Available in PDF, EPUB and Kindle. Book excerpt: The major focus of this thesis involves a new approach to spin-crossover (SCO) in iron(II) dinuclear complexes. In order to acquire SCO properties, a series of novel ligands (L1M, L2M, L3M, L4M), together with the corresponding iron(II) and/or nickel(II) complexes were synthesized and characterized. Beyond the successful synthesis and characterization of these ligands, some interesting aspects of their synthesis are discussed. The reported ligands are methyl-derivatives from ligands previously studied in the group. The methyl group in the position 6 of the pyridine group is introduced to induce steric hindrance in the complexes and decrease the ligand field strength. As a result, the influence of the methyl group may induce SCO in low spin (LS) complexes, or tune the SCO properties (such as transition hysteresis or transition temperature) in complexes with SCO. Ligands were divided into three different types according to their spacers: a ligand with an aromatic spacer (L1M), ligands with aliphatic spacer (L2M, L3M) and a ligand with no spacer (L4M). The difference between their structures helped us to better understand the coordination chemistry of the corresponding complexes. The nickel(II) complexes formed provided useful background information on the coordination chemistry of iron(II), such as the crystal field stabilization energy (10Dq) value. An approximation of 10Dq value of iron(II) was calculated based on the 10Dq value of analogous nickel(II) complexes using an empirical rule, to provide a prediction of SCO property.

Book excerpt: Herein is described the syntheses and characterization of iron dicarbonyl complexes derived from tridentate ortho-substituted Schiff base pyridine/thioether ligands ([subscript R]NNS) and (Fe(CO)4(Br)2). Two general synthetic routes are reported for the isolation of such iron carbonyls. First, metalation of [subscript R]NNS ligands within Et2O at room temperature affords the complex species of type (([subscript R]NNS)Fe(CO)2Br)(Fe(CO)3(Br)3). Alternatively, metalations of [subscript R]NNS (R = CH3, OCH3) at low temperature (-78 °C) in CH2Cl2/MeCN mixtures with (Fe(CO)4(Br)2) afford complexes of formula ([subscript R]NNS)Fe(CO)2Br)Br. Reactions under similar conditions with more sterically demanding ligands (R = quinoline (Q), chlorophenyl (ClPh)) afford a complex salt of the form (([subscript R]NNS)Fe(CO)2Br)(Fe(CO)3(Br)3). Complexes have been characterized using X-ray crystallography, IR spectroscopy and low temperature 1H NMR spectroscopy. Photostability of the complexes has been studied using UV-vis and solution IR spectroscopies. Relevance to the apo-active site of mono-iron hydrogenase is discussed.

Book excerpt: Lena Daumanns's thesis describes structural and functional studies of the enzyme Glycerophosphodiesterase (GpdQ) from Enterobacter aerogenes. It also examines the properties of small mimics of this enzyme and related binuclear metallohydrolases such as the metallo-ß-lactamases to enhance our understanding of hydrolytic cleavage of important substrates like phosphoesters and β-lactams. Overall, this project has led to a better understanding of the metal ion binding and active site structural features of the enzyme GpdQ. Daumann describes how she successfully immobilized phosphoesterase and related biomimetics on solid supports for potential applications in the area of bioremediation of organophosphate pesticides. Analysis shows that both the enzyme and biomimetics can be stored on the solid support without loss of activity. Furthermore, the author specroscopically and mechanistically characterized a number of Zn(II), Cd(II) and Co(II) complexes, some of which are among the most active biomimetics towards organophosphates reported to date. This thesis makes excellent reading for non-specialists because each chapter includes a short introduction section.

This book is intended to reminisce this event, to take inventory, and to point out new lines of development in this field. Divided in 6 sections and 22 chapters, the book provides an up-to-date account on topics such as - the chemistry and biochemistry of cisplatin, - the clinical status of Pt anticancer drugs, - the impact of cisplatin on inorganic and coordination chemistry, - new developments in drug design, testing and delivery. It also includes a chapter describing the historical development of the discovery of cisplatin. The ultimate question - How does cisplatin kill a cell? - is yet to be answered, but there are now new links suggesting how Pt binding to DNA may trigger a cascade of cellular reactions that eventually result in apoptosis. p53 and a series of damage recognition proteins of the HMG-domain family appear to be involved. The book addresses the problem of mutagenicity of Pt drugs and raises the question of the possible relevance of the minor DNA adducts, e.g. of interstrand cross-links, and the possible use of trans-(NH3)2Pt(II)-modified oligonucleotides in antisense and antigene strategies. Our present understanding of reactions of cisplatin with DNA is based upon numerous model studies (from isolated model nucleobases to short DNA fragments) and application of a large body of spectroscopic and other physico-chemical techniques. Thanks to these efforts there is presently no other metal ion whose reactions with nucleic acids are better understood than Pt. In a series of chapters, basic studies on the interactions of Pt electrophiles with nucleobases, oligonucleotides, DNA, amino acids, peptides and proteins are reported, which use, among others, sophisticated NMR techniques or X-ray crystallography, to get remarkable understanding of details on such reactions. Reactivity of cisplatin, once bound to DNA and formerly believed to be inert enough to stay, is an emerging phenomenon. It has (not yet) widely been studied but is potentially extremely important. Medicinal bioinorganic chemistry - the role of metal compounds in medicine - has received an enormous boost from cisplatin, and so has bioinorganic chemistry as a whole. There is hardly a better example than cisplatin to demonstrate what bioinorganic chemistry is all about: The marriage between classic inorganic (coordination) chemistry and the other life sciences - medicine, pharmacy, biology, biochemistry. Cisplatin has left its mark also on areas that are generally considered largely inorganic. The subject of mixed-valance Pt compounds is an example: From the sleeping beauty it made its way to the headlines of scientific journals, thanks to a class of novel Pt antitumor agents, the so-called "platinum pyrimidine blues". In the aftermath diplatinum (III) compounds were recognized and studies in large numbers, and now an organometalic chemistry of these diplatinum (III) species is beginning to emerge. The final section of the book is concerned with new developments such as novel di- and trinuclear Pt(II) drugs with DNA binding properties different from those of cisplatin, with orally active Pt(IV) drugs which are presently in clinical studies, and with attempts to modify combinatorial chemistry in such a way that it may become applicable to fast screening of Pt antitumor drugs. The potential of including computational methods in solving questions of Pt-DNA interactions is critically dealt with in the concluding chapter.

