Customized storage protocols are often necessary to address differing strategies and program goals for specific pine species. Subtropical pine pollen has been and will be important for forestry programs around the world, and the evaluation of pollen storage protocols for these species permits the determination of successful strategies to maintain viability over time. Germination of pollen grains using in vitro agar gel methodology has been shown to successfully predict in vivo germination of pollen tubes. Five methods commonly used for temperate pine pollen storage were evaluated for their efficacy for maintaining temperate (P. taeda, P. radiata) and subtropical species (Pinus tecunumanii, P. oocarpa, P. maximinoi) pollen after 6 months. For best results, cryopreservation at -196 & deg;C should be used for subtropical pine pollen storage if available. When reliable resources for cryopreservation are limited, direct freezing at -20 & deg;C is recommended. Like the P. taeda and P. radiata pollen evaluated in this study, subtropical pine pollen exhibited increased in vitro germination rates when exposed to the lower temperatures evaluated after 6 months in storage. Temperate species pollen storage protocols evaluated in this study were used successfully for subtropical and tropical pine species. The 3 subtropical pine species' pollen assessed diverged from temperate pollen behavior when only prestored pollen is considered. Cryogenic and direct freezing storage methods yielded higher in vitro germination rates for subtropical species than temperate species (P. taeda and P. radiata). Vacuum drying/freezing and refrigeration, however, yielded higher in vitro germination rates for temperate species. Additional research into the physiology of these differences and the correlations with evolutionary phylogeny information is recommended to determine the caus.

Book excerpt: Cryopreservation of pollen offers a simple and effective method of long-term pollen storage. Pollen may be stored in liquid nitrogen (LN) for many years without loss of its essential capabilities to pollinate, fertilize, and set normal fruit and seed when used in plant breeding, for controlled pollinations, or for the conservation of plant genetic resources. Pollen of Corylus, Fragaria, Pyrus, Rubus and Vaccinium was frozen in LN either freshly collected, air dried, or after equilibration with atmospheres of 0, 20, 40, 60, 80, 90, or 100% relative humidity. Survival was assayed by in vitro germination on a sucrose-agar medium and by controlled pollinations (in Corylus only). All pollen survived freezing in LN provided the pollen moisture content was adjusted properly. Pollen of the wind-pollinated genus Corylus was not tolerant of desiccation and survived freezing at very high moisture content levels relative to the insect-pollinated Rosaceae pollens which survived freezing only when dried prior to their immersion in LN. Guidelines are presented for the establishment of a pollen bank at the National Clonal Germplasm Repository (NCGR) in Corvallis, using cryopreservation for the long-term conservation of plant genetic resources.

Horticultural crops, as high-value crops, have an important role to play in revitalizing rural economies and can add significantly to national economies. Moreover, horticulture provides more than twice the number of jobs compared to traditional cereal crop production, and the shifting of conventional agriculture towards high-value horticulture has increased employment opportunities in developing countries. To exploit this potential, researchers need a vast array of horticultural genetic resources and information on new traits. Horticultural crops, which are only a part of PGRFA (Plant Genetic Resources for Food and Agriculture), are characterized by a wide and varied range of species. In fact, there are five major horticultural crop groups: fruit and nut crops, vegetables, food legumes, roots and tubers, and lastly the ornamental and medicinal group. In this context, the present book provides a comprehensive overview of the current state of conservation and utilization of horticultural genetic resources, addressing contemporary approaches to conservation in connection with different technologies, including biotechnological approaches as practised in India and in some cases, globally. It includes a brief chapter on the unique nature of horticultural genetic resources, providing a rationale for viewing them as being distinct from field crop genetic resources. Subsequent chapters share insights on protocols for the conservation of selected horticultural crops ex situ, and focus on the increased need to complement these efforts with in situ conservation approaches. Book excerpt: Ex situ preservation of germplasm for higher plant species has been accom plished using either seeds or clones, but storage of these under typical condi tions does not provide the extreme longevities that are needed to minimize risk of loss. Costs of maintenance and regeneration of stocks are also high. Systems that provide virtually indefinite storage should supplement existing methods and it is within this context that cryopreservation is presented. The use of low temperature preservation was initially more a concern of medicine and animal breeding, and was expanded to plants in the 1970s. Sur vival after cryogenic exposure has now been demonstrated for diverse plant groups including algae, bryophytes, fungi and higher plants. If survival is com monplace, then the eventual application is a cryopreservation system, whereby cells, tissues and organs are held indefinitely for use, often in the unforeseen future. The increasing interest and capabilities for application could not have occurred at a more opportune time since expanding human populations have placed unprecedented pressures on plant diversity. This book emphasizes cry opreservation of higher plants and was initially driven by the concern for loss of diversity in crops and the recognized need that this diversity would be essential for continued improvement of the many plants used by society for food, health and shelter. The interest in cryopreservation has been expanded by conservationists and their concerns for retaining, as much as possible, the diversity of natural populations. The need for cryopreservation, thus, is well established.

Book excerpt: The germ plasm of numerous plant species, especially those of forest trees, some agricultural crops, and medicinal plants, is endangered and threatened with extinction. This depletion of germplasm pools and the shrinkage of naturally occurring genetic resources have caused international concern. Conventionally, the germplasm of plants is conserved through seeds, tubers, roots, corms, rhizomes, bulbs, cuttings, etc. However, the germ plasm of a number of trees and plantation crops (such as coconut, cocao, coffee, oil palm, rubber, mango, horse chestnut, etc. ) cannot be preserved since their seed are short-lived (recalcitrant). Likewise, germplasm of vegetatively propagated crops (such as potato and cassava) cannot be stored on a long term basis and has to be grown and multiplied periodically in nurseries and fields. The plants are thus exposed to unpredictable weather conditions and diseases, with the result that instances are known where entire genetic stocks are lost. Therefore, unconventional methods are being developed for the storage and international exchange of germplasm. For this purpose in vitro cultures have been employed, but they can only enable short-to medium term preservation; moreover, cell cultures upon repeated subculture undergo genetic erosion. In view of the recent developments in the in vitro induction of genetic variability through somaclonal variation, somatic hybridization, recombinant DNA technology, etc. , new methods need to be employed for the storage of desirable cultures. In this regard freeze preservation of cells in liquid nitrogen (-196 0q, like that of semen, enables long-term storage, theoretically, for an indefinite period of time.
