Homopolymer peptides, which are made up of only a single type of amino acid, are far less ubiquitous. The only two amino-acid homopolymers known to occur in nature are presented in this volume. Poly-epsilon-L-lysine is a polycationic peptide and exhibits antimicrobial activity against a wide spectrum of microorganisms. It is both safe and biodegradable and is therefore used as a food preservative in several countries. In addition, there has been great interest in medical and other applications of poly-lysine and its derivatives. In contrast, poly-gamma-glutamic acid is an unusual anionic polypeptide. It is also water soluble, biodegradable, edible, non-toxic and non-immunogenic and can be chemically modified to introduce various drugs. These features are very useful for pharmaceutical and biomedical applications. Poly-glutamic acid is also a highly attractive as a food ingredient.

Book excerpt: Cells maintain fidelity in protein synthesis through various mechanisms including aminoacyl-tRNA synthetases (aaRSs) that are able to discriminate tRNAs and their cognate amino acids. Substrate specificity along with editing functions of aaRSs act to prevent or remove incorrect amino acids charged onto tRNAs. Editing domains are highly conserved and found in both prokaryotes and eukaryotes; knocking out the editing function of an aaRS does not seem to affect cell viability. To determine the biological relevance of the editing domains found on aaRSs, it is essential to determine the contribution of editing to protein fidelity in vivo. Bacillus subtilis serves as an ideal model to study this phenomenon because it possesses a small gene-enocoded, 10 amino acid pheromone peptide, ComX. ComX is processed, modified, and secreted by B. subtilis as a signaling molecule that can easily be purified. The amino acid sequence of ComX can be altered to some degree while maintaining cellular activity enabling different strains that produce variants of ComX pheromone to be compared for levels of mistranslation. Ultimately future experiments will reveal the impact that editing by an aaRS has on the fidelity of protein synthesis in vivo by quantifying the rates of mistranslation in strains of B. subtilis that express an editing-defective tRNA synthetase and comparing it to levels of mistranslation to wild type strains.

Book excerpt: In its natural environment, the soil bacterium Bacillus subtilis permanently encounters nutrient limitations and increases in osmotic stress. The present study investigated the systemswide response of B. subtilis to different simultaneously imposed stresses by combining chemostat experiments under conditions of carbon and nitrogen limitation, ionic and nonionic osmotic stress and osmoprotection with multi-omics analyses of the transcriptome, proteome, metabolome and fluxome. Surprisingly, the flux through central carbon and energy metabolism is very robust under all conditions studied. The key to achieve this robustness is the increased production of several enzymes of central carbon metabolism to compensate for their reduced activity in the presence of high salt. A major response of the cell during osmotic stress is the production of the compatible solute proline through the concerted adjustment of multiple reactions around the 2-oxoglutarate node, which drives metabolism towards the precursor glutamate. Co-regulations between the individual cellular components under the investigated stress conditions indicate that the fine-tuning of the transcriptional and metabolic networks involves functional modules that outreach single pathways. Additionally, the work shortly describes the salinity-decoupled fed-batch production of the compatible solute ectoine in Corynebacterium glutamicum and the contribution of precursor amino acids of the aspartate family.

Book excerpt: The threonyl-tRNA synthetase is an enzyme that catalyzes the attachment of the amino acid threonine to the threonyl-tRNA so that protein synthesis at the ribosome can occur. In order to maintain translational fidelity, this tRNA synthetase must employ an editing function that is structurally and functionally separate from this enzyme's aminoacylation capacity. Here, we studied the importance editing function of the Bacillus subtilis threonyl synthetase, ThrRS, by making a single amino acid substitution in the separate editing domain. The data in this study suggest that disrupting editing function in the major B. subtilis ThrRS, thrS, is toxic to the cells under normal growth conditions. A future goal in this project is to study the effect of knocking out the editing function of the minor B. subtilis ThrRS, thrZ.
