Abstract: Abstract of thesis entitled IRON AND RUTHENIUM COMPLEXES WITH NITROGEN AND OXYGEN DONOR LIGANDS FOR ANTI-CANCER AND ANTI-VIRAL STUDIES Submitted by Ella Lai-Ming Wong For the degree of Doctor of Philosophy at The University of Hong Kong in February 2006 Development of drug resistance in cancer chemotherapy has stimulated the search for new anti-cancer drugs. Due to the versatile oxidation states and diverse coordination chemistry, iron and ruthenium compounds are potential candidates for new anti-cancer agents and metallopharmaceuticals. In this study, iron(II) complexes containing polypyridines as auxiliary ligand and ruthenium-oxo oxalato cluster were synthesized and characterized by X-ray crystallography, and their anti-cancer and anti-viral properties were examined. The iron(II) complexes, [Fe(R-qpy)(CH CN) ](ClO ) [qpy = 3 2 4 2 2,2′ 6′,2″ 6″,2′″ 6′″,2″″-quinquepyridine, R = H (1a) and Ph (1b)] and [Fe(Py -OH)(CH CN)](ClO ) (2) [Py-OH = 5 3 4 2 5 2,6-bis[hydroxybis(2-pyridyl)methyl]pyridine] were stable in physiological buffers and exhibited significant cytotoxicities against some established human cancer cell lines. According to the MTT assay, their IC values were 0.1 - 70 50 M; they exhibited more pronounced cytotoxicities against hepatocellular carcinoma cells than against other type of cancer cells. Flow cytometry studies revealed that 1a could cause cell cycle arrest of HeLa cells at the S-phase, which involved DNA synthesis. Complex 1a was found to catalyze epoxidation, aziridination and amidation of hydrocarbons with good substrate conversions and high product yields. The iron(IV)-oxo and -imido reactive intermediates were generated in situ and their formulations were verified by mass spectroscopy. By Evan's method, the result supported the iron(IV) formulation with a low-spin d electronic configuration (S = 1). A ruthenium-oxo oxalato cluster formulated as Na [Ru ( -O) (C O ) ] (3) 7 4 3 4 2 4 6 was prepared. Structural studies and magnetic measurements revealed that the ruthenium atoms adopted the oxidation states of +III, +III, +III and +IV. Complex 3 showed anti-HIV properties on infected cells. Using the HIV-1(BaL) virus infected Hut/CCR5 cells, 3 was found to exhibit > 90% inhibition on viral replication at 5 M concentration. Similar findings were obtained with other HIV-infected cell lines including GHOST/CXCR4 and PBMCs. Complex 3 was non-cytotoxic to these cells even at 50 M concentration. However, the reactivity studies on those nitride complexes are sparse in the literature. As a result, the reactivities of nitridoruthenium(VI) complexes toward nucleophilic, electrophilic and oxidizing agents were studied. [Ru^CNXL^Cl] [H2L1 = 2,6-bis(2,2-diphenyl-2-hydroxyethyl)pyridine] underwent nitrogen atom transfer reaction with PPI13, PPhMe2 (dimethylphenylphosphine) and dppe [l,2-bis(diphenylphosphino)ethylene] to give [RuIII(HL1)(PPh3)Cl2], [Ru^XPPhMe^Cl] and [RuIII(L1)(dppe)Cl], respectively. In the presence of py or Hpz, the reaction of [Ru^^XL^Cl] gave [RuIII(L1)(py)2Cl] and [RuIII(L1)(Hpz)3]Cl as respective products. An unsymmetrical u-nitrido dinuclear ruthenium complex, [Ru^L^ChCu-N^u^L'XCsHioN^l], was formed upon reaction of [Ru^CNXL^Cl] with 2,6-dimethylaniline. Excess (Me30)BF4 reacted with [Ru^CNXL^Cl] to give a dinuclear u-OH complex, [Ru2(N)2(L1)2(OH)]PF6. For the analogous reaction with [n-Bu4N][MVI(NXL)] [L = L2 and L3; M = Os and Ru; H4L = l,2-dichloro-4,5-bis(2-hydroxybenzamido)benzene, H4L = l,2-bis(2-hydroxybenzamido)benzene], protonation at the auxiliary ligand occurred to give [M^CNXHL)] as product. Oxidation of [n-Bu4N][OsVI(N)(L2)] by PhI(OAc)2 produced [n-Bu4N][OsVI(N)(L202)] in 10% yield. Based on X-ray crystal analysis, the coordinated phenoxy moiety was converted to a benzoquinone moiety while the Os=N group remained intact. Book excerpt: Since the discovery of nonlinear effects, both the theory and measurement techniques have