KIMBALL BACKGROUND It was observed in 1930 that seminal plasma of a number of species contained very large amounts of substance(s) capable of altering uterine motility. These substances were subsequently demonstrated to be prostaglandins (PGs); this began the early and continued association of this group of compounds with reproduction. The activity was found to be associated with a fraction containing lipid-soluble acids, derived from prostanoic acid. Restrictions imposed by obscure sources and small amounts of material available for testing were initially impediments to their study.

Book excerpt: Prostagtandin Abstracts Volume II contains abstracts of the significant scientific Iiterature published in 1971, 1972, and 1973 on the role of prostaglandins in human reproduction. Of the more than 4000 articles which were published between 1971 and 1973 and deal with prostaglandins, endogenous and exogenous, in mal es and females, nearly 1100 involve human reproduction. Prostagtandin Abstracts, Volume I (Plenum Publishers, 1973) contains abstracts of all articles on prostaglandins published between 1906, when the action of these compounds was first noted, and 1970, when clinical research with these compounds was weil underway. Clinical use of prostaglandins during the 1960s proved safe but provided evidence of troublesome side effects. Du ring the 1970s, the development of new analogues, various protocols for international testing, and improved routes of administration indicates that prostaglandins still hold promise as a useful drug in the management of fertility. Research into potential contraceptive and other clinical uses of prostaglandins continues to be an exciting and expanding field of fundamental and clinical research. The citations and abstracts which are included in this volume have been selected and reproduced from POPINFORM, a computerized on-line information retrieval system which includes in its data base more than 25,000 documents on regulation of fertility. Also included as an Appendix is a bibliography (without abstracts) of 1971 -1973 material on aspects of prostaglandin research that do not involve reproduction. This bibliography was prepared by the Upjohn Company of Kalamazoo, Michigan and is printed here with permission.

During the last decade obstetricians have become familiar with these drugs for inducing labour and for terminating second-trimester pregnancy. Within the last year their use has been extended, in association with antiprogestin, to first-trimester termi nation. Although the effectiveness of prostaglandins in these pharma cological roles is clear, their full potential has still to be explored, and much remains to be learned about their physiology. Prostaglandins playa central role in the initiation of labour. Further clarification of this role could lead to improvements in preventing or treating preterm labour, which still remains the most important cause of perinatal mortality in this country. Prostaglandins are also inti mately involved in the mechanism of menstruation. More detailed understanding of this role should lead to more effective treatments for menorrhagia and dysmenorrhoea. These substances also playa part in the mechanism of implantation, and further research in this area may lead to more effective therapy for infertility.

The fields of molecular genetics and immunology have experienced dramatic breakthroughs, capturing the imagination of both the scientific community and the general public. With less public notice, receptor biology has brought a cascade of new discoveries and insights. The entire science of pharmacology has been virtually rewritten in terms of receptor phenomenology. In particular, the discovery of specific receptors for steroid and protein hormones has been of seminal importance. With this new information, we have advanced our understanding of the mechanism and specifity of hormone action. We can now explain how hormones interact selectively with specific target cells and how hormones alter biochemical events within the target cells. These facts have already impacted on applied problems of clinical medicine, particularly in diagnosis and treatment of cancer and some metabolic diseases. Now, a new and important application of basic receptor biology and chemistry looms ahead. Within a few short years since the discovery of the progesterone receptor, chemists have synthesized molecules with a greater affinity for the receptor than progesterone itself and which, while occupying the receptor, fail to trigger the events which transform a target cell from the unstimulated to the stimulated state. This is the basis of the competitive inhibitory action of the anti-progestational agent, synthesized by the chemists at Roussel Uc1af, Paris, and designated RU 486.

To focus on this and related problems an International Congress to examine many aspects of male and female Reproductive Health Center Care was held on 10-15 October 1982 in Maui, Hawaii, USA. This volume is a result of the proceedings from the 'Symposium on Male Fertility and its Regulation' which was a part of the Reproductive Health Care Congress. The organizers of this symposium recognized the need to focus male reproductive understanding on contraceptive development. The ultimate objective was and still is to produce a variety of safe and effective male contraceptives similar to that accomplished in the female. Speakers were invited to review the state of the art in several areas related to male contra ception, reproductive toxicity and reproductive biology. The abstracts of these sessions were published as a special issue of Archives of Andrology (Vol. 9, No.1, August, 1982). Subsequently, this volume was assembled from key papers presented at the Symposium. Additionally, invited man uscripts from leaders in specific areas were solicited to provide additional range to the topics covered.
