal synapses are at the heart of several human diseases, both neurological disorders and psychiatric conditions. As synapses present specialized cellular junctions between neurons and their target cells, it may not come as a surprise that neural cell adhesion molecules (CAMs) are of special importance for the genesis and the maintenance of synaptic connections. Genes encoding adhesive molecules make up a significant portion of the human genome, and neural CAMs even have been postulated to be a major factor in the evolution of the human brain. These are just some of the many reasons why we thought a book on neural CAMs and their role in establishing and maintaining neuronal synapses would be highly appropriate for summarizing our current state of knowledge. Without question, over the near future, additional adhesive proteins will join the ranks of synaptic CAMs and our knowledge, and how these molecules enable neurons and their targets to communicate effectively will grow.

This is especially the case with regard to so-called "higher" functions such as per ception, memory or the coupling between sensory input and motor output. There are a number of different reasons for this. Some may be related to experimental limitations allowing the simultaneous recording of the activities of only a restricted number of neurones. But there are also conceptual difficulties hindering the transition from "single-neurone" schemes, in which neurones are assigned relatively specific tasks (such as feature detection), to more complex schemes of nerve cell as semblies (for a discussion of some of the difficulties see Abeles 1982; von der Malsburg 1981; Kriiger 1983). Whilst much is known about the basic properties and functions of single neu rones, whose operations we hope to understand in the foresee able future, this does not hold true in the same way for the working of large assemblies of neurones.
