Book excerpt: In this study the use of HPMA copolymer to deliver Doxorubicin (DOX) has resulted in a favorable modification of the pharmacokinetic behaviour of DOX. This study has attempted to demonstrate whether the efficacy of PK1 is enhanced as a result of enhanced permeability and retention (EPR) or whether it is due to altered pharrnacokinetics and drug exposure parameters using two different models of murine tumour. The two models were MAC15A, which is poorly differentiated and poorly vascularised and MAC26 which is well differentiated and well vascularised. This study has shown that the method used for extraction of DOX and PK1 from plasma and tumour was valid with acceptable sensitivity. Evaluation of this method showed both acceptable extraction efficiencies and good reproducibility for extraction of PK1 and DOX from plasma. Preliminary pharmacokinetic work on both PK1 and DOX showed different pharmacokinetic profiles with both compounds. Pharmacokinetic parameters obtained from the data showed a higher AUC for PK1 in plasma of NMRI mice compared to that for DOX with slightly longer half life in the case of PKI . On other hand, there was a small amount of free DOX released from PKI in plasma in the case of MAC15A tumour model but there was no free DOX from PKI seen in the case of MAC26 tumour model. Analysis of plasma samples showed that PKI was still detectable at the 72h time point. The most interesting points in this study were the tumour drug levels and the associated pharmacokinetic parameters. Analysis of samples obtained from both MAC15A and MAC26 tumours showed very high levels of PK1 and free DOX released from PK1 within the tumours compared with DOX 10 mg/kg (iv). In general, AUC of PK1 and released DOX following a single 490 mg/kg which equates to 40mg/kg of DOX was very high when compared with free DOX and....
