Book excerpt: With the continued rapid expansion of neurochemical research, there has been no shortage of new developments in methodology for this third volume of Research Methods in Neurochemistry. As in previous volumes we have again tried to provide some balance in the subjects represented. The wisdom of this policy may be questioned since it can lead to delay in publica tion, but there are many approaches to the chemical study of the nervous system and a methods book needs to stand on its own as well as be part of a series. In one respect, however, the present volume departs from this policy, in that we have included two chapters on micromethods for analyzing amines and amino acids, both giving special emphasis to dansylation techniques. These chapters are complementary and we feel justified in publishing them in one volume in view of the importance of such micromethods for the study of neural systems. At the other end of the scale, particular attention may be drawn to the chapter by D. D. Gilboe and colleagues describing their remarkable procedures for studying metabolism in the isolated canine brain. We were fortunate also in persuading S. S. Oja to extend the general prin ciples of transport systems he described in Volume 2 to amino acids in brain slices. In addition, there are the usual chapters on components of neural tissues, which once again we have found convenient to divide into enzymes, macromolecules, and other constituents.

Book excerpt: On picking up this first volume of a new series of books the reader may ask the two questions: (a) why research methods? and (b) why in neurochem istry? The answers to these questions are easy - they more than justify the volumes to come and show the strong need for their existence. It is customary to think of methods as a necessary but unexciting means to an end - to relegate advances in methodology to a minor role in the creative, original portion of advances in science. This is not the case; the pace-setting function of methodology is well illustrated in most areas of neurobiology. To formulate our questions to Nature (which is the essence of experimental design), methodology is needed; to get answers to our ques tions we have to devise yet new methods. The chapters of the present volume fully illustrate how the development of a new method can cut a new path how it can open new fields, just as the microscope founded histology. Heter ogeneity of structures presents a formidable challenge for methodology in the nervous system, yet methods for separating the structures are essential if we ever want to decipher the enigma of functional contribution of the ele ments to the whole. The problem is not only physical separation-clearly methods are essential to study complex structures in situ.

Book excerpt: Neurochemistry, having the objective of elucidating biochemical processes subserving nervous activity, emerged as an application of chemistry to the of neurobiological problems as a post-World War II phenomenon. investigation However, only in the last 40 years has the chemical community recognized neurochemistry as a distinct, if hybrid, discipline. During this period great strides have been made. However, recently neurochemistry, along with neu rophysiology, neuropharmacology, neuroanatomy, and the behavioral sci ences, has emerged to form neuroscience, a new community of scientists with its own national society, journals, and meetings. Actually, this recently formed hybrid, neuroscience, is in the process of merging with another well-established discipline, molecular genetics (frequently called molecular biology, and itself a hybrid), which appears to have sufficient hybrid vigor to form yet a new community of scientists, which, for want of a more imaginative term, has been called molecular genetic neuroscience. Clearly, advantages resulting from such mergers or hybridizations accrue not only from the merging discipline (neurochemistry in this case) to the new community (molecular genetic neuroscience), but also in the reverse direction. This Foreword will be concerned primarily with examples of this latter process.

They are the results of many millions of years of evolution, from the time when biological membranes first came into being and were folded to produce the first cells within which the earliest enzymes were wrought. Countless changes have taken place within them, so that, now, only those enzymes exist that play specific roles in the functions of the living cells of today. Those in the nervous system possess a mUltiple role: in the creation, maintenance, and ultimate breakdown of the component cells and in enabling consciousness, perception, memory, and thought to become possible. But though life may go on forever, the enzymes that make life possible will undergo the many changes involved in the evolutionary process.

The proceedings were edited by S. Berl, D. D. Clarke and D. Schneider and published as Volume 6 of the NATO ASI Life Science series by Plenum Press. The object of the present meeting was to review and discuss the present status of amino acids as chemical transmitters. Several issues such as electrophysiological response, localization, synthe sis, release and receptor binding of transmitter candidates were discussed. The possible morphological correlates to these func tions were also reviewed. During the meeting 24 leading papers were given. In addition, several of the participants presented important new findings during the discussion. Some of these have been included as short reports. The main financial support was obtained from NATO, Scientific Affairs Division.

Book excerpt: The division of the mature mammalian brain and spinal cord into regions of "white" matter and "gray" matter is observable upon the most cursory inspection. Microscopic examination indicates that the "white" matter regions are characterized by a multilamellar sheath (myelin) which sur rounds the axons. A morphologically similar myelin imparts the white color to tracts of the peripheral nervous system, although, as is empha sized throughout the book, there are very significant morphological and chemical differences between central and peripheral myelin. The rapid postnatal accumulation of myelin is temporally associated with increasing functional capacity and presumably indicates some important role related to neuronal activity. The ability of myelinated axons to conduct electrical impulses much more rapidly and with much lower use of energy, relative to unmyelinated axons, is now generally accepted. As is evident from the above capsule description, a considerable body of information concerning myelin is available. However, as in other areas of research the unknown expands before us more rapidly than we are able to add to our core of knowledge. This is a corollary of the axiom that a good experiment raises more questions than it answers. Among the areas currendy under investigation in many laboratories are the following: interaction of myelin-forming cells with neurons, assembly and metabo lism of myelin, molecular architecture of myelin, and the details of the functional role of myelin.

The concept was developed in the laboratory of Dr. Heinrich Waelsch as a result of studies related to the metabolism of glutamic acid and glutamine. It was welcomed by a number of neurochemists who felt that the concept had validity and would help explain metabolic phenomena that were otherwise quite puzzling. The concept gradually achieved general acceptance, and by 1971 the amount of information that could profit by being examined from the point of view of metabolic compartmentation had increased sufficiently to warrant a symposium on the subject ("Metabolic Compartmentation in the Brain," Balazs and Cremer, eds. , MacMillan, 1973). Almost all the participants at that initial symposium were present at the second, on which this volume is based.

Neuroethology, which is concerned with the experimental analysis ofthe releasing and control mech anisms of behavior, is a young discipline. Results from this multidisciplinary branch of research, which uses physical, chemical, and mathematical methods, have not yet been extensively treated in textbooks of neurophysiology and ethology. This book is intended as a first attempt to pose major questions of neuroethology and to demonstrate, by means of selected research examples, some of the ways by which these questions are being approached. Inevitably this cannot be a complete and in depth detailed treatment of all of the neurobiology examples, and I realize that such a selection is of a subjective nature. The overall goal ofthe book is to present an introduction. After outlining some of the very basic neurophysiological and ethological concepts (Chaps. 2 and 3), neuroethological questions and methods are demonstrated extensively by means of a particular example (Chap. 4). There are two reasons to choose the visually guided prey-catching and avoidance behavior of the Common Toad: (1) it is a system which I have inves tigated for about fifteen years and therefore know best, (2) the toad story is one of the most comprehensive neuro ethological approaches so far. Thus, it is possible here to outline the major concepts of neuroethology and to pose the basic questions.
