Book excerpt: The fundamental knowledge of Cr(III) chemistry has been enhanced via detailed structural and spectroscopic studies of largely novel compounds that may potentially be active tri- and tetramerisation precursors. The compounds are based on various monodentate and bidentate nitrogen and phosphorus ligands which have been coordinated to [CrCl3 (thf) 3]. The few compounds that have been synthesised previously have in this study been made via novel synthetic routes and incorporate a combination of new and more detailed analysis than was carried out previously. The eight structures determined, in addition to offering novel crystallographic data, also provided insights into the synthetic pathways leading to compound formation. The isolation of monomeric structures suggests direct ligand substitution, while the cationic-anionic structures suggest the presence of dimeric intermediates which have been cleaved asymmetrically. Infrared and Raman spectra of these structures were able to add weight to these pathway proposals and, by means of vibrational comparisons, assisted in the general band assignments of the compounds' spectra where structures were not available. Vibrational shifts relative to the free ligands, as well as metal ligand vibrations in the far infrared region, were also of significant value in terms of ligand coordination and geometry. Closely associated with the infrared and Raman spectra analysis was the generation of theoretical spectra using Density Functional Theory calculations. The excellent agreement between the calculated and experimental spectra confirmed the vibrational assignments. Also generated by computational means were the highest occupied and lowest unoccupied molecular orbitals (HOMO and LUMO) of the compounds which indicated the sites of potential nucleophilic and electrophilic attack. 1H NMR spectroscopy is a technique normally avoided when studying paramagnetic materials. However, by employing a largely novel approach, information pertaining to both ligand coordination and reaction times was obtained. FAB-MS assisted in the confirmation that the single crystal determinations did indeed reflect the composition of the bulk precipitated samples. It also provided additional structural information through the identification of fragmentation patterns which could not be gained by techniques such as elemental analysis.

Book excerpt: Metalloproteins comprise approximately 30% of all known proteins, and are involved in a variety of biologically important processes, including oxygen transport, biosynthesis, electron transfer, biodegradation, drug metabolism, proteolysis, and hydrolysis of amides and esters, environmental sulfur and nitrogen cycles, and disease mechanisms. EPR spectroscopy has an important role in not only the geometric structural characterization of the redox cofactors in metalloproteins but also their electronic structure, as this is crucial for their reactivity. The advent of x-ray crystallographic snapshots of the active site redox cofactors in metalloenzymes in conjunction with high-resolution EPR spectroscopy has provided detailed structural insights into their catalytic mechanisms. This volume was conceived in 2005 at the Rocky Mountain Conference on Analytical Chemistry (EPR Symposium) to highlight the importance of high-resolution EPR spectroscopy to the structural (geometric and electronic) characterization of redox active cofactors in metalloproteins. We have been fortunate to have enlisted internationally recognized experts in this joint venture to provide the scientific community with an overview of high-resolution EPR and its application to metals in biology. This volume, High-Resolution EPR: Applications to Metalloenzymes and Metals in Medicine, covers high-resolution EPR methods, iron proteins, nickel and copper enzymes, and metals in medicine. An eloquent synopsis of each chapter is provided by John Pilbrow in the Introduction. A second volume, Metals in Biology: Applications of High-Resolution EPR to Metalloenzymes, will appear later this year covering the complement of other metalloproteins. One of the pioneers in the development of pulsed EPR and its application to metalloproteins was Arthur Schweiger, whose contribution we include in this volume. Unfortunately, he passed away suddenly during the preparation of this volume. The editors and coauthors are extremely honored to dedicate this volume to the memory of Arthur Schweiger in recognition of his technical advances and insights into pulsed EPR and its application to metalloproteins. Arthur was extremely humble and treated everyone with equal respect. He was a gifted educator with an ability to explain complex phenomena in terms of simple intuitive pictures, had a delightful personality, and continues to be sadly missed by the community. It is an honor for the editors to facilitate the dissemination of these excellent contributions to the scientific community. Suggestions for future volumes are always appreciated.

Book excerpt: The polarized crystal spectra of a series of tetragonal trans- diacidobis (ethylenediamine) chromium (III) complexes, trans-[Cr(en)2XY], where X= Y = Br-, H2O, F-, Cl- and X= H2O, Y = OH-, F-, have been measured at liquid nitrogen temperature, achieving better resolution of the split components of the cubic spin-allowed bands, and also definite energy level assignments. The observed band positions have been fitted with the predicted transition energies by the use of the appropriate energy equations for d^3 configuration immersed in tetragonal fields with full configuration interaction. The ligand field parameters, which include the cubic and axial, and the electron correlation parameter B have been evaluated by such a fitting. The derived parameters have been analyzed and their significance discussed. Using thick crystals, we have uncovered a rich doublet structure in some of the complexes and tentative values of the electron correlation parameter C have been obtained for all the complexes.
