Book excerpt: 1,2-Dialkylhydrazines are cleaved at the N~ bond on reaction with oxotrichlorobis(triphenylphosphine)rhenium{V) to give alkylimido complexes of the type [Re(NR)C13(PPh3)2J. 1,2-Dibenzylhydrazine reacts anomalously to give a Re(IV) amido complex of the type Retention of the N-N bond occurs on reaction of I ,2-dioC'll::.oy111ydrazinc with oxotrichlorobis(triphenylphosphine)rhenium(V) in benzene-ethanol, and a novel chelated benzoylhydrazido complex is formed. Employment of benzene-acetone as reaction medium permits the isolation of new benzoylhydrazone complexes. The formation of chelate rings containing nitrogen is not restricted m,aroyl hydrazines, and dibenzamidc gives Cl complex analogous to the mown rhenium acetylacetonates. The chelated benzoylhydrazido complexes react rapidly with cyanides, pyridine, phosphines, halogens and carbon monoxide to give rhenium(III) benzoy.lazo complexes, containing the -N=NCOPh group. Prolonged treatment of the benzoylhydrazidc complex with ditertiary and certain monotertiary phosphincs gives a series of new rhenium(I) tctrakis(phosphine)dinitJ'ogcn complexes. The first fluorophosphine and phosphite dinitrogcn complexes have been prepared by this method. The complex [ReCl(N2)(PMe2Ph)4] has also been prepared from dinitr~gen gas by reduction of [RcC13(PMe2Pll)3] under a dinitrogen pressure of 80 atmospheres. 15N2 labelling has been --1 employed to confinn the assignment of strong infrared bands in 1900-2100cm region to v{N=N). Carbon .monoxide reacts with the chelated benzoylhydrazido or benzoy.l.a.co complexes to give bis{carbonyl)dinitrogen complexes. The N=N and C=O stretching vibrations are shown to be st.rongly coupled by 15NZ labelling, infrared bands in the 1900-2100cm- 1 and no assignments of region can be made.The rhenium(l) tetrakis(phosphine)dinitrogen complexes react with halogens and oxidising nletal salts to give the first series of paramagnetic dinitrogen complexes. Transition metal salts such as [MOCl4(Etp)2] react with [ReCl(N2)(P}.fe2Ph)4]to give dinitrogen bridged complexes with abnonnally low N=N stretching frequencies (1680-1850CJlt-1). Treatment of the bis(carbonyl)dinitrogen complexes with mono- and ditertiaryphosphincs provides a facile method for the preparation of rhenium(I) mono- and bis -carbonyl complexes.

Book excerpt: This dissertation investigates the synthesis and characterization of several rhenium Schiff base complexes as potential radiopharmaceuticals for imaging and therapeutic purposes. A series of acetylacetone derived tetradentate N 2 O 2 Schiff base ligands (i.e., acac 2 en, acac 2 pn, acac 2 dmpn) were reacted with a rhenium (V) starting material (i.e., n-Bu 4 N[ReOCl 4], ReOCl 3 (PPh 3) 2) to yield a series of compounds. In most cases, reaction's involving [n-Bu 4 N][ReOCl 4] with two equivalents of ligand in ethanol yielded monomer complexes of the form trans -[ReO(Ligand)X]. The type of axial substituent, X = Cl -, H 2 O, ReO 4 -, MeO -, found trans to the rhenium oxo group can be adjusted by altering the reaction conditions. A similar reaction utilizing ReOCl 3 (PPh 3) 2, one equivalent of the ligand, and triethylamine yielded [mu]-oxo dimer complexes, trans -[Re 2 O 3 (Ligand) 2]. The conversion of rhenium (V) Schiff base monomer complexes into [mu]-oxo dimer complexes was investigated to gain a better understanding of conditions that yield monomer compounds. Experiments reacting trans -[ReO(acac 2 pn)Cl] with various solution conditions led to the proposal of a dimer formation mechanism that involves two pathways: (1) addition of base(OH - ) and (2) addition of water to generate a trans aquo species and additional water molecules to deprotonate the coordinated water molecule. The rhenium (V) monomer complexes, trans -[ReO(acac 2 en)OH 2]Cl and trans -[ReO(acac 2 pn)Cl], were also reacted with phosphine ligands to potentially generate a bis -phosphine(Schiff base)rhenium(III) cationic complexes, trans -[Re(Ligand)(PR 3) 2] + . The trans -[ReO(acac 2 en)H 2 O]Cl complex yielded the Re (III) complex when reacted with phosphines, while the Schiff base ligand of the [trans -ReO(acac 2 pn)]Cl complex underwent an intramolecular rearrangement to yield an asymmetric complex, [cis -ReO(acac 2 pn)PR 3]X. A mechanism is proposed for the rearrangement of the Schiff base ligand in the complex upon treatment with phosphine ligands. Further experiments with other soft monodentate ligands (i.e., thiocyanate, cyanide) yielded neutral asymmetric complexes. A series of experiments investigated the removal radioactive Tc-99 from solution through precipitation. The study focused on precipitating agents that could be added to a simulated waste stream at a uranium reprocessing plant to remove Tc-99 as a pertechnetate ion pair or a reduced Tc (IV) complex.

