Book Studies on Divergent Crp Dependent Promoters

Download or read book Studies on Divergent Crp Dependent Promoters written by Mohamed Elrobh and published by LAP Lambert Academic Publishing. This book was released on 2010-07 with total page 276 pages. Available in PDF, EPUB and Kindle. Book excerpt: Bacterial genomes contain genes that are packed closely together. For many genes, promoter regions overlap, and regulatory segments that carry divergent promoters are common. In this study, I have focussed on promoters that are regulated by the cyclic AMP receptor protein (CRP) of Escherichia coli. CRP is a global regulator that directs the activity of more than 100 genes, some of which are oriented back-to-back, i.e., in a divergent fashion. CRP is known to activate transcription initiation through direct protein-protein contact with RNA polymerase and CRP-dependent promoters are classified into two classes, Class I and Class II, according to the location of the CRP-binding site. I have investigated whether a single bound CRP, either at a Class I or a Class II location, can activate divergent transcription.

It has implications for other biological problems, such as developmental biology and cancer, brings insights into genetic engineering and expression systems, and has consequences for important aspects of applied research. For example, the molecular basis of bacterial pathogenicity has implications for new antibiotics and in crop development. Prokaryotic Gene Expression is a major review of the subject, providing up-to-date coverage as well as numerous insights by the prestigious authors. Topics covered include operons; protein recognition of sequence specific DNA- and RNA-binding sites; promoters; sigma factors, and variant tRNA polymerases; repressors and activators; post-transcriptional control and attenuation; ribonuclease activity, mRNA stability, and translational repression; prokaryotic DNA topology, topoisomerases, and gene expression; regulatory networks, regulatory cascades and signal transduction; phosphotransfer reactions; switch systems, transcriptional and translational modulation, methylation, and recombination mechanisms; pathogenicity, toxin regulation and virulence determinants; sporulation and genetic regulation of antibiotic production; origins of regulatory molecules, selective pressures and evolution of prokaryotic regulatory mechanisms systems. Over 1100 references to the primary literature are cited. Prokaryotic Gene Expression is a comprehensive and authoritative review of current knowledge and research in the area. It is essential reading for postgraduates and researchers in the field. Advanced undergraduates in biochemistry, molecular biology, and microbiology will also find this book useful.

Available in PDF, EPUB and Kindle. Book excerpt: The birth and the development of molecular biology and, subsequently, of genetic engineering and biotechnology cannot be separated from the advancements in our knowledge of the genetics, biochemistry and physiology of bacteria and bacter- phages. Also most of the tools employed nowadays by biotechnologists are of bacterial (or bacteriophage) origin and the playground for most of the DNA manipulations still remains within bacteria. The relative simplicity of the bacterial cell, the short gene- tion times, the well defined and inexpensive culturing conditions which characterize bacteria and the auto-catalytic process whereby a wealth of in-depth information has been accumulated throughout the years have significantly contributed to generate a large number of knowledge-based, reliable and exploitable biological systems. The subtle relationships between phages and their hosts have produced a large amount of information and allowed the identification and characterization of a number of components which play essential roles in fundamental biological p- cesses such as DNA duplication, recombination, transcription and translation. For instance, to remain within the topic of this book, two important players in the or- nization of the nucleoid, FIS and IHF, have been discovered in this way. Indeed, it is difficult to find a single fundamental biological process whose structural and functional aspects are better known than in bacteria.

Book Bacterial Cell to Cell Communication

Download or read book Bacterial Cell to Cell Communication written by Donald R. Demuth and published by Cambridge University Press. This book was released on 2006-02-23 with total page 340 pages. Available in PDF, EPUB and Kindle. Book excerpt: Many bacterial diseases are caused by organisms growing together as communities or biofilms. These microorganisms have the capacity to coordinately regulate specific sets of genes by sensing and communicating amongst themselves utilizing a variety of signals. This book examines the mechanisms of quorum sensing and cell-to-cell communication in bacteria and the roles that these processes play in regulating virulence, bacterial interactions with host tissues, and microbial development. Recent studies suggest that microbial cell-to-cell communication plays an important role in the pathogenesis of a variety of disease processes. Furthermore, some bacterial signal molecules may possess immunomodulatory activity. Thus, understanding the mechanisms and outcomes of bacterial cell-to-cell communication has important implications for appreciating host-pathogen interactions and ultimately may provide new targets for antimicrobial therapies that block or interfere with these communication networks.

Book Bacterial Cellular Metabolic Systems

Download or read book Bacterial Cellular Metabolic Systems written by K. Shimizu and published by Elsevier. This book was released on 2013-03-26 with total page 497 pages. Available in PDF, EPUB and Kindle. Book excerpt: The metabolic regulation of a cell system is of critical importance in systems biology, and a robust model of these mechanisms is essential in predicting the effects on the metabolism of both the culture environment and the knockout of specific genes. Bacterial cellular metabolic systems focuses on this highly topical subject in relation to culture environment and provides a detailed analysis from gene level to metabolic level regulation, as well as offering a discussion of the most recent modelling approaches. The book begins with an introduction to metabolic mechanisms and to the metabolic regulation of a cell, before moving on to discussing the action of global regulators in response to a specific culture environment. The second half of the book examines conventional flux balance analysis and its applications, 13C-metabolic flux analysis, and the effect of a specific gene knockout on the metabolism. - Comprehensive account of metabolic regulation via global regulators in response to changes in the culture environment - Basic formulation of 13C-metabolic flux analysis based on 13C-labelling experiments - Systems biology approach for the modelling and computer simulation of the main metabolic pathways of a cell system
