Book excerpt: The longterm goal of our work is to elucidate the pattern of expression of the five somatostatin receptor (SSTR1-5) isoforms in breast cancer, to determine which SSTR subtypes and signalling mechanisms mediate the antiproliferative effects of somatostatin (SST), whether the available SST analogs are effective in binding to these antiproliferative SSTR subtypes, and whether the pattern of SSTR expression in tumors can provide an independent prognostic marker. Towards these goals, we have characterized the pattern of expression and relative abundance of mRNA and proteins for SSTR1-5 in 101 primary breast cancers. All tumors expressed at least one SSTR and the majority expressed multiple SSTRs including subtypes 2, 3, and 5 which bind to octapeptide SST analogs. A good correlation was found between SSTR protein expression by immunocytochemistry and mRNA analysis by RT-PCR. SSTRs exert significant antitumor activity both by cytostatic and cytotoxic (apoptotic) actions in breast cancer cells. Apoptosis is dependent on estrogen receptor expression and is potentiated by tamoxifen. Apoptosis is induced uniquely via the SSTR subtype and is associated with induction of wild type p53 and of Bax and endonuclease 2. By contrast, the other four SSTR subtypes signal cell cycle arrest with the following rank order - SSTR5> SSTR2> SSTR4> SSTR1. These changes are associated with the induction of the retinoblastoma protein pRB and the cyclin-dependent kinase inhibitor p21.

The secretion of these hormones is under the control of releasing and inhibiting factors released from the hypothalamus. The neuropeptide somatostatin (SOM) is the major hypothalamic regulator of pituitary gland hormone secretion. Five receptor subtypes (SSTR1-5) bind and transduce SOM signaling. Because SSTR2 is the major receptor subtype, my goal was to understand the physiological trafficking of SSTR2 following SOM binding in neuroendocrine cells. Several observations had suggested that SSTR2 cell surface expression is regulated by SSTR5 co-expression. Thus, we aimed to understand how SSTR5 regulates SSTR2 trafficking by activating SSTR2 with a selective agonist (L-779,976) in the presence or absence of SSTR5. The results from this thesis demonstrate that SSTR5 does not regulate SSTR2 trafficking but rather agonistic property of L-779,976 is responsible for selective SSTR2 trafficking pattern induced by this agonist. Additionally, we demonstrated that following SOM binding, SSTR2 traffics to a juxta-nuclear compartment. Excitingly, the effort to characterize this compartment resulted in the identification of a new class of storage vesicles in pituitary cells homologous to GLUT4 storage vesicles present in myocytes and adipocytes. We called those vesicles GLUT4 like storage vesicles (GLSVs) and showed that they harbor internalized SSTR2 and they are responsible for the regulated surfacing of SSTR2 in response to activation of hypothalamic releasing factor receptors on pituitary cells. We demonstrate that this mechanism fine-tunes pituitary gland hormones release. Therefore, in this doctoral thesis we have provided a detailed characterization of SSTR2 trafficking and identified a new storage carrier in pituitary cells that stores internalized SSTR2 and contributes to the regulation of pituitary hormone secretion"--