Rhenium has been widely used as a surrogate of technetium for a long time, and the promising physical features of 186Re and 188Re, make 186/188Re-complexes promising candidates as therapeutic radiopharmaceuticals.Similarly, the interesting photoactive and photoluminescence properties of non-radioactive Re-complexes make them excellent catalysts, luminescent materials and imaging sensors.Thus, in this work, our goal was to (i) develop, using a click chemistry strategy, multidentate ligands for the stabilization of different rhenium cores [Re(CO)3]+ and [ReO]3+ (M = Re or 188Re) as well as the analogous 99mTc-cores in some examples, (ii) assess the potential of the rhenium(technetium) complexes as imaging (natRe or 99mTc) or therapeutic (188Re) agents. To do so, two rhenium(technetium) specific-chelating systems were used: a semi-rigid tripodal system in the second chapter and a pyta moiety in the third chapter, these two chelators being developed previously in our group. Thus, based on a N2O tridentate click ligand, two different studies were carried out in chapter II. In the first one, two synthetic pathways to a range of potentially N3O tetradentate ligands, designed to coordinate rhenium cores as well as their coordination behaviors towards different rhenium cores (oxidation states +I and +V), were investigated. The first radiolabeling results combined with the recent work reported by Dugave and co-workers indicated that this ligand could be a promising 99mTc-chelator for nuclear imaging applications. As perspectives to this work, the extension of the radiolabelling work using the [188ReVO]3+ core should be performed, and the in vitro stability should be tested under physiological conditions in human plasma and by cysteine exchange experiments. The second study was focused on the development of novel hypoxia-selective 99mTc radiopharmaceuticals. Our semi-rigid tripodal click framework was decorated with an appended nitro group (either a nitrobenzyl group or a metronidazole (Mtz) unit). Different positions were considered and at least only two metronidazole (Mtz)-containing ligands and one nitro group-containing ligand as well as their corresponding tricarbonyl rhenium(I) complexes were obtained and characterized, in particular by electrochemistry. The reduction potentials of NO2 group in complexes [Re(CO)3Cl(L2)] and [Re(CO)3(L6)] were similar to those of reported hypoxic imaging agents, prompting us to further investigate other properties of these complexes. Chapter III was focused on the study of AIE (aggregation-induced emission) effect in tricarbonyl Re(I) complexes, the association of this effect with the intrinsic properties of Re(I) complexes being expected to lead to very attractive compounds. To do that, we combined an organic fluorophore (PBO) which exhibits excellent stability and optical properties, with a tricarbonylrhenium(I) complex based on a pyta unit (either a 2-pyridyl-1,2,3-triazole or a 2-pyridyl-1,2,4-triazole ligands). Four compounds were studied. The X-Ray structures revealed spectacular discrepancies between the two first triazole-based complexes ReL8 and ReL9. Moreover, this study being a novel orientation in our group, this work is a great starting point for further investigations. Various organic dyes and/or structural modifications of the organic moiety will soon be considered in order to develop highly emissive rhenium(I) luminescent probes.

Chapter 2 describes the rhenium labeling of four somatostatin analogues. Confirmation for the formation of nonradioactive rhenium labeled peptides was accomplished with LC-MS. Purification was done by analytical HPLC. Chapter 3 describes the synthesis, characterization and determination of acid dissociation constant values for the two rhenium complexes with ethylenediamine type ligands. This chapter also describes the synthesis and characterization of a rhenium amine complex with the 2-aminoethanethiolate ligand and a phosphine ligand.
