Book excerpt: Nucleophilic ring opening of oxabenzonorbornadiene (OBD) has been used in natural product synthesis as it enables precise creation of multiple stereocenters in a single step. The work presented in this thesis investigates how modifications to the OBD framework influences the regio- and stereochemistry of the products. The scope of the nickel catalyzed arylation of OBD was broadened by including C1-substituted substrates. Different 1,2-dihydronaphthol or naphthalene regioisomers could be obtained by varying the substituents' electronic properties. The latter chapters focus on the ring opening chemistry of OBD derivatives. Intramolecular ring opening of OBD was explored through the synthesis of OBDs with C1-tethered nucleophiles. Subsequent ring opening of these compounds yielded tricyclic and tetracyclic products. Finally, the scope of the palladium catalyzed Type 2 ring opening and the acid catalyzed Type 3 ring opening of the OBD derivative, cyclopropanated oxabenzonorbornadiene (CPOBD), was expanded to include substituted CPOBD substrates.

Book excerpt: This thesis presents a general approach to accessing nitrogen-substituted hetero- and carbocycles. In short, the annulation reactions developed in the thesis make it possible to access nitrogen-substituted four-, five- and six-membered rings, all essential building blocks for the synthesis of bioactive molecules. Many natural products display a saturated polycyclic core allowing a well-defined arrangement of functional groups in space. As such, they can interact with biological targets with a high degree of affinity and selectivity, surpassing many synthetic drugs. Nevertheless, the efficient synthesis of such complex ring systems poses a challenge for organic chemistry. Through careful tuning of the electronic properties of a nitrogen donor group and a diester acceptor group, the first [3+2] annulation reaction between aminocyclopropanes and enol ethers or carbonyl compounds is now possible. The reaction proceeded under mild catalytic conditions, and the building blocks obtained can be found at the core of bioactive alkaloids, drugs such as Ramipril and biomolecules such as DNA and RNA. Thanks to the dynamic kinetic asymmetric annulation of aminocyclopropanes with enol ethers and aldehydes, access to enantioenriched compounds is also now possible. Lastly, a synthesis of donor-acceptor aminocyclobutanes via [2+2] cycloaddition using a cheap iron catalyst was developed, allowing them to be used in [4+2] annulations to access cyclohexylamines.

Book excerpt: The present work demonstrates the first examples of carboxylic acid-tethered cyclopropanated oxabenzonorbornadienes (CPOBDs) to undergo ring-opening reactions in mild acidic conditions. Two regioisomers are formed but the reactions are highly regioselective towards type 3 ring-opened products. It was observed that substitution at the C-5 and aryl positions of CPOBD significantly hinders the ring-opening reactions leading to poor yields of ring-opened product, although high regioselectivity for the type 3 ring-opened product is still maintained. The second part of this thesis outlines a study on the regio- and stereo-chemistry of the iridium-catalyzed hydroacylation of C1-substituted OBDs with salicylaldehyde. The scope of the reaction was expanded to include various electron-donating, electron-withdrawing and bulky substituents to investigate the electronic and steric effects of the C1- substituent on the hydroacylation reaction. It was observed that the addition occurred exclusively at the less hindered, C3 position to give the corresponding adducts.

Book excerpt: 2-oxazolines are extremely versatile substrates in organic synthesis and their synthesis, chemistry and uses have been well documented. The aim of this study is to investigate the synthesis and transformations of 5-benzeneselenenyl and -sulphenyl-substituted 2-oxazolines. Chapter one reviews various aspects of the synthesis and chemistry of 2-oxazolines in general. Reaction of N-allylic amides with selenenyl and sulphenyl halides and the subsequent Lewis-Acid catalysed cyclisation into 2-oxazolines is shown to be an effective and generally high-yielding route for the preparation of a variety of oxazolines differing in their substitution pattern. Structural proof for the oxazoline moiety was obtained in some cases by deselenation experiments and in general by spectroscopic methods. A competing oxazoline-oxazine forming process was observed and the effect of the sulphur and selenium electrophiles and Lewis Acid noted. Cyclisation of a cyclohexene derived allylic amide also produced a novel bicyclic, oxazine product. The synthetic utility of 5-benzeneselenenyl and sulpheny substituted oxazolines is demonstrated through the oxidation, reduction and rhydrolysis reactions of these systems in particular. Synthesis of various sulpoxide/sulphone systems, b-aminoalcohols b-hydroxyamides was achieved from the 2-oxazolines prepared in this study. Selective oxidation of the b-hydroxyamides at sulphur or at the alcohol group was also studied. The introduction of further functionality a-acetoxysulphide derived oxazoline was achieved, and this substrate was successfully subjected to a substitution reaction with allyltrimethylisilane in the presence of SnC14. Various novel reactions of the sulphone and sulphoxide derivatives: reductive ring opening with LiA1H4, reaction with n-butyllithium and a sequential Pummerer-phenylthio group migration oxazoline ring opening process, were also observed.

Book excerpt: Oxabenzonorbornadiene (OBD) and derivatives thereof have become key substrates of interest within synthetic organic chemistry and natural product synthesis as they enable the formation of multiple stereocenters in a single transformation. The present work expands on previous studies to investigate various intramolecular transformations, and regiochemical implications regarding ring-opening reactions of asymmetric oxabicyclic alkenes. Acid-catalyzed intramolecular ring-opening reactions of cyclopropanated oxabenzonorbornadiene (CPOBD) derivatives were investigated to afford novel medium-sized ring systems in moderate to good yields and good to excellent regioselectivity. Similarly, a Ni catalyzed intramolecular ring-opening reaction of alkyl halide tethered OBD compounds was optimized, thus generating a naphthalene derivative in excellent yields. Lastly, iridium-catalyzed ring-opening reactions of unsymmetrical C1-substituted OBD derivatives is presented using alcohols and water as nucleophiles which allows for the selective synthesis of previously inaccessible substrates.