been developed significantly, especially since the invention of the laser. However, structure-NLO property relationships for organometallics as well as their NLO mechanisms are far less explored than those of organic molecules and inorganic salts. The greater flexibility and exceptionally large NLO responses of organometallic compounds attracts chemists to this field. Modification of coordinated co-ligands in organometallic systems has influence on the NLO merit by introducing new electronic charge-transfer transitions, oxidation state and coordination sphere of the metal centers. In this work, the donor sets of the ruthenium complexes were modified from the most investigated (P-P)(P-P) to (N-N)(P-P) and (N-P)2 and a series of ruthenium complexes were synthesized and characterized. In Chapter 2, the study of the Ru(N-N)(P-P) complexes is detailed. Three bidentate diphosphine ligands (dppe, dppb and dppf) and one diimine ligand (tert-Bu-bpy) were selected for this study as the diphosphine and diimine ligands, respectively. Ruthenium halide and mono-alkynyl complexes were obtained successfully. Their optical, electrochemical and spectroelectrochemical properties were examined and are discussed. The formation of eta3- and eta1-butenynyl complexes was confirmed by single-crystal X-ray diffraction. Attempts towards bis-alkynyl complexes were made, but no conclusive evidence could be obtained to confirm the successful synthesis of this species. In Chapter 3, the focus of the work is the study of ruthenium complexes with a (N-P)2 donor set. Two iminophosphine ligands, 2-(diphenylphosphino)pyridine (PPh2py) and 8-(diphenylphosphino)quinoline (PPh2qn), were selected for this study. The cis-RuCl2(N-P) complexes and the corresponding dimers were synthesized and characterized. Their optical and electrochemical properties were measured and are discussed. In Chapter 4, the quadratic and cubic nonlinear optical properties of organometallic complexes were explored by hyper-Rayleigh scattering (HRS) and frequency-depentdent Z-scan techniques, respectively. The first hyperpolarizabilities of the ruthenium halide and mono-alkynyl complexes described in Chapter 2 were determined by HRS measurements. The second hyperpolarizabilities of some organometallic complexes synthesized by the Humphrey group and the collaborators were measured by the Z-scan technique.

Els aquo complexes mononuclears i dinuclears de ruteni amb lligands polipiridílics han estat estudiats a fons pel nostre grup de recerca i d'altres, convertint-se en catalitzadors útils per a l'oxidació d'aigua i per a l'epoxidació d'alquens. Aquesta tesi vol presentar i discutir els següents temes relacionat: 1.La immobilització de dos nous complexes moleculars del tipus "Ru-Hbpp" capaços d'oxidar aigua, sobre òxids metàl·lics com són el TiO2 i el SiO2 així com fent ús del polímer Nafion per tal d'assolir l'objectiu final de construir una cel·la foto-electroquímica. El comportament catalític d'aquests nous sistemes serà discutit i comparat tant amb les espècies homòlogues moleculars com amb d'altres sistemes similars. 2.La síntesi i caracterització d'un complex dinuclear de ruteni que conté un lligand pont amb disposició bis-facial. La seva reactivitat envers l'oxidació d'aigua i d'olefines i la discussió comparant amb sistemes ja descrits a la literatura també formen part d'aquesta secció. 3.En aquest capítol s'ha plantejat la síntesi d'una nova família de lligands híbrids tetradentats capaços de coordinar el ruteni tant per N com per C, camp en el que se n'han fet diversos avanços. Així mateix descrivim un nou complex dinuclear de ruteni contenint un d'aquests nous lligands. 4.La preparació i caracterització tant espectroscòpica com electroquímica d'una família de complexos que tenen com a fórmula general [RuII(PY4Im)(X)]n+ (X = Cl, n = 1 or X = H2O, n = 2), on PY4Im és el lligand pentadentat 1,3-bis(bis(2-piridil)metil)imidazol-2-ilidè.
