Book excerpt: This work focuses on the synthesis, the characterization and the catalytic evaluation in the reduction of protons into dihydrogen, of new complexes of Ni(II), Co(III), Cu(II) and Pd(II) based ligands Type thiosemicarbazone. The catalytically active species during the process of the proton reduction was studied by cyclic voltammetry and mechanisms were formulated on the basis quantum chemical calculation.The first chapter introduces the scientific context, the goals and the main objectives of this work. The second chapter concerns the synthesis and the characterization of the N2S2 ligands and their associated mononuclear complexes, Ni, Cu and Pd. The third chapter presents the synthesis and the characterization of binuclear Co and trinuclear Ni based on N2S2 ligand.Electrochemical studies of these complexes in DMF in the presence of a proton source (trifluoroacetic acid), allowed us to evaluate their catalytic efficiency. Our results show that Cu and Pd complexes have a specific irreversible wave for the reduction of protons, but decomposition is observed during electrolysis, which makes these uninteresting complexes for the reduction of protons.On the contrary, Ni and Co complexes showed an electrochemical stability and good catalytic performances. In particular, the new mononuclear Ni complex exhibits remarkable catalytic properties that rank it among the best catalysts for the reduction of protons reported in the literature. All this work provided a complete description of the electrochemical behavior of N2S2 thiosemicarbazone ligands complexed to transition metals. It allows considering future developments in improving the catalytic properties of these complexes.

Book excerpt: This research presents an investigation of synthetic model complexes with relevance to the active site of Ni(II) acireductone dioxygenase (Ni-ARD) and Ni(II) superoxide dismutase (Ni-SOD). Acireductone dioxygenases (ARDs) are a unique set of enzymes found in the methionine salvage pathway that catalyze the oxidation reaction of acireductone (1, 2-dihydroxy-3-oxo-5-(methylthio)pent-1-ene). These enzymes share a common polypeptide sequence but bind different metal ions, Ni 2+ or Fe2+, at the active site. The Ni-ARD enzyme is responsible for the off pathway shunt in the pathway. Using the tridentate nitrogen donor ligands hydrotris(3,5-dimethyl-1-pyrazolyl)borate (Tp*) and the newly developed tris(1, 2-dimethyl-4-imadozyl)carbinol, (4-TIC Me, Me) several reactions involving the acireductone analog 2-hydroxy-1, 3-diphenylpropan-1, 3-dione and O2 were investigated for similarities to the Ni-ARD active site. Superoxide dismutases (SODs) play a key role in protecting cells against oxidative damage by regulating the cellular concentration of the superoxide radical (O2.- ) which is an unwanted byproduct of cellular metabolism. This process is accomplished by converting the superoxide radicals to hydrogen peroxide and molecular oxygen. Several small-molecule complexes were synthesized and characterized in an effort to model the reduced state of the Ni-SOD using the Tp* ligand. The structures for these complexes have been determined using X-Ray Crystallography.

Book excerpt: Transition metal complexes of nickel(II) and zinc(II) with 5-bromo- N -(8-quinolyl)salicylaldimine (HqsalBr, HL 1 ) and 3,5-dibromo- N -(8-quinolyl)salicylaldimine (HqsalBr 2 , HL 2 ) have been synthesized and successfully characterized using several techniques: IR, 1 H-NMR, mass spectrometry and thermogravimetric analysis (TGA). The molecular structures of nickel(II) and zinc(II) complexes with HL 2 were determined by single crystal X-Ray crystallography. Ligands HL 1 and HL 2 both act as a tridentate ligand and coordinate to metal ion center via deprotonated hydroxyl oxygen atom, nitrogen of azomethine functional group and nitrogen of quinoline substituent. The interaction of these complexes with DNA was investigated using calf thymus DNA (CT-DNA) in which the studies by electronic absorption and luminescence titrations reveal that DNA binding mode of the complexes is intercalation. All the synthesized complexes provide great values of binding constant (K b ) and these results are consistent with the quenching capability defined by quenching constant values (K q ). Amongst the synthesized complexes, zinc(II) with ligand HL 2 complex, [Zn(qsalBr) 2 ], exhibits the most superior ability in the interaction with CT-DNA and the verification mode of binding by the competitive reaction with ethidium bromide (EB) also indicates the strongest intercalation by such complex. Additionally, ligands and complexes were also examined in anti-lung cancer activity against A549 cell line and compared with standard compounds, Etoposide and Cisplatin.

Book excerpt: Superoxide dismutases are ubiquitous enzymes that efficiently catalyze the disproportionation of superoxide radical anions to protect biological molecules from oxidative damage. Several SODs have been identified having different metals at their active sites. These include Mn SOD, Fe SOD, Cu/Zn SOD and, most recently, Ni SOD. The catalytic center of Ni SOD resides in the N-terminal active-site loop, where a Ni(II) is coordinated by the amine N of His-1, the amide N of Cys-2, and two thiolate S atoms of Cys-2 and Cys-6. In the oxidized form, Ni(III) adds the imidazole N of His-1 as an axial ligand. For the past decade, we have been developing methodology using 2, 2'-dithiodibenzaldehyde (DTDB) for the synthesis of metal complexes with mixed N/S coordination. We are reporting on the application of this methodology to the synthesis of model complexes for the active site of NiSOD, in which we have successfully synthesized and characterized three NiIIN2S2 complexes of imine/amine N donors: Ni(NNS)SPh (1), Ni(NNS)SPhNO2 (2) and Ni(NNS)StBu (3). These may be used as a model for reduced NiSOD, with future plans of comparing to complexes with amide/amine N donors, thus establishing the role of the amide.
