Book excerpt: Although retroviruses have long been associated with a variety of animal diseases, active research in the field of human retroviruses dates from the discovery of human immunodefici ency virus (HIV) in association with acquired immunodefici ency syndrome (AIDS). The enormous research efforts in this field have been directed toward understanding the nature of the virus and toward its elimination through preventive vaccin ation and the cure of the disease. Human T-cell leukemia virus (HTLV-l) was the first member of the human retrovirus family to be discovered. It was implicated as the cause of adult T-cell leukemia (ATL) even before the association of HIV and AIDS was established. Research on HTL V -1 has, however, been lagging behind that of HIV because of the importance of AIDS. Today HTLV-1 and possibly closely related HTLV-2 are associated with a variety of human neurologic diseases, and research activities in this field may show that human retroviruses can cause a variety of human diseases in addition to those affecting the nervous system. Papers in this volume attempt to acquaint the reader with the present state of research into retrovirus infection and related diseases of the nervous system.

Book excerpt: Retroviruses arguably belong to the most fascinating of all viruses because of their unusual and highly efficient mode of replication involving reverse transcription and integration of the viral genome and a complex system of transcriptional and post transcriptional regulatory mechanisms. The importance of ret roviruses as human and animal pathogens has also enhanced scientific and medical interest in this diverse group of viruses and has spurred an intensive search for novel and improved antiviral agents. More recently, analysis of retroviral replication and in particular understanding the formation and composition of the virus particle has received additional attention because of the promise of retroviral vectors as vehicles for human somatic gene therapy. Many recent advances have been made in our understanding of the molecular mechanisms governing as sembly and release of infectious retrovirus particles. This book attempts to summarize these recent developments and to provide an overview of our current knowledge on retrovirus particle formation. The individual chapters of the book deal with specific steps in the pathway of retroviral morphogenesis and maturation, starting at the time when the components of the virus have been synthesized within the infected cell and ending once the infectious virion has been released from the cell. An introductory chapter provides a comparative description of the structure and morphology of various retroviruses.

Book excerpt: An integrated retrovirus effectively becomes part of the cellular genome, but with the difference that the virus to a large extent retains control over its own expression through nontranslated sequences in the long terminal repeat (L TR). Some retroviruses also code for nonstructural proteins that further regulate proviral expression. Integration changes the cell genome; it adds viral genes, and in the case of transducing retroviruses also adds cell-derived oncogenes that have been incorporated into the viral genome. Integration can also have consequences for cellular genes. The transcriptional signals in a provirus can activate expression of neighboring cellular genes; the integration even can disrupt and thus inactivate cellular genes. These effects of retroviral genomes take place in cis; they are referred to as insertional mutagenesis and are the subject of this volume. Almost 10 years have passed since W. Hayward, S. Astrin, and their colleagues found that in B cell lymphomas of chickens, induced by avian leukosis virus, transcription of the cellular proto-oncogene myc was upregulated through the integration of a complete or partial provirus in its vicinity. This landmark discovery suggested a mechanism by which retro viruses that do not carry cellular oncogenes in their genome ("nonacute retroviruses") can cause cancer. It contributed the first evidence for the carcinogen potential of oncogenes that are not part of a viral genome.

For the first time, it was demonstrated that a retrovirus could play a role in the development of a human disease, in this case adult T cell leukemia (ATL). Several years later, the acquired immunodeficiency syndrome (AIDS) epidemic began, and it was dem- strated that a retrovirus, originally designated the human T cell lymp- tropic virus type 3, was the causal agent of this syndrome. This virus, later named the human immunodeficiency virus type 1 (HIV-1), has since been extensively studied in terms of its pathogenesis as well as its ability to elicit immune responses. In that time, a tremendous amount of information has been obtained about the virus. Although recent drug regimens have been useful in significantly lowering viral loads and perhaps maintaining an asymptomatic state among individuals infected with HIV-1, an established “cure” for AIDS eludes us. In addition, the effective drug therapies are very expensive, and are not available to infected people in the third world, where greater than 90% of new infections occur. Furthermore, the development of viral resistance against the drug therapies is an additional concern. Despite extensive study, no effective vaccine has been developed. One of the problems in developing an effective vaccine against HIV-1 is the ability of the virus, particularly in the immunogenic envelop glycoprotein, to undergo amino acid hypervariability.

Such a procedure seemed the more appropriate as the workshop was intended to bring together specialists from two rather diverse fields: RNA-tumor virology and immunobiology. While this created some initial problems of terminology, it was quite effective in making representatives of one field more aware of the significance and the contributions ofthe other. Quite a few of these viruses have been characterized in the past few years and their varied nomenclature according to source, discoverer, date of isolation or biological properties could result in some con fusion among those attempting to follow the literature. Information will be included on the molecular biology of the sarcoma viruses, rather more of which is available than when the review was fITst conceived, although in this respect the review will inevitably be out of date by the time of publication. If any bias of content is introduced, this will be towards a more detailed coverage of the author's own area of interest, the gene products of the defective sarcoma viruses. Rous sarcoma virus (RSV) serves as the model for much of this work and will frequently be referred to for comparative purposes, as will the mammalian defective transforming viruses. Recent reviews provide more complete coverage of these topics (l-4a). This review is complemented by a discussion ofthe avian acute leukaemia viruses, which appears elsewhere in this volume [5]. As will be seen, concentrating primarily on the defective sarcoma viruses and comparing them to RSV can be justified in terms of their biochemical properties as well as their similar biology.
