The influence they have had on literature is hinted at by some of the quotations I have used as chapter headings. Their in fluence on folklore and on medicine has been even greater. The scientific study of alkaloids may be said to have begun with the isolation of morphine by SERTURNER in 1804. Since that time they have remained of great interest to chemists, and now in any month there appear dozens of publications dealing with the isolation of new alkaloids or the determination of the structures of previously known ones. The area of alkaloid biochemistry, in comparison, has received little attention, and today is much less developed. There is a certain amount of personal arbitrariness in defining "biochemistry", as there is in defining "alkaloid", and this arbitrariness is doubtless compounded by the combination. Nevertheless, it seems to me that in any consideration of the bio chemistry of a group of compounds three aspects are always worthy of attention pathways of biosynthesis, function or activity, and pathways of degradation. For the alkaloids, treatment of these three aspects is necessarily lopsided. Much has been learned about routes of biosynthesis, but information on the other aspects is very scanty. It would be possible to enter into some speculation regarding the biosyn thesis of all the more than 1,000 known alkaloids.

The purpose is to inform the reader about significant advances in the biology and metabolism of alkaloids in plants. Since alkaloids are generally referred to as "secondary metabolites," the reactions discussed are not, for the most part, involved with the main metabolic pathways. The reactions that we are interested in are pathways that have been developed for the formation of these secondary metabolites, using as their starting mole cules one of the compounds produced via a main or primary metabolic path way. The primary metabolic pathways are common to all plants, indeed to most living organisms, whereas the highly specialized branches leading to alkaloid formation are found in only about 10 to 20 % of the known plants. The reason for these diversities in plant metabolism is not clear; however, it seems likely that the formation of highly individualized and specialized pathways resulted as a response to the pressure of natural selection. Nevertheless, the genetic peculiarity that controls alkaloid production has provided many extremely interesting problems for scientists and consti tutes convincing evidence of nature's superior ability in biochemistry.

The alkaloids are also categorized by their molecular weights and structural types. Chapter 2, by Toh-Seok Kam, is a review of alkaloids derived from Malaysian flora. Malaysia's position near the Equator confers on it a tropical climate characterized by high temperatures, humidity, and rainfall, conditions favorable for plant life that has resulted in a rich flora of about 15,000 species of higher plants. This review concentrates on work published during the past twenty years and where appropriate compares the occurrence of alkaloids with studies of similar plants from countries neighboring to Malaysia, especially Thailand and Indonesia. Chapter 3, by Jie Jack Li, presents a collection of very interesting total syntheses of naturally occurring indole alkaloids where palladium chemistry plays a central role in the syntheses. Five different types of palladium-mediated reactions are treated: (1) oxidative cyclization reactions promoted by palladium (II) species; (2) transmetallation reactions with organoboranes, organostannanes, and organozinc reagents; (3) inter- and intramolecular Heck reactions; (4) reactions with &pgr;-allylpalladium as the intermediate; and (5) reactions using C-N bond formation as the key step for the synthesis.

Thus, the alkaloids have been isolated from marine and terrestrial sources and human beings have had the aptitude to determine the chemical structure of many derivatives of simple and big complexity as well as observing the biological effects of every compound in the living organism. Different natural sources as well as the synthesis of many alkaloids of big therapeutic activity have been the basis for the hundreds of drugs that are applied successfully in the scope of the health and combating diverse diseases. Alkaloids' low cytotoxicity in many cases and versatility in transforming into stable salt have generated diverse drugs of easy administration in the organism without the side effects associated with the ingestion of organic and inorganic salt of difficult tolerance. In this sense, this contribution covers several chapters which include: mechanisms and strategies against cancer, wherein certain types of alkaloid take control of important and selective form; the use of boldine as the alkaloid of current reference in the traditional medicine and used actively as natural antioxidant; alkaloids from vegetable origin as coming from the Amaryllidaceae; curious brominated alkaloids from marine sources between several outstanding examples; alkaloids derived from the Erythrina including the synthesis and pharmacological applications; the technological approaches of some derivatives originated from Tropane; an interesting contribution of the application of Trabectedin as alkaloid of clinical use in the treatment of ovarian cancer; the mention of a small group of alkaloids called oxoisoaporphines as the big medical tool in the treatment of mental disorders such as depression; and finally a complete review on the Daphniphyllum alkaloids.
