Book excerpt: Members of the genus Clostridium synthesize and secrete many antigenic substances into the nutrient medium in which the microorganisms have been cultivated. The study of the antigenic nature of these enzymes and/or exotoxings is important because the organisms of this genus which cause gas gangrene are extremely pathogenic and their taxonomic position can cause confusion at the time of a clinical diagnosis of the disease. A rapid serological method was described for differentiating two similar pathogens; i.e., Clostridium chauvoei and Clostridium septicum. Antisera were prepared against five strains of Cl. chauvoei, two strains of Cl. novyi type B, and five strains of Cl. septicum. Rabbits were used as the experimental animals for production of hyperimmune serum. Geinea pigs, as a source of antiserum specific for Cl. chauvoei, proved to be unsatisfactory. The bacterins for these immunizations were derived from Clostridium cultures grown by both conventional and cellophane sac techniques. Formaldehyde was used for inactivation of the cells and toxin and the bacterins were alum precipitated. Various immunization schedules were tested to find one advantageous for demonstration of antigenic similarity. The results obtained by antigenic analysis with the double immunodiffusion technique are reported. Using the hyperimmuned and commercial antiserum, the following reactions occurred between Cl. chauvoei and other species: There are at least four antigenic supstances produced by Cl. chauvoei that are precipitated by Cl. septicum antisera and vice versa. The antisera prepared against Cl. novyi type B reacted with Cl. chauvoei antigen without the production of lines of identity. Cl. chauvoei antisera and Cl. novyi types B and D antigen precipitatied as a dense zone around the well containing antiserum. The use of the cellophane sac technique is described in preparation of the antigen for both immunization and immunodiffusion. This technique has been used to concentrate the bacterial cells and their soluble products. This was accomplished by growing the microorganisms inside a cellophane sac that was suspended into a specially developed medium. The small diameter of the pores in the membrane prevents diffusion of molecules large enough to be antigenic. Therefore, the final antigen preparation was free of antigenic medium contituents. The microorganisms used for inoculation are grown in liquid media, centerfuged, and the packed cells constitute the inoculum deleting the major portion of the seed media antigens from the cellophane sac. The application of the results obtained from the immunodiffusion analyses aids in clinical diagnosis of the disease producing agents of blackleg and malignant edema. When antisera are available, a serological differentiation can be made based on immunodiffusion patterns obtained with homologous and/or heterologous antisera. This can be accomplished in forty-eight hours by inoculating a guinea pig with a sample from the infected bovine, inoculating a tube of nutrient medium with a piece of the dead guinea pig's liver, and inoculating the media and redording where the lines of precipitation develop.

In this volume, we have sought to remedy this misconception by compiling aseries of chapters which, together, provide a practically-oriented handbook of the biotechnologie potential of the genus. Clostridium is a broad grouping of organisms that together undertake a myriad of biocatalytic reactions. In the first two chapters, the reader is introduced to this diversity, both taxonomically and physiologically. In the following chapter, the current state of genetic analysis of members of the genus is reviewed. The remaining chapters concentrate on specific, exploit able aspects of individual Clostridium species-highlighting their range of unique capabilities (of potential or recognized industrial value), particu larly in the areas of biotransformation, enzymology, and the production of chemical fuels. Fittingly, the final chapter demonstrates that even the most toxic of the clostridia can be of therapeutic value. The contributors to this volume reflect the trans national interest in Clostridium, and we are indebted to each of them for making this volume possible. We particularly wish to acknowledge the contributions, both to this volume and to microbiology in general, of Dr. Elizabeth Cato, who, sadly, died shortly be fore publication ofthis volume. In addition to the PPLO specialists, the conference invited Julius Marmur to compare PPLO DNA to DNA of other organisms; David Garfinkel, who was one of the first to develop computer models of metabolism; Cyrus Levinthal to talk about coding; and Henry Quastler to discuss information theory constraints on very small cells. The conference was an announcement of the role of PPLO in the fundamental understanding of molecular biology. Looking back 40-some years to the Connecticut meeting, it was a rather bold enterprise. The meeting was international and inter-disciplinary and began a series of important collaborations with influences resonating down to the present. If I may be allowed a personal remark, it was where I first met Shmuel Razin, who has been a leading figure in the emerging mycoplasma research and a good friend. This present volume is in some ways the fulfillment of the promise of that early meeting. It is an example of the collaborative work of scientists in building an understanding of fundamental aspects of biology.
