Book excerpt: The use of Compound-specific Stable Isotope Analysis (CSIA) is increasing in many areas of science and technology for source allocation, authentication, and characterization of transformation reactions. Until now, there have been no textbooks available for students with an analytical chemical background or basic introductory books emphasising the instrumentation and theory. This book is the first to focus solely on stable isotope analysis of individual compounds in sometimes complex mixtures. It acts as both a lecture companion for students and a consultant for advanced scientists in fields including forensic and environmental science. The book starts with a brief history of the field before going on to explain stable isotopes from scratch. The different ways to express isotope abundances are introduced together with isotope effects and isotopic fractionation. A detailed account of the required technical equipment and general procedures for CSIA is provided. This includes sections on derivatization and the use of microextraction techniques in GC-IRMS. The very important topic of referencing and calibration in CSIA is clearly described. This differs from approaches used in quantitative analysis and is often difficult for the newcomer to comprehend. Examples of successful applications of CSIA in food authenticity, forensics, archaeology, doping control, environmental science, and extraterrestrial materials are included. Applications in isotope data treatment and presentation are also discussed and emphasis is placed on the general conclusions that can be drawn from the uses of CSIA. In this regard, nitrogen-15 (15N)-labelled fertiliser was used as a biochemical and physical stable isotope tracer in laboratory and field experiments. The major outcome of the research was the development, validation and application of a new compound-specific amino acid 15N stable isotope probing method for assessing the assimilation of fertiliser nitrogen by soil microbial biomass. The novelty of the method lies in its tracing of incorporated nitrogen into newly biosynthesised microbial protein in time-course experiments using gas chromatography-combustion-isotope ratio mass spectrometry. The approach provides previously unattainable insights into the microbial processing of different nitrogen fertilisers in different soils. Further, it identifies the mechanistic link between molecular-scale processes and observations of field-scale fertiliser nitrogen immobilisation studies. The method and the results presented here will have far-reaching implications for the development of enhanced recommendations concerning farm-based soil management practices for increasing soil productivity and reducing nitrogen losses, which is essential to minimising environmental impacts.

Yet, while the age of information has exponentially increased our access to knowledge, we can see, as clearly as ever, that scientific knowledge is neither apolitical nor dogma-free, and it certainly is not disconnected from power. It is hard to imagine another era when the separation between science and politics has been this blurred as it is today. At the same time, it is true that no other topic than climate change has been so politically charged, with one side dominating the scientific narration and branding anyone opposing the mainstream as a “climate change denier,” and the other standing in staunch defiance that climate change exists. In an age of political and scientific turmoil, how can we navigate out way to coming towards a more objective understanding of the scientific issues surrounding the climate change debate?
