Book excerpt: From small beginnings in the early 1970s, the study of complement regulatory proteins has grown in the last decade to the point where it dominates the complement field. This growth has been fueled by the discovery of new regulators, the cloning of old and new regulators, the discovery that many of the regulators are structurally and evolutionarily related to each other and the development of recombinant forms for use in therapy. There are now more proteins known to be involved in controlling the complement system than there are components of the system and the list continues to grow. The time is ripe for a comprehensive review of our current knowledge of these intriguing proteins. This book does just that. The first few chapters discuss the "nuts-and-bolts" of the complement regulators, describing their structures, functional roles and modes of action. The roles of the complement regulators in vivo are then described, focusing on the consequences of deficiency, roles in the reproductive system, interactions with pathogens and exploitation for therapy. The interesting developments in defining the complement regulators expressed in other species are also discussed. The book is written as a monograph, albeit by two people. The text is as readable as possible without compromising on scientific accuracy and completeness. The conversational style very evident in some sections is deliberate! Placing all references in a single bibliography at the end of the text further improves readability. The reader will go to the book to discover a specific fact but be persuaded to read more and derive pleasure from the process. Book excerpt: The complement system is a group of proteins which plays a major role in the processing and removal of microorganisms and tissue breakdown products from the circulation and extracellular spaces. The system is activated by a wide range of targets, and activation leads to the production of opsonins, chemotaxis of granulocytes, cell lysis and other biological activities. Inappropriate overactivation of the system contributes to inflammatory tissue damage in the host, while inadequate activation leads to accumulation of immune complexes and other debris in the circulation, and susceptibility to infection. The biology and biochemistry of the system is now adequately understood, and attempts can be made to manipulate the activation and activities of the system for potential therapeutic purposes. The reviews in this volume summarise what is known of the ways in which the complement system can be activated, by interaction with antibodies, microorganisms, cell debris, and complex carbohydrates and how the activities and activation of the system have been modified, accidentally or by design, in vitro or in vivo by drugs, venoms, particulate carbohydrates, specific antibodies, synthetic peptides and other reagents.

While antimicrobial peptides, after cleavage of a preproprotein, have the ability to insert directly in non host membranes, complement requires a sequential enzymatic activation in the fluid phase in order to produce a transmembrane membrane attack complex. Its insertion is controlled by membrane bound regulators. Deficiencies are described for both effectors and relate to increased susceptibility of infection. In addition, however, antimicrobial peptides and complement each influence the activity of inflammatory cells as recent data in the respective research areas shows. This series of articles draws together for the entities of antimicrobial peptides and complement a balance of contributions in the areas of evolution, roles, functions and preclinical applications. By comparing and contrasting antimicrobial peptides and complement, greater cross-disciplinary appreciation will be derived for their individual and overlapping spectra of activity, circumstances of activation and their general ability to more completely inform the inflammatory and cellular response.

Book excerpt: The idea of Professors Bolis and Gilles to gather together for a 3 days' meeting in the splendid environment of Crans-Montana in Switzerland a limited number of people around the subject of calcium and calcium bind ing proteins seemed at first particularly attractive, and when they asked me to take charge of the scientific organization of the symposium, I accepted with enthusiasm. It rapidly became clear that the major problem would be the selection of the topics, since it was impossible to cover completely and in depth such a broad and dynamic area of research. In our view, one imperative was to associate as intimately as possible the structural and the functional aspects of the areas covered. Apart from one whole day focused on the fascinating roles played by calmodulin in cellular activities, the other sessions were devoted to calmodulin-related calcium binding proteins in muscle and non muscle tissues and to some selected biological systems such as mitochondria, secretory cells or sarcoplasmic reticulum in which calcium also plays a crucial role. The presentations were made by leading investigators in their field. Some of them do not, however, appear in the present volume, for which there are two reasons: first, some of the contributions were somewhat outside the scope of the book; second, three speakers, for valid reasons, simply found no opportunity to write a manuscript in the allotted time.

Book excerpt: The concept of innate immunity refers to the first-line host defense that serves to limit infection in the early hours after exposure to microorganisms. Recent data have highlighted similarities between pathogen recognition, signaling pathways, and effector mechanisms of innate immunity in Drosophila and mammals, pointing to a common ancestry of these defenses. In addition to its role in the early phase of defense, innate immunity in mammals appears to playa key role in stimulating the subsequent clonal response of adaptive immunity. Recent exciting information has determined that the templates that are laid down in primitive life forms, like flowering plants and insects, form the basic principles of first line host defense that are conserved in mammalian systems. The next frontier in the field is to understand the dynamic adaptive changes that occur as a result of the inter play between host defenses and infectious agents. One emerging theme is that microorganisms are constantly seeking ways to co-opt host defenses. On the other hand, host defense to infection is mediated by the coordinate action of pattern recognition molecules and receptors that, in mammals, are important and probably necessary antecedents to the development of an adaptive immune response. Innate Immunity aims to explore the intersection between host pathogen interactions across an evolutionary spectrum that will inform our understanding of the dynamic interplay between infectious agents and host defense in man.
