The first chapter deals with Receptor Tyrosin Kinases (RTKs) with emphasis on the Epidermal Growth Factor Receptor (EGF receptor) and the Platelet Derived Growth Factor Receptor (PDGF receptor). The second chapter deals with the RTK c-Met and with how this RTK becomes carcinogenic. The third chapter reviews recent understanding on the mechanisms of action of the numerous fibroblast growth factors and their receptors. In the fourth chapter we learn about the trafficking of and signalling from the Growth Hormone Receptor and how this receptor is controlled by ubiquitination. The fifth chapter is devoted to the Interleukin II receptor, essential for activation of T cells. Links between ubiquitination, signalling, endocytosis, and sorting are reviewed. The last chapter discusses current views on how monoubiquitination controls both signalling and trafficking and thereby the final outcome of receptor activation.

A short account of the history of the effort led to the Phosphate Workshops is appro priate and can be of interest to the reader. The idea for Phosphate Workshops was born in the early days of November, 1974. One of us (S. G. M. ) suggested the thought to a group of scientists gathered for a luncheon in one of the attrac tive small restaurants in Weisbaden, Germany. The purpose of the workshop was to bring together interested scientists to discuss the newer developments and the recent advances in the field of phosphate metabolism and the other related minerals. An Organizing Committee made of Shaul G. Massry (USA), Louis V. Avioli (USA), Philippe Bordier (France), Herbert Fleisch (Switzerland), and Eduardo Slatopolsky (USA) was formed. The First Workshop was held in Paris during June 5-6, 1975 and was hosted by Dr. Philippe Bordier. Its proceeding was already published. The Second Workshop took place in Heidelberg during June 28-30, 1976 and was hosted by Dr. Eberhard Ritz. Both of these workshops were extremely successful scientific endeavors, and the need for them was demonstrated by the great interest they generated among the scientific community. The Or ganizing Committee, therefore, decided to continue with the tradi tion to hold additional Workshops annually or every other year.

Book excerpt: The fibroblast growth factor (FGF) family of ligands and the corresponding family of FGF receptor tyrosine kinases comprise one of the most versatile and diverse growth factor signaling families in vertebrates, found virtually in every tissue and cell type where they regulate metabolic and physiologic function, maintain tissue homeostasis, and mediate injury response, tissue repair, and regeneration. As a result, FGFs play critical roles in a wide variety of normal biological and abnormalities in FGF signaling can lead to a variety of disease states from developmental malformations, to atherosclerosis, pulmonary hypertension and cancer among many others. A thorough understanding of this system is necessary for critical insights into both normal and disease biology and is essential to development of rationale therapeutic strategies aimed at treatment of FGF-dependent diseases states.Recent years have seen advances in our understanding of FGFs role in endothelial and, more general, vascular biology, the subject of this book. In particular, FGFs have been implicated in the maintenance of vascular homeostasis and control of blood vessel permeability while aberrant FGF signaling has been shown to be central to the development of pulmonary hypertension, and endothelial-to-mesenchymal transition, a process implicated in the development of atherosclerosis. Finally, FGFs ability to stimulate blood vessel growth has been explored in therapeutic angiogenesis approaches.With this in mind, the present monograph is structured to provide comprehensive information to a reader interested in FGFs in general and vascular biology in particular. To this end, we explore everything from the structure and signaling of FGFs and their receptors (Chapters 1, 2 and 3) to their role in the regulation of vascular homeostasis (Chapter 4) and cardiovascular diseases (Chapters 5 and 6). We then address new developments in FGF therapeutics (chapter 7) and explore FGF biology in the epithelium (Chapter 8) thereby providing a complete analysis of this growth factor family biology in two closely related but distinctly different environments — endothelium and epithelium. Finally, we examine FGF biology in eye disease and in cancer.It is our hope that this comprehensive treatment of FGF vascular biology and its application will provide the reader with the accurate and timely summary of this rapidly moving field.

Fortunately, the most significant conclusion to be drawn from the experimental embryology of the last two decades is the manner in which orthologous or closely related molecules are deployed to mediate similar - velopmental processes in both vertebrates and invertebrates. The molecular mechanisms regulating processes fundamental to most animals, such as axial patterning or axon guidance, are frequently conserved during evolution. (It is now widely believed that the differences between phyla and classes are the result of new genes, arising mostly by duplication and divergence of extant sequences, regulating the appearance of derived characters. ) Other vertebrates are obviously most likely to use the same devel- mental mechanisms as humans and, within the vertebrate subphylum, the - parent degree of conservation of developmental mechanism is considerable. It has long been recognized that particular vertebrate species offer either d- tinct advantages in investigating particular stages of development or are - pecially amenable to particular manipulations. No single animal can provide all the answers because not all types of experiments can be carried out on a single species.
