Though many of the participating authors are urologists, there are also valuable contributions from medical oncologists, laboratory inv- tigators, and pathologists. One of the inherent problems in publishing a book that describes molecular techniques, especially those techniques that are rapidly evolving, is that some of these may become obsolete after a few years. However, many of the procedures detailed here are standard approaches that have already wi- stood the test of time. Reverse transcriptase PCR, or RT-PCR, a technique that produces m- tiple copies of selected sequences of DNA, has a sensitivity of a millionfold amplication of a single cell, and thus constitutes a very precise technique for measuring the presence of tumor cells. By combining this technique with genomic hybridization, cadherin,ß and metalloproteinase expression we may then reveal other important factors in the detection, staging, aggressiveness, and treatment of this disease.

Under anaerobic conditions S. oneidensis MR-1 attaches to insoluble minerals such as Fe(III) and Mn(IV) oxides and can use these as alternative electron acceptors. Biofilms of S. oneidensis MR- 1 were previously found to be dependent on the mxd gene cluster (mxdABCD). This study identified environmental and genetic factors regulating mxd -dependent and -independent biofilm formation in S. oneidensis MR-1 and a putative function for mxdB in LPS biosynthesis. Physiological experiments determined electron donor starvation as a factor responsible for mxd induction. Tn5 mutagenesis identified the two-component signaling system (TCS) ArcS/ArcA and a putative TCS BarA/UvrY as regulators of mxd expression. ccmC, important for cytochrome c maturation, was found to be involved in biofilm formation in S. oneidensis MR-1. Microarray analysis identified ccmC to play a role in aspects of anaerobic and fermentative metabolism in S. oneidensis MR-1. In conclusion, these data indicate a potential connection between energy metabolism of the cell, mxd and biofilm formation in S. oneidenis MR-1. The natural life cycle of the phylogenetically closest relative V. cholerae A1552 involves the transitioning of cells between different environmental surfaces such as the chitinous shell of Crustaceae and the epithelial layer of the human intestine and vice versa. Biofilms of V. cholerae A1552 vpsA and luxO mutants grown under hydrodynamic flow conditions were found to form pronounced, three-dimensional biofilms that resemble all aspects of wild type biofilms. By genetic experiments, it was shown that in hydrodynamically-grown biofilms this independence of vpsA is due to the expression of rpoS, which is a negative regulator of vpsA expression. Biofilms also underwent substantial metabolically controlled dissolution after 96 h that could be induced by a simple stop of medium flow across the biofilm. These studies indicate that metabolic conditions control the reversible attachment of cells to the biofilm matrix and are key in regulating biofilm cell physiology via rpoS. Furthermore, these results redefine the roles of vps and quorum-sensing in V. cholerae A1552 biofilms.

Book excerpt: Life presumably arose in the primeval oceans with similar or even greater salinity than the present ocean, so the ancient cells were designed to withstand salinity. However, the immediate ancestors of land plants most likely lived in fresh, or slightly brackish, water. The fresh/brackish water origins might explain why many land plants, including some cereals, can withstand moderate salinity, but only 1 – 2 % of all the higher plant species were able to re-discover their saline origins again and survive at increased salinities close to that of seawater. From a practical side, salinity is among the major threats to agriculture, having been one of the reasons for the demise of the ancient Mesopotamian Sumer civilisation and in the present time causing huge annual economic losses of over 10 billion USD. The effects of salinity on plants include osmotic stress, disruption of membrane ion transport, direct toxicity of high cytoplasmic concentrations of sodium and chloride on cellular processes and induced oxidative stress. Ion transport is the crucial starting point that determines salinity tolerance in plants. Transport via membranes is mediated mostly by the ion channels and transporters, which ensure selective passage of specific ions. The molecular and structural diversity of these ion channels and transporters is amazing. Obtaining the detailed descriptions of distinct ion channels and transporters present in halophytes, marine algae and salt-tolerant fungi and then progressing to the cellular and the whole organism mechanisms, is one of the logical ways to understand high salinity tolerance. Transfer of the genes from halophytes to agricultural crops is a means to increase salt tolerance of the crops. The theoretical scientific approaches involve protein chemistry, structure-function relations of membrane proteins, synthetic biology, systems biology and physiology of stress and ion homeostasis. At the time of compiling this e-book many aspects of ion transport under salinity stress are not yet well understood. The e-book has attracted researchers in ion transport and salinity tolerance. We have combined our efforts to achieve a wider, more detailed understanding of salt tolerance in plants mediated by ion transport, to understand present and future ways to modify and manipulate ion transport and salinity tolerance and also to find natural limits for the modifications.
