Book excerpt: In response to cytoskeletal perturbations Saccharomyces cerevisiae cells trigger a morphogenesis checkpoint resulting in a G2 arrest. The arrest is due to the Swe1p kinase, which phosphorylates and inhibits the CDK Cdc28p. Dephosphorylation of Cdc28p is catalyzed by the phosphatase Mih1p, and allows for cell cycle progression. Recent work on regulation of the kinase Swe1p has shown a complex pathway of localization specific degradation. Several of the components involved in Swe1p degradation appear to have conserved roles in degradation of the Wee1 family kinases in multicellular systems. Despite this little is known about the steps leading up to the initiation of Swe1p degradation. In this study we have challenged the model that activation of the Swe1p negative regulator, Hsl1p, results from relief of autoinhibition due to septin interaction. Rather, we conclude Hsl1p is subject to phosphorylation of threonine 273, promoted by the septin-associated kinase Elm1p. We identified elm1 mutants in a screen for defects in Swe1p degradation. Additionaly regulation of the morphogenesis checkpoint appears to be acting though regulation of the phophatase Mih1p. In other eukaryotes, Mih1p homologs (Cdc25-family phosphatases) are thought to be regulated by other checkpoints through control of their phosphorylation. We have shown that Mih1p is phosphorylated in response to morphogenesis checkpoint stress. We have also identified a NLS in Mih1p, mutation of which greatly reduces its in vivo activity. These data combined with data from a competing lab suggest that phosphorylation of Mih1p is indeed regulated. We have also investigated the checkpoint response of budded yeast cells. Specifically we have shown that environmental stresses that depolarize the actin cytoskeleton result in a Swe1p mediated G2 delay.

Book excerpt: The morphogenesis checkpoint in the budding yeast Saccharomyces cerevisiae monitors bud emergence. If environmental perturbations delay budding, the cells arrest in G2 until they are able to form a bud. The morphogenesis checkpoint is mediated by Swe1p, the budding yeast homolog of Wee1, which phosphorylates and inhibits Cdc28p, the CDK that drives cell division. In unstressed cells, Swe1p is degraded, a process that is dependent on the kinase Hsl1p. Hsl1p localizes to the septins, which form a collar-like structure at the bud neck. Hsl1p binds to Hsl7p, an adapter protein that recruits Swe1p to the neck. When budding is perturbed, Hsl1p is inactive, thus allowing Swe1p to accumulate and inhibit Cdc28p. Bud formation activates Hsl1p, enabling it to recruit negative regulators of Swe1p to the bud neck, where Swe1p is marked for degradation. Interestingly, our lab has shown that Hsl1p is regulated by cell shape. Here, we reexamine a model that suggests Hsl1p is regulated by an autoinhibitory domain and discover that the model cannot be supported by our data. In addition, we present evidence suggesting that Hsl1p is regulated by an upstream kinase, Elm1p, that phosphorylates Hsl1p within the activation loop. Finally, we identify functional and regulatory regions within Hsl1p. We identify regions that participate in septin binding and Hsl7p binding. But our most interesting discovery is a regulatory region that makes Hsl1p responsive to bud emergence.
