Bioremediation and biotransformation processes harness the naturally occurring ability of microbes to degrade, transform or accumulate a wide range of organic pollutants. Major methodological breakthroughs in recent years through detailed genomic, metagenomic, proteomic, bioinformatic and other high-throughput analyses of environmentally relevant microorganisms have provided us unprecedented insights into key biodegradative pathways and the ability of organisms to adapt to changing environmental conditions. The degradation of a wide spectrum of organic pollutants and wastes discharged into the environment by anthropogenic activities is an emerging need today to promote sustainable development of our society with low environmental impact. Microbial processes play a major role in the removal of recalcitrant compounds taking advantage of the astonishing catabolic versatility of microorganisms to degrade or transform such compounds. New breakthroughs in sequencing, genomics, proteomics, bioinformatics and imaging are generating vital information which opens a new era providing new insights of metabolic and regulatory networks, as well as clues to the evolution of degradation pathways and to the molecular adaptation strategies to changing environmental conditions. Functional genomic and metagenomic approaches are increasing our understanding of the relative importance of different pathways and regulatory networks to carbon flux in particular environments and for particular compounds. New approaches will certainly accelerate the development of bioremediation technologies and biotransformation processes in coming years for natural attenuation of contaminated environments

Book excerpt: The EPA commissioned The National Academies to provide advice on the vexing question of whether and, if so, under what circumstances EPA should accept and consider intentional human dosing studies conducted by companies or other sources outside the agency (so-called third parties) to gather evidence relating to the risks of a chemical or the conditions under which exposure to it could be judged safe. This report recommends that such studies be conducted and used for regulatory purposes only if all of several strict conditions are met, including the following: The study is necessary and scientifically valid, meaning that it addresses an important regulatory question that can't be answered with animal studies or nondosing human studies; The societal benefits of the study outweigh any anticipated risks to participants. At no time, even when benefits beyond improved regulation exist, can a human dosing study be justified that is anticipated to cause lasting harm to study participants; and All recognized ethical standards and procedures for protecting the interests of study participants are observed. In addition, EPA should establish a Human Studies Review Board (HSRB) to evaluate all human dosing studiesâ€"both at the beginning and upon completion of the experimentsâ€"if they are carried out with the intent of affecting the agency's policy-making.

Diazinon acts on a wide range of insects on crops, gardens, livestock, and pets, but most uses have been restricted in the USA, Canada, and the European Union since the 1980s. Glyphosate is the most heavily used agricultural and residential herbicide in the world, and has been detected in soil, air, surface water, and groundwater, as well as in food. Malathion is one of the oldest and most widely used organophosphate insecticides, and has a broad spectrum of applications in agriculture and public health, notably mosquito control. The insecticide parathion has been largely banned or restricted throughout the world due to toxicity to wildlife and humans. Tetrachlorvinphos is banned in the European Union, but continues to be used in the USA and elsewhere as an insecticide on animals, including in pet flea collars. The IARC Monographs Working Group reviewed epidemiological evidence, animal bioassays, and mechanistic and other relevant data to reach conclusions as to the carcinogenic hazard to humans of these agents.

Since house flies first developed resistance to DDT in 1946, more than 428 species of arthropods, at least 91 species of plant pathogens, five species of noxious weeds and two species of nematodes were reported to have developed strains resistant to on~ or more pesticides. A seminar of U. S. and Japanese scientists was held in Palm Springs, California, during December 3-7, 1979, under the U. S. -Japan Cooperative Science Program, in order to evaluate the status of research on resistance and to discuss directions for future emphasis. A total of 32 papers were presented under three principal topics: Origins and Dynamics of Resistance (6), Mechanisms of Resistance (18), and Suppression and Management of Resistance (8). The seminar was unique in that it brought together for the first time researchers from the disciplines of entomology, plant pathology and weed science for a comprehensive discussion of this common problem. Significant advances have been identified in (a) the development of methods for detection and monitoring of resistance in arthropods (electrophoresis, diagnostic dosage tests) and plant pathogens, (b) research on biochemical and physiological mechanisms of resis tance (cytochrome p450, sensitivity of target site, gene regulation), (c) the identification and quantification of biotic, genetic and operational factors influencing the evolution of resistance, and (d) the exploration of pest management approaches incorporating resis tance-delaying measures.
