The skin serves as a protective interface between the body and the noxious environment. Ultraviolet (UV) radiation is one of the most important sources of environmental stress, inducing photochemical changes in the skin that may lead to sunburn, premature skin aging and skin cancer. The skin and UV also play a leading part in the vitamin D endocrine system. When UVB photons strike the epidermis, their energy can be used for the synthesis of vitamin D3. Furthermore, the skin is one of the direct target issues for vitamin D3 action. The close interplay between UV and vitamin D3 production in the skin evokes the possibility of mutual interactions. The active form of vitamin D, calcitriol (1,25(OH)2D3) thought the binding to vitamin D nuclear receptor (VDR), modulates expression of hundreds of genes. In an additions activity of VDR could be regulated on various levels. Here we presenting our preliminary results concerning effects of VDR expression, alternative splicing, expression of coactivator RXR on the activity of 1,25(OH)2D3 in human keratinocytes (HaCaT, A431 and primary cells). At least three splicing variants of VDR receptor mRNA were expressed in human keratinocyte cells, and its expression was modulated by 1,25(OH)2D3. Interestingly, keratinocytes cell lines used in the study differ in the patter of expressed VDR isoforms, which could at least partially explain observed various response to 1,25(OH)D3. Next it was shown, that overexpression of VDR splicing variants (VDR1 or VDR3) in keratinocytes had a modulatory effect on VDR signalling including delayed translocation of VDR to the nucleus and partial selectivity in target genes. The translocation of VDR1 and VDR3 was observed live by using fusion constructs of VDR isoform with green fluorescent protein (GFP). On the other hand, attenuation of VDR expression by RNAi resulted in downregulation of classic VDR targets in keratinocytes, however some phenotypic changes including induction of differentiation by 1,25(OH)2D3 were not full diminished what suggested involvement of alternative pathways. Thus, we are planning to investigate non geneomic pathways activated by vitamin D.Summarising, our preliminary results suggest that alternative splicing of VDR may modulate response of keratinocytes to 1,25(OH)2D3 and could be essential regulatory factor in maintenance of epidermal barrier and skin physiology.

Book excerpt: "1,25-dihydroxyvitaminD$ sb3$ (1,25(OH)$ sb2$D$ sb3$) is anti-proliferative and pro-differentiative in a variety of cell types, including human keratinocytes. Exposure of cultured normal human keratinocytes to 1,25(OH)$ sb2$D$ sb3$ markedly reduces ($ sp3$H) thymidine incorporation and cell number and arrests these cells in the G1/G0 phase of the cell cycle. Calcium and 1,25(OH)$ sb2$D$ sb3$ act in concert to modulate the expression of two important cell-cycle associated genes, c-fos and p53, and of markers of keratinocyte differentiation. In contrast, inhibition of cell growth and of the cell cycle associated oncogene, c-myc, in the malignant keratinocyte cell line, HPK1Aras, requires 10 to 100 fold higher concentrations of 1,25(OH)$ sb2$D$ sb3$ than do the immortal non-malignant human keratinocyte cell line, HPK1A. Cell cycle analysis also reveals that 10-100 fold higher concentrations of 1,25(OH)$ sb2$D$ sb3$ are required to induce cell cycle arrest in HPK1Aras cells as compared to HPK1A cells. Analysis of the vitamin D receptor (VDR) from these two cell lines reveals identical sizes, numbers, and ligand binding characteristics. Furthermore, the sequence of the DNA binding domain of this receptor is unchanged in the vitamin D resistant HPK1Aras cells. Gel mobility shift assays using extracts from both cell lines reveals that the complexes formed by HPK1Aras nuclear extracts in the presence of a vitamin D DNA response element (VDRE) probe contain VDR but not its dimerization partner, the retinoic acid X receptor (RXR). In contrast, HPK1A nuclear extracts form complexes that contain both VDR and RXR. Overexpressing wild type RXR$ alpha$ in HPK1Aras cells results in VDRE-binding complexes containing VDR/RXR heterodimers. However, the sequence of the RXR$ alpha$ is unchanged in HPK1Aras cells and its expression in both cell lines is the same. Western blot analysis of RXR$ alpha$ from ras transformed keratinocytes suggests that its post-translational modification is" --
