Book Starvation survi v al Studies on a Marine Vibrio Sp and Other Selected Marine Bacteria

Download or read book Starvation survi v al Studies on a Marine Vibrio Sp and Other Selected Marine Bacteria written by Penny Susan Amy and published by . This book was released on 1982 with total page 296 pages. Available in PDF, EPUB and Kindle. Book excerpt: The starvation-survival patterns of 16 freshly isolated open ocean bacteria and in a psychrophilic Vibrio sp., Ant-300, were determined. These isolates displayed three survival patterns as determined by viable cell count. These were (a) a rapid increase followed by a decrease to a constant value (pattern of Ant-300 and 7 isolates), (b) a rapid decrease to a constant value (4 isolates), (c) a gradual increase to a constant value (5 isolates). Isolates changed shape with most becoming coccobacilli by 8-9 mo starvation. All isolates decreased in size, thereby increasing their surface to volume ratio. One isolate from each starvation survival pattern and Ant-300 were incubated under simulated open ocean conditions with [14C]glutamic acid and monitored for respiration and cellular in corporation of radiolabel. In each case a rapid linear respiration and cellular incorporation of labeled glutamic acid was seen. Physiological studies on Ant-300 were conducted both during 6 weeks starvation and recovery (the addition of nutrients) from starvation. During long-term starvation, macromolecules on a per cell basis rapidly decreased as cells fragmented (division without biomass increase) in the first 24-48 h. At one week starvation, the quantity of protein and DNA became constant whereas the level of RNA increased. Also after one week, a small peak in activity was seen in endogenous respiration, respiration potential (substrate added), and ATP level per viable cell; and glutathione was resynthesized after an initial decline when appropriate amino acids were present. After recovery from starvation Ant-300 displayed characteristics of both exponential and growing cells. Individual cell size and macromolecules increased exponentially with the addition of nutrients after lag periods which related to the prior length of starvation. Cells of Ant-300 displayed greater degrees of dormancy with increased starvation time. Protein fingerprint patterns formed by two dimensional gel electrophoresis showed changes in individual protein spots during long-term starvation. Cells grown under pressures comparable to those from which Ant-300 was isolated, showed similar protein patterns but lower concentrations when pressures higher than 100 atmospheres were employed. Cells recovering from starvation also showed much the same pattern as those under other conditions of growth, indicating that [35S]methionine is synthesized into all the proteins of a growing cell.

Download or read book Starvation in Bacteria written by S. Kjelleberg and published by Springer Science & Business Media. This book was released on 2013-11-11 with total page 288 pages. Available in PDF, EPUB and Kindle. Book excerpt: Concerted efforts to study starvation and survival of nondifferentiating vegeta tive heterotrophic bacteria have been made with various degrees of intensity, in different bacteria and contexts, over more than the last 30 years. As with bacterial growth in natural ecosystem conditions, these research efforts have been intermittent, with rather long periods of limited or no production in between. While several important and well-received reviews and proceedings on the topic of this monograph have been published during the last three to four decades, the last few years have seen a marked increase in reviews on starvation survival in non-spore-forming bacteria. This increase reflects a realization that the biology of bacteria in natural conditions is generally not that of logarithmic growth and that we have very limited information on the physiology of the energy-and nutrient-limited phases of the life cyde of the bacterial cello The growing interest in nongrowing bacteria also sterns from the more recent advances on the molecular basis of the starvation-induced nongrowing bacterial cello The identification of starvation-specific gene and protein re sponders in Escherichia coli as weIl as other bacterial species has provided molecular handles for our attempts to decipher the "differentiation-like" responses and programs that nondifferentiating bacteria exhibit on nutrient limited growth arrest. Severallaboratories have contributed greatly to the progress made in life after-log research.

Book Molecular Approaches to the Study of the Ocean

Download or read book Molecular Approaches to the Study of the Ocean written by K.E. Cooksey and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 562 pages. Available in PDF, EPUB and Kindle. Book excerpt: Marine biological science is now studied at the molecular level and although research scientists depend on information gained using molecular techniques, there is no book explaining the philosophy of this approach. Molecular Approaches to the Study of the Ocean introduces the reasons why molecular technology is such a powerful tool in the study of the oceans, describing the types of techniques that can be used, why they are useful and gives examples of their application. Molecular biological techniques allow phylogenetic relationships to be explored in a manner that no macroscopic method can; although the book deals with organisms near the base of the marine food web, the ideas can be used in studies of macroorganisms as well as those in freshwater environments.

This book was released on 2012-12-06 with total page 355 pages. Available in PDF, EPUB and Kindle. Book excerpt: This text on viable but non-culturable organisms provides information on topics including: morphological changes; the role of membranes; genetics and genetic regulation; molecular methods for detection; as well as survival dominancy and related phenomena. The main purpose of the text is to elucidate the phenomenon and to distinguish it from other seemingly related but different phenomena such as spore formation, dormancy, starvation, and injury. It covers a cross section of morphology, metabolism, genetics, ecology and epidemiology.
