Chapters in that series contained physical-genetic maps but very little DNA sequence information. Now the complete DNA sequence is known for some phages, and the se quences for others will soon follow. During the past 10 years two phages have come into common use as reagents: A phage for cloning single copies of genes, and Ml3 for cloning and DNA sequencing by the dideoxy termi nation method. Also during that period the use of alternative sigma fac tors by RNA polymerase has become established for SPOl and T4. This seems to be a widely used mechanism in bacteria, since it has been implicated in sporulation, heat shock response, and regulation of nitro gen metabolism. The control of transcription by the binding of A phage CII protein to the -35 region of the promoter is a recent finding, and it is not known how widespread this mechanism may be. This rapid progress made me eager to solicit a new series of reviews. These contributions are of two types. Each of the first type deals with an issue that is exemplified by many kinds of phages; chapters of this type should be useful in teaching advanced courses. Chapters of the second type provide comprehensive pictures of individual phage families and should provide valuable information for use in planning experiments.

Book excerpt: To investigate the mechanism of transcription termination by bacteriophage RNA polymerases, templates encoding variants of the transcription-termination structure in the regulatory region of the threonine (thr) operon of Escherichia coli were used. The thr terminator comprises two distinct structural elements: a G+C-rich inverted repeat, which encodes an RNA hairpin structure, and A+T-rich regions, one of which contains a continuous sequence of template deoxyadenosine residues within which the transcript terminates. Fourteen terminator variants were analyzed by T7 RNA polymerase and we find that not only the hairpin structure itself but also its sequence influences termination. Furthermore, the formation of a hairpin in the RNA encoded by the A+T-rich regions of the attenuator is not mandatory for termination. Similar studies were also applied to T3 and Sp6 RNA polymerases. T3 RNA polymerase has very similar termination properties as T7 RNA polymerase does with both wild-type and mutated thr terminators. Sp6 RNA polymerase shows 63% termination efficiency compared to the 43% termination efficiency of T7 RNA polymerase on thr terminator, and the thr variants have less effect on Sp6 RNA polymerase than on T7 RNA polymerase. A series of seven deletion variants that successively shorten the deoxyadenosine tract in the terminator template were also analyzed. Results from these experiments with T7 and Sp6 RNA polymerases indicate that complete readthrough occurs when there are four or fewer deoxyadenosine residues. With 5 template deoxyadenosine residues there is 5 percent termination increasing to 32 percent with 8 deoxyadenosines--the value produced by the wild-type terminator. In addition, a comparison with E. coli RNA polymerase shows that bacteriophage RNA polymerases require a more perfect region of dyad symmetry and a longer deoxyadenosine tract than does the bacterial enzyme to terminate with maximum efficiency.

Book excerpt: The motivation for us to produce a treatise on regulation was mainly our convic tion that it would be fun, and at the same time productive, to approach the subject in a way that differs from that of other treatises. We had ourselves written reviews for various volumes over the years, most of them bringing together all possible facts relevant to a particular operon, virus, or biosynthetic system. And we were not convinced of the value of such reviews for anyone but the expert in the field reviewed. We thought it might be more interesting and more instructive-for both author and reader-to avoid reviewing topics that anyone scientist might work on, but instead to review the various parts of what many different scientists work on. Cutting across the traditional boundaries that have separated the subjects in past volumes on regulation is not an easy thing to do-not because it is difficult to think of what interesting topics should replace the old ones, but because it is difficult to find authors who possess sufficient breadth of knowledge and who are willing to write about areas outside those pursued in their own laboratories. For example, no one scientist works on suppression per se. He may study the structure of suppressor tRNAs in Escherichia coli, he may study phenotypic suppression of various characters in drosophila, he may study polarity in gene expression, and so on.
