Book excerpt: Recent progress in brain science has been remarkable, especially with regard to advances in the area of neuroscience. Particularly in the past decade, there have been many important discoveries about signal transduction in the brain. With this background, biological research in affective disorders has become well developed in relation to neural signaling. However, this field is fairly hard to understand comprehensively, and there is relatively little integrative work with clinical psychiatry in spite of the involvement of a wide variety of scien tific disciplines. This monograph brings together up-to-date reviews from several young Japanese scientists who work in basic and clinical neuroscience. The intention is to explain in plain language the information that has evolved on signal transduction in terms of the biological abnormalities and mechanisms of anti depressants. The model shown on the cover (and on page 24) is intended to help the reader understand signal transduction in the brain and the patho physiology of affective disorders as well as the mechanisms of antidepressants.

In each cascade the first messenger gives rise to the appearance of a second messenger such as cyclic AMP, cyclic GMP, or diacylglycerol, which in turn triggers a third messenger, a fourth messenger, and so forth. Many advances in elucidating such pathways have been made, including efforts to link messenger molecules to brain processes operative in health or disease. However, the latter type of information, relating signaling pathways to brain function, is scattered across a variety of publication media, which makes it difficult to integrate the multiple roles of different signaling cascades into our understanding of brain function in health and disease. The primary aim of Cerebral Signal Transduction: From First to Fourth Messengers, therefore, is to offer a comprehensive picture of the recent advances made in the signaling field as it relates to neuronal and cere bral function. The current state of progress provides an exciting opportunity for such a comprehensive focus because molecular tools have become available to selectively remove, reduce, or enhance spe cific components in the signaling pathways, e. g. , by interfering with the genes encoding key proteins. First, application of the full range of modern techniques in behavioral, neurochemical, and molecular biology have greatly improved our understanding of basic clock mechanisms. In several species the genetic basis of the circadian clock is being progressively delineated. A complete picture of the neurochemical and neuroanatomical structure of the mammalian clock is emerging and the complex pattern of control mechanisms involving endogenous clock mechan isms and photic and nonphotic zeitgebers is being built up as a result of behavioral studies. Secondly, in parallel with these exciting developments in basic science, clinical applications are being convincingly demonstrated in the general fields of pharmacology and medicine as well as in specific areas, e.g., jet lag, shiftwork maladaption syndrome, blindness, and cardiovascular system.
