Book excerpt: The feasibility of sperm cells to act literally as gene vehicles was discussed using one of the most modern techniques in this aspect which is known as restriction enzyme mediated integration sperm mediated gene transfer (REMI-SMGT)technique. Several objectives were pursued in this book; such as to test the validity of REMI-SMGT, to evaluate the efficiency of this technique in generating mammalian species rather than the sheep that made only by one group and to observe the possibility of doing so by using rabbits as a model for this approach. Add to that, it was aimed to reduce the cost of REMI-SMGT by substituting liposomes and highly cost effective media with a high efficient, non-cost effective substitute. Moreover, it was directed to identify if any band substitution was occurred by simple seminal proteins - exogenous DNA incubation by one gel double stained non-radioactive method to identify the role of seminal proteins charge on the nature of seminal fluid inhibitory effect that exerted against the invading DNA. This might help us to approach closely to the nature of inhibitory activities in seminal proteins that prevent the entry of foreign DNA into the head of the sperm.

Book excerpt: The establishment of microinjection protocols about 20 years ago for cultured cells and shortly thereafter for the generation of transgenic mice by microinjection of DNA into fertilized mouse eggs greatly influ enced many fields of biology. Not only have the data generated using these approaches contributed to a large extent to our present under standing of gene regulation and cellular function of higher eukaryotic cells, but current knowledge and future developments in this area will certainly have a great impact on basic and applied research for many years to come. This laboratory manual describes the current state of the art in this research area and focuses primarily on both the experimental strategies with an extensive bibliography and the detailed procedures. A large number of studies are presently being performed and a great variety of new experimental designs are rapidly being developed. The book con tains protocols on injection of somatic cells as well as on injection of embryos, the use of similar equipment being a common feature. In the articles dedicated to somatic cells, full descriptions of the manual and automatic injection systems are given as well as the methods for the analysis of injected cells by video-microscopy, electron microscopy or in situ hybridizations. In addition, comprehensive protocols are given for injection experiments with very different purposes, such as to study sig nal transduction or microtubule dynamics.

No entanto esta metodologia apresenta diversosfatores negativos: consome tempo e trabalho, exige qualificação técnica, háelevada mortalidade de embriões, a percentagem de integração é freqüentementebaixa, a opacidade dos ovos de peixes não permite a visualização do pró-núcleo, além da membrana coriônica ser muito resistente em muitas espécies de peixestornando-se uma barreira à microinjeção. Uma alternativa de otimização doprocesso de inserção de DNA exógeno, teoricamente consiste na transfecçãomassal de células espermáticas pré fertilização in vitro. A transferência gênicamediada por espermatozóides (SMGT) vem sendo utilizada no estudo e geraçãode diversas espécies de animais transgênicos, inclusive peixes. Algumasferramentas como eletroporação, diferencial osmótico e incubação espermáticasão alternativas que podem potencializar a SMGT em peixes. Este estudo teve porobjetivo avaliar o potencial espermático do jundiá (Rhamdia quelen) submetido adiferentes tratamentos de inserção de DNA exógeno para gerar jundiástransgênicos. Para atingir este objetivo utilizou-se dois vetores pCMV-GFP epcBA-GFP, linearizados, em 5 tratamentos propostos: desidratação/reidratação (DR); eletroporação (E); desidratação/reidratação/eletroporação (DRE);incubação com plasma seminal (INC) e incubação sem plasma seminal (INCSP).Como controle sêmen diluído em solução isotônica sem DNA exógeno. Paraavaliar o potencial espermático foram mensurados atividade espermática (AT), tempo de permanência em atividade (TPA), integridade de membrana efertilização in vitro (FIV) através das TXFIV (taxa de fertilização in vitro) e TXECLO(taxa de eclosão). Book excerpt: Heritable human genome editing - making changes to the genetic material of eggs, sperm, or any cells that lead to their development, including the cells of early embryos, and establishing a pregnancy - raises not only scientific and medical considerations but also a host of ethical, moral, and societal issues. Human embryos whose genomes have been edited should not be used to create a pregnancy until it is established that precise genomic changes can be made reliably and without introducing undesired changes - criteria that have not yet been met, says Heritable Human Genome Editing. From an international commission of the U.S. National Academy of Medicine, U.S. National Academy of Sciences, and the U.K.'s Royal Society, the report considers potential benefits, harms, and uncertainties associated with genome editing technologies and defines a translational pathway from rigorous preclinical research to initial clinical uses, should a country decide to permit such uses. The report specifies stringent preclinical and clinical requirements for establishing safety and efficacy, and for undertaking long-term monitoring of outcomes. Extensive national and international dialogue is needed before any country decides whether to permit clinical use of this technology, according to the report, which identifies essential elements of national and international scientific governance and oversight.
