This study was conducted to determine whether the M1 and M2 macrophage densities in NSCLC are associated with patient's survival time.

Book excerpt: Malignant pleural mesothelioma (MPM) is a rare thoracic cancer that derives from the mesothelial cells of the pleura and is causally associated with exposure to asbestos. Because of its aggressiveness and resistance to therapies, MPM prognosis is extremely poor and the 5-year survival rate is approximately 10%. Despite its poor effectiveness, chemotherapy with cisplatin and pemetrexed has long remained the standard-of-care for upfront treatment with no second line therapy available. Recently, the approval of ipilimumab (anti-CTLA-4) plus nivolumab (anti-PD-1) for the first-line treatment of unresectable MPM has marked a significant milestone, in particular for the non-epithelioid MPM subtype, which is more resistant to chemotherapy and associated with a poorer outcome. However, most MPM patients are still resistant to cancer therapies. Beyond histologic subtypes, recent studies have highlighted extensive genetic variation and gene expression deregulation both between and within MPM patients. This molecular heterogeneity highlights the need for a personalized treatment approach. Therefore, both multiple therapeutic options and predictive biomarkers that can guide clinical decision-making are urgently needed. Tumor cell immunogenicity and tumor microenvironment composition (TME) are the major determinants of the success of immunotherapy. Despite the low tumor mutational burden, recent studies indicate that chromoanagenesis and chromotripsis are molecular features of MPM. Since chromosomal instability can lead to the expression of neo-antigens, it could also influence the efficacy of immune checkpoint inhibitors (ICIs). The immunosuppressive TME is an additional important hurdle that limits the efficacy of immunotherapy. The combination of ICIs with strategies to boost anti-tumor immunity by restraining immunosuppressive myeloid cells or by reprograming “cold tumor” into “hot tumor” is an active area of research. This Research Topic aims to discuss recent advances in understanding and targeting MPM immune microenvironment for therapeutic purposes along with potential predictive biomarkers to select patients who most likely will respond to immunotherapies. We are interested in basic and clinical studies that explore the crosstalk between tumor and immune cells in order to highlight new molecular targets, approaches, and combination treatments to enhance anti-tumor immunity in MPM and assess potential predictive biomarkers of response to immunotherapy.

With the on-going debate about the requirement of EMT for cancer metastasis, an emerging focus on intermediate states of EMT and its reverse process mesenchymal-epithelial transition (MET) offer new ideas for metastatic requirements and the dynamics of EMT/MET during the entire metastatic cascade. Therefore, we would like to initiate discussions on viewing EMT and its downstream signaling networks as a fulcrum of cellular plasticity, and a facilitator of the adaptive responses of cancer cells to distant organ microenvironments and various therapeutic assaults. We hereby invite scientists who have prominently contributed to this field, and whose valuable insights have led to the appreciation of epithelial-mesenchymal plasticity as a more comprehensive mediator of the adaptive response of cancer cells, with huge implications in metastasis, drug resistance, tumor relapse, and patient survival.

Book excerpt: This book is focused on the analysis of the role played by immune cell components in the angiogenic process associated with inflammation and tumor growth. Both innate and adaptive immune cells are involved in the mechanisms of endothelial cell proliferation, migration and activation, through the production and release of a large spectrum of pro-angiogenic mediators. These may create the specific microenvironment that favors an increased rate of tissue vascularization. The link between chronic inflammation and tumorigenesis was first proposed by Rudolf Virchow in 1863 after the observation that infiltrating leukocytes are a hallmark of tumors and first established a causative connection between the lymph reticular infiltrate at sites of chronic inflammation and the development of cancer. Tumors were described as wounds that never heal and surgeons have long described the tendency of tumors to recur in healing resection margin and it has been reported that wound healing environment provides an opportunistic matrix for tumor growth. As angiogenesis is the result of a net balance between the activities exerted by positive and negative regulators, this book will also provide information on some anti-angiogenic properties of immune cells that may be utilized for a potential pharmacological use as anti-angiogenic agents in inflammation as well as in cancer. The work is written for researchers in the field and also for graduate students which approach this matter.
