Book excerpt: Steroid Hormone Regulation of the Brain focuses on the advancement of knowledge of the properties of brain steroid receptors and steroid target nerve cells, including the actions of gonadal steroids, adrenocortical steroids, and androgenic steroids on the brain. The selection features the work of authors who have inspiringly conducted extensive studies on the action of steroid hormones. The book starts with discussions that point out that steroid hormones not only function through intracellular steroid receptors, but possibly through steroid membrane actions as well. Studies to support this claim are presented. As to the action of steroid hormones on presynaptic transmitter mechanisms, the book suggests that adrenocortical and gonadal steroids can create alterations in noradrenaline, adrenaline, and dopamine levels and turnover within the hypothalamus and preoptic area. The effects of catecholestrogens on the brain are also elaborated. A contribution to this paper stresses that estrogens have antidopaminergic action in the extrapyramidal system. This idea is identified through a series of behavioral experiments and measurements of the levels of striatal acetylcholine. This selection is recommended to scientists interested in studying the effect of hormones on the brain.

Contributors explore how the steroid receptor complex modulates transcription of RNA and cover topics ranging from the sequential acquisition of responsiveness to estrogen in the rat uterus to the ontogeny of steroid receptors in the guinea pig. This book is comprised of 29 chapters and begins with a review of estrogen and estrogen effects in rat uterus and pituitary in culture. The embryonic chick Müllerian duct and several fetal guinea pig organs are then considered, followed by discussions on mammalian and chick liver, multi-hormonal control in rat liver; progesterone and decidualization; and rat mammary gland in culture. Glucocorticoids in developing pancreas, lung, and liver are examined, along with ecdysteroids in both locusts and Drosophila. Refractoriness is exemplified by gonadotropin action in the ovary. The remaining chapters deal with the role of steroid hormone receptors in brain development; the neural trigger for ovulation; aromatization and development of responsiveness of the brain to gonadal steroids; and neuroendocrine correlates of female-offspring interaction in maternal rats. Book excerpt: Intracellular Receptors: New Instruments for a Symphony of Signals In the late eighteenth century, it was proposed on theoretical grounds that each of the body's organs, beginning with the brain, must be "a factory and laboratory of a specific humor which it returns to the blood", and that these circulating signals "are indispensable for the life of the whole" (Bordeu 1775). During the nineteenth cen tury, some remarkable physiological experiments revealed the actions of humoral factors that affected the for and function of multiple tissues, organs and organ sys tems within the body (Berthold 1849); much later, the chemical and molecular na ture of some of those factors was determined. Against this deep historical backdrop of the founding studies of intercellular signaling, molecular biology sprang into existence a mere forty years ago, rooted in the revelation of regulable gene expression in bacteria. But contemporaneous with those classical analyses of transcriptional regulation of the lactose operon, the mod em era of signal transduction was inaugurated by the identification of cAMP as a second messenger -- an intracellular mediator of hormonal activation of glycogen catabolism (Sutherland and RaIl 1960). Later in that same decade, it emerged that cAMP is a critical signal not only in metazoans, but even in bacteria, where it serves an analogous function as a critical switch that activates expression of genes re quired for catabolism of complex carbon sources, including those of the lactose operon.

This book captures the detailed presentations made at the first conference on Steroid Receptors in Health and Disease held at Meadow Brook Hall, Oakland University in the fall of 1987.
