Book excerpt: A NATO Advanced Study Institute on "New Developments and Methods in Membrane Research and Biological Energy Transduction" was held in order to consider some of the most recent developments in membrane research methodologies and results, with particular emphasis on studies of biological energy transduction. The partic ipants in the Institute dealt with three general areas of membrane study: membrane structure (with emphasis on lipid and protein components), membrane component assembly (with particular emphasis on mitochondria and chloroplasts), and the specialized functions of certain membrane systems. This last area included discussions of topics such as drug transformation, the role of membrane electron transport in the generation of oxygen radicals, the effect of oxygen radicals on cellular homeostasis and on the structure, organization and function of the acetylcholine receptor. Lectures and posters were concerned with two central questions: what is the function of membrane structure in energy transduction and how can energy trans duction be effectively measured and assessed? This text presents the content of the major lectures and important posters presented during the Institute's program. In issuing this book, the editor hopes to convey the proceedings of the Institute to a larger audi ence and to offer a comprehensive account of those developments in membrane research that were considered on the Island of Spetsai between August 16 and August 29, 1984. It describes a novel approach for how these advances might be used in combination with existing methodologies to further the understanding of mechanisms of developmental toxicity, to improve the assessment of chemicals for their ability to cause developmental toxicity, and to improve risk assessment for developmental defects. For example, based on the recent advances, even the smallest, simplest laboratory animals such as the fruit fly, roundworm, and zebrafish might be able to serve as developmental toxicological models for human biological systems. Use of such organisms might allow for rapid and inexpensive testing of large numbers of chemicals for their potential to cause developmental toxicity; presently, there are little or no developmental toxicity data available for the majority of natural and manufactured chemicals in use. This new approach to developmental toxicology and risk assessment will require simultaneous research on several fronts by experts from multiple scientific disciplines, including developmental toxicologists, developmental biologists, geneticists, epidemiologists, and biostatisticians.

Less attention, however, has been paid to the mesoscale-it is at this scale, in the range extending from large molecules (10 nm) through viruses to eukaryotic cells (10 microns), where interesting ensemble effects and the functionality that is critical to macroscopic phenomenon begins to manifest itself and cannot be described by laws on the scale of atoms and molecules alone. To further explore how knowledge about mesoscale phenomena can impact chemical research and development activities and vice versa, the Chemical Sciences Roundtable of the National Research Council convened a workshop on mesoscale chemistry in November 2014. With a focus on the research on chemical phenomena at the mesoscale, participants examined the opportunities that utilizing those behaviors can have for developing new catalysts, adding new functionality to materials, and increasing our understanding of biological and interfacial systems. The workshop also highlighted some of the challenges for analysis and description of mesoscale structures. This report summarizes the presentations and discussion of the workshop.

Water confers to lipid membranes unique features in terms of surface and mechanical properties. The analysis of the hydration forces, plasticiser effects, controlled hydration, formation of microdomains of confined water suggests that water is an active constituent in a water-lipid system. The chapters describe water organization at the lipid membrane–water interphase, the water penetration, the long range water structure in the presence of lipid membranes by means of X-ray and neutron scattering, general polarization, fluorescent probes, ATR-FTIR and near infrared spectroscopies, piezo electric methods, computer simulation and surface thermodynamics. Permeation, percolation, osmotic stress, polarization, protrusion, sorption, hydrophobicity, density fluctuations are treated in detail in self-assembled bilayers. Studies in lipid monolayers show the correlation of surface pressure with water activity and its role in peptide and enzyme interactions. The book concludes with a discussion on anhydrobiosis and the effect of water replacement in microdomains and its consequence for cell function. New definitions of lipid/water interphases consider water not only as a structural-making solvent but as a mediator in signalling metabolic activity, modulating protein insertion and enzymatic activity, triggering oscillatory reactions and functioning of membrane bound receptors. Since these effects occur at the molecular level, membrane hydration appears fundamental to understand the behavior of nano systems and confined environments mimicking biological systems. These insights in structural, thermodynamical and mechanical water properties give a base for new paradigms in membrane structure and function for those interested in biophysics, physical chemistry, biology, bio and nano medicine, biochemistry, biotechnology and nano sciences searching for biotechnological inputs in human health, food industry, plant growing and energy conversion.

In April 1997, many of the authors met for a two-day meeting devoted entirely to intracellular cholesterol trafficking. The rationale for this meeting was that investigators interested in this topic worked in a variety of fields, and rarely, if ever, all met together. Everybody knew each other's papers but mostly worked in isolation from one another. Understanding of cholesterol trafficking also appeared to have reached the point where it would start to rapidly expand beyond these few laboratories. Understanding of cholesterol trafficking was moving from a largely descriptive science into the molecular age. It seemed a good time to get together and see how much we agreed upon up to this point. More authors contributed to this volume than attended the St. Louis meeting. That meeting was generously funded by grants from Bristol-Myers Squibb, Merck and Company and Parke-Davis, however, the total funding available limited the size of the meeting. For the book, we are not so limited and have tried to be as inclusive as possible and pretty much invited everyone who is presently active in this area. We were quite fortunate to successfully recruit the authors we sought for each of these chapters. The authors and their contributions can be organized by particular interests and particular areas of expertise.

Book excerpt: This book addresses the possibilities and challenges in mimicking biological membranes and creating membrane-based sensor and separation devices. Recent advances in developing biomimetic membranes for technological applications will be presented with focus on the use of integral membrane protein mediated transport for sensing and separation. It describes the fundamentals of biosensing as well as separation and shows how the two processes are working in a cooperative manner in biological systems. Biomimetics is a truly cross-disciplinary approach and this is exemplified using the process of forward osmosis will be presented as an illustration of how advances in membrane technology may be directly stimulated by an increased understanding of biological membrane transport. In the development of a biomimetic sensor/separation technology, both channels (ion and water channels) and carriers (transporters) are important. An ideal sensor/separation device requires the supporting biomimetic matrix to be virtually impermeable to anything but the solute in question. In practice, however, a biomimetic support matrix will generally have finite permeabilities to water, electrolytes, and non-electrolytes. These non-protein mediated membrane transport contributions will be presented and the implications for biomimetic device construction will be discussed. New developments in our understanding of the reciprocal coupling between the material properties of the biomimetic matrix and the embedded proteins will be presented and strategies for inducing biomimetic matrix stability will be discussed. Once reconstituted in its final host biomimetic matrix the protein stability also needs to be maintained and controlled. Beta-barrel proteins exemplified by the E. Coli outer membrane channels or small peptides are inherently more stable than alpha-helical bundle proteins which may require additional stabilizing modifications. The challenges associated with insertion and stabilization of alpha-helical bundle proteins including many carriers and ligand and voltage gated ion (and water) channels will be discussed and exemplified using the aquaporin protein. Many biomimetic membrane applications require that the final device can be used in the macroscopic realm. Thus a biomimetic separation device must have the ability to process hundred of liters of permeate in hours – effectively demanding square-meter size membranes. Scalability is a general issue for all nano-inspired technology developments and will be addressed here in the context biomimetic membrane array fabrication. Finally a robust working biomimetic device based on membrane transport must be encapsulated and protected yet allowing massive transport though the encapsulation material. This challenge will be discussed using microfluidic design strategies as examples of how to use microfluidic systems to create and encapsulate biomimetic membranes. The book provides an overview of what is known in the field, where additional research is needed, and where the field is heading.
