Available in PDF, EPUB and Kindle. Book excerpt: Subtilisin is the most extensively studied model system for protein engineering. The primary motivating factor for the interest in subtilisin is the commercial utility of this class of proteases. The subtilisin symposium was the first international meeting to bring together a large number of groups that have focused on the subtilisins and the subtilases-the protein superfamily of subtilisin-like enzymes. The results presented at the symposium are in this way a unique compendium of a broad spectrum of work largely focused on harnessing the potential of site-directed mutagenesis to understand and deliberately alter the function of these enzymes toward a desired end. This sort of protein engineering has been extremely successful in subtilisin, with many such "engineered" enzymes now widely used in commer cial enterprises. In this regard the experience derived from subtilisin does represent practical protein engineering. It is becoming clear that subtilisin represents a larger class of enzymes, the subtilases, that include many of the human pro hormone-converting enzymes. As international collabo rative efforts to sequence entire genomes continue, we can reasonably expect that additional members of the subtilase class will be encountered. Whenever interest in a member of this class of enzyme arises, the work on subtilisin will serve as a guide to the analysis for what in bacillus, fungi, and industry is an everyday workhorse enzyme.

Book Handbook of Proteolytic Enzymes Volume 1

Download or read book Handbook of Proteolytic Enzymes Volume 1 written by Alan J. Barrett and published by Elsevier. This book was released on 2012-12-02 with total page 1182 pages. Available in PDF, EPUB and Kindle. Book excerpt: Handbook of Proteolytic Enzymes, Second Edition, Volume 1: Aspartic and Metallo Peptidases is a compilation of numerous progressive research studies on proteolytic enzymes. This edition is organized into two main sections encompassing 328 chapters. This handbook is organized around a system for the classification of peptidases, which is a hierarchical one built on the concepts of catalytic type, clan, family and peptidase. The concept of catalytic type of a peptidase depends upon the chemical nature of the groups responsible for catalysis. The recognized catalytic types are aspartic, cysteine, metallo, serine, threonine, and the unclassified enzymes, while clans and families are groups of homologous peptidases. Homology at the level of a family of peptidases is shown by statistically significant relationship in amino acid sequence to a representative member called the type example, or to another member of the family that has already been shown to be related to the type example. Each chapter discusses the history, activity, specificity, structural chemistry, preparation, and biological aspects of the enzyme. This book will prove useful to enzyme chemists and researchers.

Book Directed Enzyme Evolution

    Book Details:
  • Author : Frances H. Arnold
  • Publisher : Springer Science & Business Media
  • Release : 2008-02-02
  • ISBN : 1592593968
  • Pages : 381 pages

Download or read book Directed Enzyme Evolution written by Frances H. Arnold and published by Springer Science & Business Media. This book was released on 2008-02-02 with total page 381 pages. Available in PDF, EPUB and Kindle. Book excerpt: Directed evolution comprises two distinct steps that are typically applied in an iterative fashion: (1) generating molecular diversity and (2) finding among the ensemble of mutant sequences those proteins that perform the desired fu- tion according to the specified criteria. In many ways, the second step is the most challenging. No matter how cleverly designed or diverse the starting library, without an effective screening strategy the ability to isolate useful clones is severely diminished. The best screens are (1) high throughput, to increase the likelihood that useful clones will be found; (2) sufficiently sen- tive (i. e. , good signal to noise) to allow the isolation of lower activity clones early in evolution; (3) sufficiently reproducible to allow one to find small improvements; (4) robust, which means that the signal afforded by active clones is not dependent on difficult-to-control environmental variables; and, most importantly, (5) sensitive to the desired function. Regarding this last point, almost anyone who has attempted a directed evolution experiment has learned firsthand the truth of the dictum “you get what you screen for. ” The protocols in Directed Enzyme Evolution describe a series of detailed p- cedures of proven utility for directed evolution purposes. The volume begins with several selection strategies for enzyme evolution and continues with assay methods that can be used to screen enzyme libraries. Genetic selections offer the advantage that functional proteins can be isolated from very large libraries s- ply by growing a population of cells under selective conditions.

Book Foldases

    Book Details:
  • Author : Natalya K. Nagradova
  • Publisher : Nova Publishers
  • Release : 2008
  • ISBN : 9781604563894
  • Pages : 254 pages

Download or read book Foldases written by Natalya K. Nagradova and published by Nova Publishers. This book was released on 2008 with total page 254 pages. Available in PDF, EPUB and Kindle. Book excerpt: A great deal of talent has been called upon over the past decade to try and understand the structure and the function of different types of foldases; as a result, a remarkable insight into how these enzymes work has been gained. Recently the main focus of research has begun to shift to unveiling the precise role of these folding catalysts in many different processes in which they are involved inside the cell. It is becoming increasingly clear that the cellular functions of foldases go far beyond accelerating the folding of proteins. This is undoubtedly an area where one can expect significant research efforts in the next few years. The present book summarises the current state of knowledge as to the structural and functional properties of four major types of foldases listed above. In structuring the material the author has sought to maintain a balance between historical development and current understanding, while also trying to look to the future and anticipate where the field is likely to move.

Book Enzymes in Action Green Solutions for Chemical Problems

Download or read book Enzymes in Action Green Solutions for Chemical Problems written by B. Zwanenburg and published by Springer Science & Business Media. This book was released on 2000 with total page 476 pages. Available in PDF, EPUB and Kindle. Book excerpt: Enzymes in Action is a timely survey of a modern development in organic chemistry. It is clear that bioreagents demand that organic chemists think in a different way. If they do so, they will open up new avenues of exciting, new chemistry that will permit problems to be solved in an elegant way. The first section covers the concepts necessary to understand enzymes in molecular operations. The second section covers heteroatom enzyme chemistry, with considerable attention being given to the use of enzymes in the detoxification of chemical warfare agents and their application in environmental problems. The final section highlights the strategic use of enzymes in organic chemistry. It is clear that the term 'green chemistry' is appropriate, since enzyme mediated processes occur under mild, environmentally benign conditions, and enzymes enable chemists to perform new chemical operations that would otherwise be difficult to achieve at all.
