Initially, phagocytosis, which comes from Greek words meaning “devouring cells,” was discovered in the late 19th century by Ilya Metchnikoff, who was awarded, together with Paul Ehrlich, the Nobel Prize in Physiology and Medicine in 1908 “in recognition of their work on immunity.” At that time Metchnikoff had already identified a function for phagocytes not only in host defense but also as scavengers of degenerating host cells during metamorphosis of tadpoles, thus providing one of the first descriptions of apoptotic cell clearance by macrophages (Kaufmann 2008). Since then, much has been learned about phagocytosis, and the previous several decades have witnessed outstanding progress in understanding the functions and the molecular mechanisms of phagocytosis. Two main types of targets are cleared by phagocytosis: microbial pathogens and dying cells. Rapid recognition and clearance of dying cells by phagocytes plays a pivotal role in development, maintenance of tissue homeostasis, control of immune responses, and resolution of inflammation. Clearance of dying cells can be divided into several stages, including sensing, r- ognition, binding and signaling, internalization, and immunological responses. In this book, our contributors address these different stages of dead cell cle- ance and examine how impaired clearance of dying cells may lead to human d- eases. We have attempted to provide sufficient cross-referencing and indexing to enable the reader to easily locate the ideas elaborated in the different chapters.

On the other hand, they contribute to essential trophic functions such as neural patterning, bone morphogenesis and ductal branching in mammary glands. Thus, macrophages are extremely versatile cells that can respond efficiently to tissue micro environmental cues by polarizing to distinct phenotypes, depending on the functions they need to perform. Indeed, functional diversity and plasticity are hallmarks of these cells. Macrophages may also play a detrimental role. An overwhelming body of literature has indicated their crucial role in pathogenesis. The list includes sepsis, cancer, metabolic syndrome, immunodeficiency, auto-immune disease-virtually impacting every major pathology that we know. These observations have suggested macrophages and their related molecules as potential targets in therapeutic applications. Available evidence proclaims macrophages as a key player in homeostasis, host defense and disease. Crucial developments in the past few years call for a re-evaluation and update of our understanding of macrophages. The present book is an endeavour that attempts provide state-of-the art knowledge of these cells in health and disease.

Book excerpt: Sixteen years have passed since the first Leiden Conference on Mononuclear Phagocytes. That first meeting led to new nomenclature, i. e. , the Mononuclear Phagocytes System (MPS), which was published in the Bulletin of the World Health Organization (R. van Furth, et aI. , Bull. WHO, 1972, 46: 845-852). The paradigm of the MPS has the advantage of being much more consistent than the Reticuloendothelial System (RES) (L. Aschoff, Ergebn. Inn. Med. Kinderheilk. 1924, 26:1), because cells with similar characteristics can be classified in the same category. The essence ota new concept is, according to Popper, that it must be susceptible to falsification. This has been attempted several times for the MPS by those who adhere to the RES. However, in biology no one can be certain of truth. Any theory must be based on experimental observations and/or theoretical consider ations and both should be presented in such a way that others can try to refute it, then or later. One may have a theoretical or a pragmatic preference for a theory which at a given time is regarded as possibly truc, but later it may be shown to be false. In other words, what we present in this volume is regarded as true at this moment, but others may consider it as not true and try to disprove it. Controversies often stimulate new research.
