The field of protein phosphorylation, including the enzymes involved in this post-translational modification, continues to advance at a fascinating pace. Since the first international meeting on this topic, held in Heidelberg in 1994, several new avenues of CK2 research have emerged despite persistent deficiencies in our understanding of the regulation of its activity. Among the significant new directions are studies related to the structure of the enzyme, especially its crystal structure, as well as an interesting aspect of CK2 function that involves its subunits as binding partners of several other proteins. In addition, new data have been gathered on the role of CK2 in transcription as well as in certain other cellular functions. To address these various aspects of the progress of CK2, a number of key scientists from different parts of the world came together at the second international meeting on `A Molecular and Cellular View of Protein Kinase CK2', held at Villard de Lans near Grenoble on September 24-26, 1997. Available in PDF, EPUB and Kindle. Book excerpt: Protein kinase CK2 is a serine/threonine kinase with a multitude of substrates and roles in many cellular processes, including mitosis. CK2 is constitutively active, yet we hypothesize that CK2 is indeed regulated in mitosis through subtle means, enabling CK2 to perform its functions unique to cell division. Our aims were to examine the roles of mitotic phosphorylation, subcellular localization, and interplay with mitotic kinases in the regulation of CK2 activity. We first examined the role of four highly conserved mitotic phosphorylation sites located in the unique C-terminus of CK2?. Phosphospecific antibodies generated against the sites show that CK2? phosphorylation is temporally regulated and occurs during prophase and metaphase during normal mitotic progression. Proper phosphorylation of CK2? is required for proper mitotic progression, as stable cell lines expressing phosphorylation site mutants of CK2? display severe mitotic defects. We next examined the impact of these phosphorylation events on the subcellular localization of CK2. We show that CK2?, but not CK2?', localizes to the mitotic spindle. Localization of CK2? to the mitotic spindle is phosphodependent, and requires the peptidyl-prolyl isomerase Pin1. These results are a rare example of functional divergence between the two catalytic isoforms of CK2, and suggest that the role of CK2? phosphorylation during mitosis is to promote localization of CK2 to the mitotic spindle. Finally, we examined the possibility that CK2 activity during mitosis is regulated through hierarchal phosphorylation events, wherein CK2 would phosphorylate proteins only after priming phosphorylation events catalyzed by other mitotic kinases, particularly Cdk1. As this phenomenon has never been systematically investigated, we have investigated the consensus requirements for CK2 primed phosphorylation, and in particular Cdk/CK2 hierarchical phosphorylation. A genome-wide search for potential mitotic substrates matching the consensus sequence suggests that Cdk1/CK2 hierarchical phosphorylation may indeed contribute to mitotic signaling, particularly on the mitotic spindle. Taken together, our results confirm the importance of CK2 in mitotic cell division, and highlight several examples of subtle regulation of CK2, through phosphorylation, subcellular localization, and interplay with other protein kinases. This helps explain how CK2, a constitutively active kinase, can participate in tightly regulated cellular processes like mitosis.

Book excerpt: This is the final report of a three-year, Laboratory Directed Research and Development (LDRD) project as Los Alamos National Laboratory (LANL). Protein phosphorylation is well established as one of the most important mechanisms of signal transduction and cellular regulation. Two of the key enzymes that catalyze these phosphorylation reactions are the cAMP- (PKA) and cGMP- (PKG) dependent protein kinases. PKA has served as the prototypic model of this class of enzymes that now comprises in excess of 300 phylogenetically related proteins. A large number of these protein kinases are critical for the regulation of cell function and a full analysis of their similarities and differences is essential to understand their diverse physiological roles. The cAMP-dependent protein kinase has the subunit structure R2C2, in which C and R refer to the catalytic and regulatory subunits, respectively. The cGMP-dependent protein kinase (PKG) is highly homologous to PKA but is distinguished from it by having the regulatory and catalytic domains on a contiguous polypeptide. The studies described here use small-angle scattering and Fourier Transform InfraRed (FTIR) spectroscopy to study domain movements and conformational changes in these enzymes in different functional states in order to elucidate the molecular bases for the regulation of their activities.
