Book excerpt: This report summarizes recent progress in the development and improvement of tunable laser systems for novel approaches to atomic and molecular spectroscopy, and in particular for new precision studies of atomic hydrogen. It reports on: (a) The development of a highly monochromatic, powerful tunable dye laser system, combining a single-frequency cw dye laser oscillator with a multistage pulsed dye laser amplifier. (b) The generation of cw single-frequency coherent radiation near 243 angstrom by summing the frequencies of a Kr ion laser and a cw dye laser in cooled ADP. In addition, the report summarizes three experiment, exploring new coherent light techniques for sensitive high resolution spectroscopy of atoms and molecules. These are (c) The first demonstration of Doppler-free laser polarization spectroscopy, a new technique, which can greatly exceed the sensitivity of saturated absorption spectroscopy. The Stark splitting of the hydrogen Balmer beta line in a glow discharge has been resolved. (d) A demonstration of polarization labeling of molecular sodium This new technique simplifies the unraveling of complex absorption spectra by identifying lines with a common lower state. (e) High resolution two-photon spectroscopy of the excited state of normal and deuterated benzene. The observed rotational contours have been reported by computer fits, yielding accurate molecular constants.

Development of the gallium-arsenic semiconductor laser (diode laser) that emits laser light at 780-830 nm has made possible development of new opto-electronic systems including laser optical recording systems, thermal writing display systems, laser printing systems, and so on. Medical applications of lasers in photodynamic therapy for the treatment of cancer were also developed. In such systems, the infrared absorbing dyes OR dyes) are currently used as effective photoreceivers for diode lasers, and will become the key materials in high technology. At the present time the chemistry of IR dyes is the most important and interesting field in dye chemistry. Laser light can be highly monochromatic, very well collimated, coher ent, and, in some cases, extremely powerful. These characteristics make diode lasers a very cheap, convenient, and useful light source for a variety of applications in science and technology. For these purposes, however, IR dyes with special characteristics are required. To develop new IR dyes, it is most important to establish the correlation between the chemical structures of dyes and other characteristics of dyes, such as their absorption spectra. Molecular design of IR dyes predicting the Amax and Emax values by molecular orbital (MO) calculations is now possible even by using a personal computer, and many types of new IR dyes have been demonstrated. Also, new opto-electronic systems using IR dyes as photoreceivers have been reported recently.

Today the major challenges to this topic include the elucidation of photochemical reactions involved in photobiology at the molecular level. This includes the use of a variety of modem probing techniques that directly measures the reactivity of excited states and free radicals involved in biological reactions. This text-book is based on such an approach and has arisen from some of the lectures delivered at the NATO ASI held at Hotel Capo Caccia near the Centre for Advanced Research in Photobiology (CARP) in Sardegia, Italy. The ASI took place from 30 September -13 October 1993 and involved a total membership of 90. The book, like the NATO ASI itself, is divided into four themes starting with fundamental aspects and ending with complex medically related systems. Thus Theme 1 covers aspects of the underlying photophysics and photochemistry with particular emphasis on modem experimental techniques to study molecular mechanisms of biological processes. Theme 2 applies many of these fundamental studies to the chemical reactions of most relevance to photobiology and photomedicine such as photo-addition, -isomerization, -sensitization and -pigmentation. The third and fourth Themes deal with the deleterious and therapeutic aspects of light with particular emphasis on the use of Photo-Dynamic Therapy (PDT) to treat cancer and on viral and micro bioi infections.