Book excerpt: Given the prominence of six-coordinate pseudo-octahedral complexes in transition metal chemistry, lower coordinate complexes in tetrahedral, trigonal pyramidal and trigonal bipyramidal configurations have garnered increased interest due to the changes in electronic structure and reactivity resultant from three-fold symmetry relative to the four-fold symmetry commonly observed in their pseudo-octahedral counterparts. Herein, we report a range of such effects in a collection of transition metal complexes supported by chelating, trianionic trispyrrolide ligands [tpaR]3-. In chapter one, two trigonal pyramidal iron(II) complexes, [(tpaPh)Fe]- and [(tpaMes)Fe]-, are discussed which are capable of undergoing oxygen atom transfer reactions and subsequent activation of strong C-H bonds. Additionally, the mesityl derivative is capable of oxygen atom transfer from nitrous oxide, an attractive but notoriously unreactive molecule. In chapter two, this family of iron(II) complexes is expanded to include tert-butyl, 2,4,6-triisopropylphenyl and 2,6-difluorophenyl substituted variants, and the magnetic properties of this homologous series of trigonal pyramidal iron(II) complexes are explored. Notably, a number of these complexes exhibit frequency dependent signals at low temperature in the imaginary component of ac magnetic susceptibility measurements performed in the presence of a small applied field. Further exploration of this behavior via magnetometry, high-field EPR and Mössbauer spectroscopy reveal a large barrier to magnetic relaxation in these complexes, in some cases comparable to the best known single molecule magnets. This phenomenon is accounted for by the unquenched orbital angular momentum present in a three-fold symmetric high spin d6 system, leading to significant uniaxial anisotropy and a barrier to spin inversion. Progress towards a magneto-structural analysis of this behavior is made for this series. Chapter three contains a through treatment of the coordination, redox and group transfer chemistry of the iron(II) platform [(tpaMes)Fe]-. In particular, it is shown that this complex is capable of supporting both high and low spin five coordinate adducts, including a rare example of a paramagnetic carbonyl complex of iron(II), [(tpaMes)Fe(CO)]-. Furthermore, the redox chemistry of this system is illustrated in the synthesis of the neutral iron(III) complex (tpaMes)Fe(i-PrNH2). As an extensiont to the oxygen atom chemistry presented in chapter one, nitrene group transfer to [(tpaMes)Fe]- is described, resulting in the synthesis of an iron(III) amido complex. The identity of this species is confirmed by high resolution electrospray mass spectrometry as well as Mössbauer, x-ray absorption and EPR spectroscopies. In chapter four, the vanadium chemistry of [tpaR]3- is introduced. In particular, the vanadium(III) complex (tpaMes)V(THF) is show to react with an aryl azide to yield an isolable vanadium(V) diazenylimido species, a member of a very rare structural type. This complex is shown to decompose unimolecularly by dinitrogen loss to generate the cooresponding vanadium(V) imido. In addition to the vanadium-nitrogen multiple bond chemistry described, the synthesis two pseudo-three-fold symmetric vanadium(IV) oxo species is discussed. Single crystal x-ray structures of both complexes reveal a distorted coordination geometry and a short vanadium-oxo distance, consistent with a triple bond. The oxidation of both of these complexes to the corresponding vanadium(V) oxo is performed and shown by x-ray crystallography to result in the relief of the distortion observed in the vanadium(IV) oxo complexes. Taken together, these results shed light on the nature of metal ligand multiple bonding in three-fold symmetric geometries. Chapter five discusses the chromium complexes of [tpaMes]3-, in particular the group transfer and redox chemistry of the system. Both chromium(II) and chromium(III) complexes are available by direct metallation, and the chromium(II) complex [(tpaMes)Cr(DME)]- is shown to undergo both oxygen atom and nitrene group transfer reactions to yield chromium(IV) oxo and imido complexes respectively. The chromium(IV) imido complex exhibits rich redox chemistry, and its oxidation to the isostructural chromium(V) and chromium(VI) complexes are demonstrated.
