Book excerpt: A study was made showing that lipid films in the dry state behave as electrical semiconductors with high activation energies. For phospholipids, these activation energies range from 4.8 to 6.3 e.v. Upon hydration, the activation energies decrease whereas the conductivities increase. An effect parallel to that of water is found with exposure of lipids to iodine vapor; here the activation energies range from 2.7 to 3.2 e.v. With the adsorption of either water or iodine, the dielectric constant, frequency dispersion and dissipation factor of the lipids increased over the dry state values. Lipid bilayers were found to exhibit an increase of 1000 to 100000 in their electrical conductivity with exposure of the bilayer to iodine solution. Spectroscopic evidence was obtained indicating that lipids form charge-transfer complexes with iodine. This charge transfer evidence is taken as suggestive of a possible electronic conduction mechanism, not only in the lipid films, but also in the lipid bilayers.

Raman scattering (RS) is a widely used vibrational technique providing highly specific molecular spectral patterns. A severe limitation for the application of this spectroscopic technique lies in the low cross section of RS. Surface-enhanced Raman scattering (SERS) spectroscopy overcomes this problem by 6-11 orders of magnitude enhancement compared with the standard RS for molecules in the close vicinity of certain rough metal surfaces. Thus, SERS combines molecular fingerprint specificity with potential single-molecule sensitivity. Due to the recent development of new SERS-active substrates, labeling and derivatization chemistry as well as new instrumentations, SERS became a very promising tool for many varied applications, including bioanalytical studies and sensing. Both intrinsic and extrinsic SERS biosensing schemes have been employed to detect and identify small molecules, nucleic acids and proteins, and also for cellular and in vivo sensing.

Free radicals form in biological systems through metabolism, but it is also realized that exogenous environmental sources, such as radiation, food, and drugs, contribute significantly to the generation of free radicals in biological systems. Being reactive species, free radicals are short-lived and do not travel far from cellular targets. Their concentration in biological systems is very low and is difficult to detect directly by electron spin resonance spectroscopy (ESR). Indirect methods of reactions of radicals with specific biomolecules are also sufficiently sensitive to detect quantitatively their presence. Thus the response of antioxidant defenses which react with radical species, can serve as an indirect measure that free radicals have been formed. Redox-based antioxidants change their oxidation state and antioxidants become free radicals themselves. Often, however, the antioxidants give rise to more persistent free radicals, sometimes owing to delocalization of the lone electron around ring structures (in vitamin E, ubiquinones, and certain carotenes). Persistent free radicals react only rarely and the precursors often can be regenerated in biological systems. In recent years, it is becoming clearer from biochemical studies on how the major lipophilic antioxidants work. Particular attention has been given to vitamin E and quinones found in animal and plant membranes and in carotenoids, for the protection of membranes in lipoprotein systems. Flavonoids form another rich and varied source of natural antioxidants.
