Book The Smallest Biomolecules Diatomics and their Interactions with Heme Proteins

Download or read book The Smallest Biomolecules Diatomics and their Interactions with Heme Proteins written by Abhik Ghosh and published by Elsevier. This book was released on 2011-10-13 with total page 627 pages. Available in PDF, EPUB and Kindle. Book excerpt: This is not a book on NO biology, nor about hemoglobin, nor about heme-based sensors per se. Of course, it covers all these topics and more, but above all, it aims at providing a truly multidisciplinary perspective of heme-diatomic interactions. The overarching goal is to build bridges among disciplines, to bring about a meeting of minds. The contributors to this book hail from diverse university departments and disciplines – chemistry, biochemistry, molecular biology, microbiology, zoology, physics, medicine and surgery, bringing with them very different views of heme-diatomic interactions. The hope is that the juxtaposition of this diversity will lead to increased exchanges of ideas, approaches, and techniques across traditional disciplinary boundaries. The authors represent a veritable Who’s Who of heme protein research and include John Olson, Tom Spiro, Walter Zumft, F. Ann Walker, Teizo Kitagawa, W. Robert Scheidt, Pat Farmer, Marie-Alda Gilles-Gonzalez, and many other equally distinguished scientists. Extremely distinguished list of authors Multidisciplinary character – equally suitable for chemists and biochemists Covers the hottest topics in heme protein research: sensors, NO biology, new roles of hemoglobin, etc.

Available in PDF, EPUB and Kindle. Book excerpt: This authoritative Fourth Edition summarizes the advances of the past decade concerning the structure, mechanism, and biochemistry of cytochrome P450 enzymes, with sufficient coverage of earlier work to make each chapter a comprehensive review of the field. Thirteen chapters are divided into two detailed volumes, the first covering the fundamentals of cytochrome P450 biochemistry, as well as the microbial, plant, and insect systems, and the second exclusively focusing on mammalian systems. Volume 1 begins with an exploration of the biophysics and mechanistic enzymology of cytochrome P450 enzymes, with a discussion of the structures of P450 enzymes and their electron donor partners, the mechanisms of oxygen activation and substrate oxidation, and the approaches and nature of cytochrome P450 inhibition. Two more chapters discuss the nature and roles of cytochrome P450 enzymes in microbes, plants and insects, and an eighth chapter is a survey of the potential utility of P450 enzymes in biotechnology. The first chapter of Volume 2 examines the roles of P450 enzymes in mammals, mainly humans. Four further chapters then deal with the genetic and hormonal regulation of P450 enzymes and their specific roles in the processing of sterols and lipids. Cytochrome P450: Structure, Mechanism, and Biochemistry is a key resource for scientists, professors, and students interested in fields as diverse as biochemistry, chemistry, biophysics, molecular biology, pharmacology and toxicology.

Book Resonance Raman Studies of Isotopically Labeled Heme Proteins

Download or read book Resonance Raman Studies of Isotopically Labeled Heme Proteins written by Freeborn Rwere and published by . This book was released on 2009 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: One effective approach for exploring structure/function relationships in heme proteins is to study proteins that have been reconstituted with modified hemes so as to systematically perturb the protein-heme interface. However, some reconstituted heme proteins may contain substantial fractions of a "non native" state in which the orientation of the heme in the folded pocket differs from the native conformation by a 180° rotation about the [alph alpha - gamma gamma] meso axis. In fact, this "non native" state has also been shown to exist in some native proteins, including several mammalian globins. In order to define changes in the active site structure associated with this "disorder", we have applied resonance Raman spectroscopy to the metMb derivatives, using selectively deuterated protohemes to associate the observed modes with specific fragments of the heme. Resonance Raman spectroscopy is also employed to characterize heme site structural changes arising from conformational heterogeneity in deoxyMb and ligated derivatives; i.e., the ferrous CO (MbCO) and ferric cyanide (MbCN) complexes. Interestingly, while substantial changes in the disposition of the peripheral vinyl and propionate groups can be inferred from the dramatic spectral shifts, the bonds to the internal histidyl imidazole ligand and those of the Fe-CO and Fe-CN fragments are not significantly affected by the heme rotation, as judged by lack of significant shifts in the [upsilon](Fe-NHis), [upsilon](Fe-C) and [upsilon](C-O) modes. We have synthesized protohemes with selectively labeled vinyl groups and have effectively reconstituted them into apo-myoglobin in order to assign the so-called "vinyl bending" modes of heme group in native and reversed forms of myoglobin to their specific molecular fragments based on their isotopic shift with these vinyl labeled protohemes. In a separate project, these vinyl labeled hemes have been employed to further define structural changes in cytochrome P450cam upon substrate binding. Substrate binding to cytochrome P450cam is known to induce the distortions of the out of plane modes such as [gamma gamma]6 and [gamma gamma]7 modes as well as the heme peripheral substituents. The detection of these low frequency modes is especially important as the disposition of these groups can modify the heme reduction potential.

Download or read book Handbook Of Porphyrin Science With Applications To Chemistry Physics Materials Science Engineering Biology And Medicine Volumes 1 5 written by Karl M Kadish and published by World Scientific. This book was released on 2010-03-16 with total page 2438 pages. Available in PDF, EPUB and Kindle. Book excerpt: This is the first set of Handbook of Porphyrin Science.Porphyrins, phthalocyanines and their numerous analogues and derivatives are materials of tremendous importance in chemistry, materials science, physics, biology and medicine. They are the red color in blood (heme) and the green in leaves (chlorophyll); they are also excellent ligands that can coordinate with almost every metal in the Periodic Table. Grounded in natural systems, porphyrins are incredibly versatile and can be modified in many ways; each new modification yields derivatives demonstrated new chemistry, physics and biology, with a vast array of medicinal and technical applications. As porphyrins are currently employed as platforms for study of theoretical principles and applications in a wide variety of fields, the Handbook of Porphyrin Science represents a timely ongoing series dealing in detail with the synthesis, chemistry, physicochemical and medical properties and applications of polypyrrole macrocycles. Professors Karl Kadish, Kevin Smith and Roger Guilard are internationally recognized experts in the research field of porphyrins, each having his own separate area of expertise in the field. Between them, they have published over 1500 peer-reviewed papers and edited more than three dozen books on diverse topics of porphyrins and phthalocyanines. In assembling the new volumes of this unique Handbook, they have selected and attracted the very best scientists in each sub-discipline as contributing authors of the chaptersThis Handbook will prove to be a modern authoritative treatise on the subject as it is a collection of up-to-date works by world-renowned experts in the field. Complete with hundreds of figures, tables and structural formulas, and thousands of literature citations, all researchers and graduate students in this field will find the Handbook of Porphyrin Science an essential, major reference source for many years to come.
