Two different approaches are discussed and contrasted with each other. The first approach is based on the classical equations of flow and transport, called 'continuum models'. The second approach is based on modern methods of statistical physics of disordered media; that is, on 'discrete models', which have become increasingly popular over the past 15 years. The book is unique in its scope, since (1) there is currently no book that compares the two approaches, and covers all important aspects of porous media problems; and (2) includes discussion of fractured rocks, which so far has been treated as a separate subject. Portions of the book would be suitable for an advanced undergraduate course. Book excerpt: The first edition of this book was based on the lectures which I gave at Cornell University during 1958 as George Fisher Baker Lecturer, and I would like to repeat my warmest thanks to Professor F. A. Long and the other members of the Department of Chemistry for their kindness and helpful advice. The present edition was largely written during the tenure of a Visiting Professorship awarded by the Royal Society and the Israeli Academy of Sciences. I am deeply indebted to both of these bodies and also to the hospitality of the Weizmann Institute of Science, in particular to Professor David Samuel and Professor F. S. Klein of the Isotopes Research Department. The subject as a whole has expanded greatly since 1959, especially in two fields, namely, the direct study of fast proton-transfer reactions (notably by the relaxation methods pioneered by Eigen), and the experi mental and theoretical study of hydrogen isotope effects. In order to keep the size of the book within reasonable bounds it has been necessary to adopt a selective policy, and this is particularly the case in Chapter 9 where I have chosen to treat a few types of reaction in some detail rather than to attempt a more complete coverage.

Book excerpt: Photochemical switches can be used to control the secondary structure of peptides and proteins. For this purpose, we created and studied a series of azobenzene cross-linkers analogous to the Kumita compound (Kumita et al., 2000), a symmetrical reagent 4,4' -bis(iodoacetamide)azobenzene, but with a range of thermal stabilities. To increase the rate of thermal reversion, derivatives with enhanced delocalization of the lone pair electrons of the para amino were designed. High reactivity and selectivity toward Cys residues was achieved by introducing methanethiosulfonyl groups. These groups enable reversible attachment of the cross-linker to peptides and proteins via disulfide linkages. The effects of the cross-linkers on the conformation of a test peptide (FK*-11) were evaluated. Second, a series of "long" azobenzene cross-linkers, containing two Cys-reactive chloroacetamide- and iodoacetamide-diphenylethynyl moieties that are linked to the azobenzene chromophore was designed and synthesized.
